Three different lines of evidence were obtained to show that trypsin modifies the actin-myosin interaction: (I) At trypsin to actomyosin or myosin ratios between 1 to 300 and 1 to 500, 30 min of trypsin treatment causes an 8-fold increase in the Ca2+-modified ITPase activity of actomyosin but has no effect on the Ca2+-modified ITPase activity of myosin alone. At these same trypsin to actomyosin ratios, the Mg2+ + Ca2+-modified ATPase activity increases by 10–30% during the first 1–2 min of trypsin digestion, and then decreases rapidly to less than 20% of its original activity after 60 min of digestion. Trypsin has no effect on the Mg2+ + Ca2+-modified ATPase activity of pure myosin. (2) The rate of turbidity response of reconstituted actomyosin suspensions is first increased and then decreased by trypsin treatment. At trypsin to actomyosin ratios of 1 to 3000, rate of turbidity response is maximal after 5 min of trypsin digestion and then decreases; after 60 min, the turbidity response is much slower than the response of the control actomyosin. (3) Supercontracted sarcomeres, shortened to less than 50% of their initial length, are lengthened to 70% of their initial length by 4 min of trypsin treatment. Myosin B from such lengthened sarcomeres has less than 35% of its myosin converted to light meromyosin and heavy meromyosin.

These results show that trypsin modifies the actin-myosin interaction in at least two ways: (1) a very rapid initial modification that increases the Mg2+ + Ca2+-modified ATPase activity and the rate of turbidity increase, and (2) a slower modification that decreases the Mg2+ + Ca2+-modified ATPase activity and rate of turbidity response, and that lengthens contracted sarcomeres. Tryptic modification is not due to cleavage of myosin to light and heavy meromyosin. Since tryptic modification occurs more rapidly than conversion of myosin to light and heavy meromyosin, all heavy meromyosin preparations will be modified.  相似文献

Summary Hard- and softwater acclimated adult rainbow trout were statically exposed to copper (12.5, 25, 50, 100, and 200 ppb) for two, 12 h periods at neutral and pH 5.0. Unidirectional Na+, and Cl, and net Na+, Cl, K+, and ammonia fluxes were monitored as a measure of branchial ionoregulatory disturbance. Copper concentrations as low as 12.5 ppb led to measurable ion losses. Net Na+, Cl, and K+ losses were concentration dependent and unaffected by prior acclimation to either hard- or softwater at both neutral pH and pH 5. From 12.5 to 50 ppb net NaCl losses arose primarily as a result of the inhibition ofJ in, and at higher concentrations,J out was also stimulated. In softwater,J in was more resistant to inhibition than in hardwater. However, in hardwater,J out recovered to normal levels during the second 12 h period, but no such recovery was found in softwater. Plasma NaCl was inversely correlated with [copper], while plasma glucose and ammonia increased with [copper]. At pH 5.0 and [copper] from 12.5 to 50 ppb, H+ contributed significantly to the total ion loss, while at 100 and 200 ppb, ion losses were no greater at pH 5.0 than at neutral pH. In no case were the effects of copper and H+ strictly additive.  相似文献

The genetic and biochemical characteristics of a particular class of mutants at the rudimentary locus are described. The mutants are pyrimidine auxotrophs, like classical rudimentary alleles, but they are unique in that they do not alter the size or shape of the wing (Falk and Nash 1974b). Aspartate transcarbamylase and dihydroorotase activities have been measured in seven different normal-winged mutants, and the results indicate that these strains are enzymologically "leaky" mutants. Previous studies have shown that three genetic functions (corresponding to the first three enzymes of pyrimidine synthesis) are associated with the rudimentary locus. Four of the seven mutants appear to affect all three of these functions. Each of the four is temperature sensitive, and a biochemical analysis of the temperature sensitivity of one of these mutants, (r)pyr1-3, suggests that a process affecting the synthesis or assembly of these enzymes is altered at high temperatures.  相似文献

Summary The rudimentary locus of Drosophila melanogaster is shown to be at least bifunctional. Mutants in different regions of the locus have either no CPSase or no ATCase activity; some mutants lack both activities. There is international recognition of the need for sustainable research ethics committees to provide ethical review of human subjects research in developing countries, but many developing countries do not have such committees (often called 'IRBs'). Theoretical and practical uncertainties encountered by an IRB on the Caribbean island of Grenada offer insight into ethical review of research in developing countries. Theoretical uncertainties include questions about whether means of ensuring confidentiality and obtaining informed consent will be effective in local settings, and whether deviations from Western norms are justifiable. International guidelines are helpful in addressing these concerns, but are subject to interpretation. Guidelines are less helpful in practical areas like selecting members or chairs. They do not address what sort of procedures and paperwork will work in a developing country, or IRBs' relationships to governments that have no mandate for them. Experiences presented here show that IRBs in developing countries can sustainably adhere to international standards. Sustainability requires knowledge, personal commitment, and an official mandate to uphold international standards. Capacity building must therefore focus on educational programs to make developing country leaders knowledgeable about the value of international guidelines to their nations. Such knowledge is needed before people will become motivated to promote, implement, and uphold the guidelines. People in developing countries must help design bridges to help their nations put international standards into practice. The structure of such bridges may, of necessity, vary in different settings.  相似文献

