Cutinsomes, spherical nanoparticles containing cutin mono‐ and oligomers, are engaged in cuticle formation. Earlier they were revealed to participate in cuticle biosynthesis in Solanum lycopersicum fruit and Ornithogalum umbellatum ovary epidermis. Here, transmission electron microscopy (TEM) and immunogold labeling with antibody against the cutinsomes were applied to aerial cotyledon epidermal cells of Arabidopsis thaliana mature embryos. TEM as well as gold particles conjugated with the cutinsome antibody revealed these structures in the cytoplasm, near the plasmalemma, in the cell wall and incorporated into the cuticle. Thus, the cutinsomes most probably are involved in the formation of A. thaliana embryo cuticle and this model plant is another species in which these specific structures participate in the building of cuticle in spite of the lack of the lipotubuloid metabolon. In addition, a mechanism of plant cuticle lipid biosynthesis based on current knowledge is proposed.  相似文献

In recent years, transposon insertion polymorphisms have been utilized as molecular markers, and a range of techniques tailored towards identification of insertion sites of various transposable elements have been developed. In the present paper we describe the application of a recently developed DcMaster transposon display system to analyse the genetic diversity of Polish breeding materials of carrot (Daucus carota) and to identify polymorphisms useful for hybrid seed purity testing. Using 3 sets of breeding materials (each consisting of the cytoplasmic male sterility stock, the maintainer, the pollinator, and the corresponding F1 hybrid), we identified 56 DcMTD markers. DcMaster insertion sites proved to be highly polymorphic in cultivated carrot, as 79% of all insertion sites differentiated between individual plants. Fourteen stock-specific DcMTD markers were further selected as potentially useful for hybrid seed purity testing.  相似文献

INTRODUCTION: The aim of this study was to evaluate the blood concentration of hepatocyte growth factor (HGF) in patients at various stages of retinopathy. We hypothesised that the high level of HGF found in diabetic patients may be an important marker of retinopathy progression and that HGF level may be an index of the risk of proliferative retinopathy. MATERIAL AND METHODS: The participants in the study were 76 patients with type 1 diabetes mellitus. Of these, 35 patients were without retinopathy and formed Group 1. Of the remaining 41 patients with retinopathy, 20 patients had non-proliferative diabetic retinopathy (NPDR) and formed Group 2, while 21 patients had proliferative diabetic retinopathy (PDR) and formed Group 3. We evaluated the concentration of HGF In the peripheral blood by an enzyme-linked immunosorbent assay. RESULTS: Mean serum concentrations of HGF in the control group were significantly lower than in the type 1 diabetic patients. We found a significant increase in HGF serum concentrations in diabetic patients with PDR compared with the control group. Mean serum HGF concentrations were significantly higher in diabetic subjects with PDR than in diabetic patients without retinopathy. CONCLUSION: HGF concentration is increased in patients with type 1 diabetes mellitus with proliferative retinopathy, and concentrations increase with the progression of retinopathy, suggesting that HGF plays a role in the pathogenesis of proliferative diabetic retinopathy.  相似文献

A series of tetramethylammonium tetrahalogenoferrates(III), [FeBr4−nCln] (n = 0, 1, 3, 4), of general formula [(CH3)4N][FeBr4−nCln], have been synthesized. The crystal and molecular structures of [(CH3)4N][FeCl4] were determined. The compound is isostructural with its [FeBr4−nCln] (n = 0, 1, 3, 4) analogues. Magnetic measurements of the powdered samples of [(CH3)4N][FeBr4−nCln] gave negative values of the Weiss constant, which suggest antiferromagnetic coupling. The strength of the antiferromagnetic interactions strongly depends on the kind of halide ligands in the coordination sphere of iron(III) and increases with an increasing number of the bromide anions.  相似文献

The flaviviral nonstructural 3 protease (NS3pro) is essential for virus replication and is therefore a pharmaceutically relevant target to fight Dengue and West Nile virus (WNV). NS3pro is a chymotrypsin‐like serine protease which requires a polypeptide cofactor (NS2B) for activation. Recent X‐ray crystallography studies have led to the suggestion that the substrate binds to the two‐component NS2B‐NS3pro enzyme by an induced‐fit mechanism. Here, multiple explicit water molecular dynamics simulations of the WNV NS2B‐NS3pro enzyme show that the active conformation of the NS2B cofactor (in which its β‐loop is part of the substrate binding site) is stable over a 50‐ns time scale even in the absence of the inhibitor. The partial and reversible opening of the NSB2 β‐loop and its correlated motion with an adjacent NS3pro loop, both observed in the simulations started from the active conformation, are likely to facilitate substrate binding and product release. Moreover, in five of eight simulations without inhibitor (started from two X‐ray structures both with improperly formed oxyanion hole) the Thr132‐Gly133 peptide bond flips spontaneously thereby promoting the formation of the catalytically competent oxyanion hole, which then stays stable until the end of the runs. The simulation results provide evidence at atomic level of detail that the substrate binds to the NS2B‐NS3pro enzyme by conformational selection, rather than induced‐fit mechanism.  相似文献

Microtubules in lipotubuloids of the Ornithogalum umbellatum stipule epidermis cells change their diameters depending on the motion of the cytoplasmic domains rich in microtubules and lipid bodies. Microtubules fixed during rotary and progressive motion of the lipotubuloids composed of the same number of protofilaments fall into two populations – wide (43–58 nm) and narrow (24–39 nm) in size. Following blockage of the motion with 2,4-dinitrophenol (DNP), the range of this diversity is smaller, microtubules become a medium-sized population (34–48 nm). When DNP is removed and the motion reactivated, 2 populations of microtubules reappear. Analysis of the structure of the microtubule wall revealed that changes in the microtubule diameters resulted from varying distances between the adjacent protofilaments, and stretching and compression of tubulin subunits in the protofilaments.A supposition has been put forward that the changes in the sizes of O. umbellatum microtubule diameters: 1) are connected with the interactions between microtubules and actin microfilaments lying along these microtubules; 2) can be the driving force of the rotary motion of lipotubuloids.  相似文献

Carrot is the most economically important member of the Apiaceae family and a major source of provitamin A carotenoids in the human diet. However, carrot molecular resources are relatively underdeveloped, hampering a number of genetic studies. Here, we report on the synthesis and characterization of a bacterial artificial chromosome (BAC) library of carrot. The library is 17.3-fold redundant and consists of 92,160 clones with an average insert size of 121 kb. To provide an overview of the composition and organization of the carrot nuclear genome we generated and analyzed 2,696 BAC-end sequences (BES) from nearly 2,000 BACs, totaling 1.74 Mb of BES. This analysis revealed that 14% of the BES consists of known repetitive elements, with transposable elements representing more than 80% of this fraction. Eleven novel carrot repetitive elements were identified, covering 8.5% of the BES. Analysis of microsatellites showed a comparably low frequency for these elements in the carrot BES. Comparisons of the translated BES with protein databases indicated that approximately 10% of the carrot genome represents coding sequences. Moreover, among eight dicot species used for comparison purposes, carrot BES had highest homology to protein-coding sequences from tomato. This deep-coverage library will aid carrot breeding and genetics. Background  

We present here the recent update of AMS algorithm for identification of post-translational modification (PTM) sites in proteins based only on sequence information, using artificial neural network (ANN) method. The query protein sequence is dissected into overlapping short sequence segments. Ten different physicochemical features describe each amino acid; therefore nine residues long segment is represented as a point in a 90 dimensional space. The database of sequence segments with confirmed by experiments post-translational modification sites are used for training a set of ANNs.  相似文献
