Seed oils are important natural resources used in the processing and preparation of food. Histone modifications represent key epigenetic mechanisms that regulate gene expression, plant growth and development. However, histone modification events during fatty acid (FA) biosynthesis are not well understood. Here, we demonstrate that a mutation of the histone acetyltransferase GCN5 can decrease the ratio of α‐linolenic acid (ALA) to linoleic acid (LA) in seed oil. Using RNA‐Seq and ChIP assays, we identified FAD3, LACS2, LPP3 and PLAIIIβ as the targets of GCN5. Notably, the GCN5‐dependent H3K9/14 acetylation of FAD3 determined the expression levels of FAD3 in Arabidopsis thaliana seeds, and the ratio of ALA/LA in the gcn5 mutant was rescued to the wild‐type levels through the overexpression of FAD3. The results of this study indicated that GCN5 modulated FA biosynthesis by affecting the acetylation levels of FAD3. We provide evidence that histone acetylation is involved in FA biosynthesis in Arabidopsis seeds and might contribute to the optimization of the nutritional structure of edible oils through epigenetic engineering.  相似文献

Two cyanobacterial genes ccaA and FBP/SBPase related to CO2 hydration and Calvin cycle were induced into rice plants. Three homologous transgenic strains were generated with ccaA and FBP/SBPase alone or in combination independently and grown under field conditions. The biochemical, physiological, and leaf anatomic results indicated that stomatal and mesophyll conductance to CO2, net photosynthetic rate, carboxylation efficiency, and other physiological and biochemical parameters increased significantly in the overexpression strains with FBP/SBPase and CcaA + FBP/SBPase but not in the CcaA strain. Leaf anatomy structure showed no significant modifications between the transgenic and wild-type strains. The CcaA protein was shown to be located in the cytoplasm. These results showed that the effect on improving photosynthetic capacity by FBP/SBPase was better than by CcaA, and only when CcaA was co-transformed with FBP/SBPase was the synergistic effect observed. The multigene-stacking approaches and their synergistic action for improving the photosynthetic capacity in rice are discussed.  相似文献

Genetic variation in CACNA1C, which codes for the L‐type calcium channel (LTCC) Cav1.2, is associated with clinical diagnoses of bipolar disorder, depression and schizophrenia. Dysregulation of the mesolimbic‐dopamine (ML‐DA) system is linked to these syndromes and LTCCs are required for normal DAergic neurotransmission between the ventral tegmental area (VTA) and nucleus accumbens (NAc). It is unclear, however, how variations in CACNA1C genotype, and potential subsequent changes in expression levels in these regions, modify risk. Using constitutive and conditional knockout mice, and treatment with the LTCC antagonist nimodipine, we examined the role of Cacna1c in DA‐mediated behaviors elicited by psychomotor stimulants. Using fast‐scan cyclic voltammetry, DA release and reuptake in the NAc were measured. We find that subsecond DA release in Cacna1c haploinsufficient mice lacks normal sensitivity to inhibition of the DA transporter (DAT). Constitutive haploinsufficiency of Cacna1c led to attenuation of hyperlocomotion following acute administration of stimulants specific to DAT, and locomotor sensitization of these mice to the DAT antagonist GBR12909 did not reach the same level as wild‐type mice. The maintenance of sensitization to GBR12909 was attenuated by administration of nimodipine. Sensitization to GBR12909 was attenuated in mice with reduced Cacna1c selectively in the VTA but not in the NAc. Our findings show that Cacna1c is crucial for normal behavioral responses to DA stimulants and that its activity in the VTA is required for behavioral sensitization. Cacna1c likely exerts these effects through modifications to presynaptic ML‐DA system function.  相似文献

Protein-protein interaction (PPI) networks of many organisms share global topological features such as degree distribution, k-hop reachability, betweenness and closeness. Yet, some of these networks can differ significantly from the others in terms of local structures: e.g. the number of specific network motifs can vary significantly among PPI networks. Counting the number of network motifs provides a major challenge to compare biomolecular networks. Recently developed algorithms have been able to count the number of induced occurrences of subgraphs with k < or = 7 vertices. Yet no practical algorithm exists for counting non-induced occurrences, or counting subgraphs with k > or = 8 vertices. Counting non-induced occurrences of network motifs is not only challenging but also quite desirable as available PPI networks include several false interactions and miss many others. In this article, we show how to apply the 'color coding' technique for counting non-induced occurrences of subgraph topologies in the form of trees and bounded treewidth subgraphs. Our algorithm can count all occurrences of motif G' with k vertices in a network G with n vertices in time polynomial with n, provided k = O(log n). We use our algorithm to obtain 'treelet' distributions for k < or = 10 of available PPI networks of unicellular organisms (Saccharomyces cerevisiae Escherichia coli and Helicobacter Pyloris), which are all quite similar, and a multicellular organism (Caenorhabditis elegans) which is significantly different. Furthermore, the treelet distribution of the unicellular organisms are similar to that obtained by the 'duplication model' but are quite different from that of the 'preferential attachment model'. The treelet distribution is robust w.r.t. sparsification with bait/edge coverage of 70% but differences can be observed when bait/edge coverage drops to 50%.  相似文献

对具超强汞富集能力的基因工程菌在重金属汞离子存在的水体中的生长状况进行了研究。实验所采用的基因工程菌由于外源基因的表达而对Hg^2 产生了一定的耐受力,而且这种耐受力经IPTG诱导后还能得到进一步的增强,耐受能力从Hg^2 浓度5mg/L提高到15mg/L。IPTG加入的时机对菌体的生长和耐受力的有效诱导是重要的。一般在OD600为0.5~0.8时加入,且加入后还应保证2h左右的诱导时间。实验结果还表明菌体在生长过程中对碳源的要求很低。  相似文献

获得特异性抗Ecp多克隆抗体,为进一步研究ecp的功能奠定基础。利用特异引物,通过RT-PCR扩增出编码Ecp蛋白的全长cDNA,克隆至谷胱甘肽S转移酶融合蛋白表达载体pGEX-4T-1(His)6C中,转染大肠杆菌DH 5α,经诱导表达后,利用谷胱甘肽琼脂糖珠从细胞裂解物中特异吸附融合蛋白,经凝血酶裂解,释放出Ecp蛋白,再经Ni-NTA亲和层析,最终获得高纯度的Ecp蛋白。用纯化的Ecp蛋白免疫新西兰家兔,亲和层析纯化抗Ecp抗体。利用该抗体进行的Western Blot结果表明:Ecp蛋白在野生型黑腹果蝇胚胎、三龄幼虫神经系统、成虫、成虫头部组织中均有明显表达,提示ecp可能是一个重要的管家基因。  相似文献