The objective of this study was to determine whether acute volume loading elevates T-wave alternans (TWA) in dogs with structurally normal hearts. TWA predicts sudden cardiac arrest in patients with left ventricular dysfunction and congestive heart failure. However, volume load and ventricular stretch may themselves precipitate arrhythmias. It is unclear to what extent volume load causes TWA. In six male mongrel dogs [25.8 kg (SD 4.2)] under general anesthesia, we measured TWA during progressive atrial pacing to 160 beats/min. Pacing was performed at baseline, at the midpoint and peak of a saline infusion designed to induce acute CHF, and then during diuresis. Dog 1 was hypothermic throughout the protocol and excluded from analysis. For dogs 2-6, 102 ml/kg (SD 30) were infused over 315 min (SD 50), causing pulmonary capillary wedge pressure to rise from 9.6 (SD 3.5) to 21.2 mmHg (SD 1.6) (P < 0.01), and heart rate variability to fall (P < 0.01). TWA magnitude (Valt) rose in all dogs with volume load (P < 0.001). Compared with baseline, TWA at peak infusion had higher magnitude [Valt 3.4 (SD 1.95) vs. 0.5 muV (SD 0.35); P = 0.011] and occurred at lower heart rates [128 (SD 6) vs. 151 beats/min (SD 12); P = 0.008]. Net volume load was linearly related to Valt (P < 0.01), with each 10 ml/kg net volume load increasing Valt by 0.23 muV. Acute volume overload elevates TWA in normal canine hearts. Although dramatic, however, this effect may contribute clinically to abnormal TWA only in patients with marked volume overload. Future studies should examine the interaction of fluid overload, myocardial disease, and arrhythmia susceptibility.  相似文献

The herpes simplex virus type 1 capsid is a protective shell that acts as a container for the genetic material of the virus. After assembly of the capsid, the viral DNA is translocated into the capsid interior through a channel formed by the portal. The portal is composed of a dodecamer of UL6 molecules which form a ring-like structure found at a single vertex within the icosahedron. Formation of portal-containing capsids minimally requires the four structural proteins (VP5, VP19C, VP23, and UL6) and a scaffolding protein (UL26.5). Recently, an interaction between UL26.5 and the portal has been identified, suggesting the scaffold functions by delivering the portal to the growing capsid shell. The aim of this study was to identify regions within UL26.5 required for its interaction with the portal. A specific region was identified by mutational analysis. Deletion of scaffold amino acids (aa) 143 to 151 was found to be sufficient to inhibit formation of the scaffold-portal complex as assayed in vitro. The aa 143 to 151 contain the sequence YYPGE, which is highly conserved among alpha herpesviruses. Although it did not bind to the portal, the Delta143-151 mutant was found to retain the ability to support assembly of morphologically normal capsids in vitro. Such capsids, however, did not contain the portal. The results suggest assembly of portal-containing capsids requires formation of a scaffold-portal complex in which intermolecular contact is dependent on scaffold aa 143 to 151.  相似文献

Conversion of grasslands to woodlands may alter the sensitivity of CO2 exchange of individual plants and entire ecosystems to air temperature and precipitation. We combined leaf‐level gas exchange and ecosystem‐level eddy covariance measurements to quantify the effects of plant temperature sensitivity and ecosystem temperature responses within a grassland and mesquite woodland across seasonal precipitation periods. In so doing, we were able to estimate the role of moisture availability on ecosystem temperature sensitivity under large‐scale vegetative shifts. Optimum temperatures (Topt) for net photosynthetic assimilation (A) and net ecosystem productivity (NEP) were estimated from a function fitted to A and NEP plotted against air temperature. The convexities of these temperature responses were quantified by the range of temperatures over which a leaf or an ecosystem assimilated 50% of maximum NEP (Ω50). Under dry pre‐ and postmonsoon conditions, leaf‐level Ω50 in C3 shrubs were two‐to‐three times that of C4 grasses, but under moist monsoon conditions, leaf‐level Ω50 was similar between growth forms. At the ecosystems‐scale, grassland NEP was more sensitive to precipitation, as evidenced by a 104% increase in maximum NEP at monsoon onset, compared to a 57% increase in the woodland. Also, woodland NEP was greater across all temperatures experienced by both ecosystems in all seasons. By maintaining physiological function across a wider temperature range during water‐limited periods, woody plants assimilated larger amounts of carbon. This higher carbon‐assimilation capacity may have significant implications for ecosystem responses to projected climate change scenarios of higher temperatures and more variable precipitation, particularly as semiarid regions experience conversions from C4 grasses to C3 shrubs. As regional carbon models, CLM 4.0, are now able to incorporate functional type and photosynthetic pathway differences, this work highlights the need for a better integration of the interactive effects of growth form/functional type and photosynthetic pathway on water resource acquisition and temperature sensitivity.  相似文献
