A new process to couple amyloglucosidase (AG) to inorganic supports is described. The technique consists in activating the support with a transition metal salt according to the metal-link method and subsequent amination and linkage of the alkylamine derivative using glutaraldehyde. The various parameters susceptible of influencing the properties of the immobilized enzyme (IME) preparation are investigated. The best result are obtained when 100 mg of 1000-Å controlled porous glass (CPG) are treated with 45 mg of TiCl4 and the activated carrier aminated using a 10-g/L solution of hexamethylenediamine (HMDA) in carbon tetrachloride (10 Ml/100 mg). Preparation obtained according to the process here described show operational stabilities much superior to those of AG immobilized on the same support by the traditional metal-link method or its variations. The mechanism involved in the preparation of the amino derivative of CPG is proposed.  相似文献

To be evolutionarily stable, sexual signals need to convey information regarding the quality or motivation of individuals. This can be achieved through direct or indirect signal costs that guarantee honest communication or through signal designs that facilitate the assessment of quality. Here, I present the case for an alternative hypothesis: that signalling exposes informative mistakes. Signalling mistakes range from occasional to frequent and from slight distortions of typical signals to grossly atypical signals. Their occurrence may be enhanced by disease or stress, thus revealing individual quality or motivation, and receivers typically respond negatively to them. By this mechanism, honest communication is due to costs of developing resistance to mistakes. Therefore, the hypothesis can function independently of signal design costs, although it can also be enhanced by signal design costs when those increase the occurrence of mistakes. This hypothesis widens the scope of signals expected to be sexually selected and creates new approaches to research in sexual selection and animal communication.  相似文献

Different low molecular mass ligands have been used to identify amyloid deposits. Among these markers, the dyes Thioflavin T and Congo Red interact specifically with the beta-sheet structure arranged in a cross-beta conformation, which is characteristic of amyloid. However, the molecular details of this interaction remain unknown. When labelled with technetium-99m, the proteinase inhibitor aprotinin has been shown to represent a very important radiopharmaceutical agent for in vivo imaging of extra-abdominal deposition of amyloid in amyloidosis of the immunoglobulin type. However, no information is available as to whether aprotinin binds other types of amyloid fibrils and on the nature and characteristics of the interaction. The present work shows aprotinin binding to insulin, transthyretin, beta-amyloid peptide and immunoglobulin synthetic amyloid fibrils by a specific dot-blot ligand-binding assay. Aprotinin did not bind amorphous precipitates and/or the soluble fibril precursors. A Ka of 2.9 microM-1 for the binding of aprotinin to insulin amyloid fibrils was determined by Scatchard analysis. In competition experiments, analogues such as an aprotinin variant, a spermadhesin and the soybean trypsin inhibitor were tested and results suggest that both aprotinin and the spermadhesin interact with amyloid fibrils through pairing of beta-sheets of the ligands with exposed structures of the same type at the surface of amyloid deposits. An electrostatic component may also be involved in the binding of aprotinin to amyloid fibrils because important differences in binding constants are observed when substitutions V15L17E52 are introduced in aprotinin; on the other hand beta-sheet containing acidic proteins, such as the soybean trypsin inhibitor, are unable to bind amyloid fibrils.  相似文献

Vertebrate fossils recovered from sites nearby the Botucaraí Hill and Candelária (Caturrita Formation) depict a diverse Late Triassic tetrapod fauna from south Brazil. These records are of key importance to the biostratigraphy of the upper sections of the Rosario do Sul Group. A lithological and biostratigraphic survey on the main fossil localities of the Botucaraí Hill area confirms the occurrence of the lower Hyperodapedon and the upper Riograndia Assemblage Zones in the region, the latter yielding early saurischians. In this paper, three incomplete dinosaur specimens, an isolated sacral vertebra, an articulated left pubis–ischium and an isolated right ischium, from the ‘Botucaraí Hill’ site are described. A comparative survey suggests that these specimens have sauropodomorph affinities, but probably more primitive than typical ‘prosauropods’ from the Norian-Early Jurassic. Regardless of the phylogenetic position of Guaibasaurus as theropod or sauropodomorph, their occurrence in the Caturrita Formation, which also yielded ‘core prosauropods’ from the Santa Maria region, suggests either the survival of early members of the clade with more derived ‘prosauropods’ or that heterochronous faunas are sampled from that stratigraphic unit.  相似文献

ABSTRACT

This study evaluated visual sensitivity to luminance contrast during a daily period. Twenty-eight young male adults (M = 24.85; SD = 2.4) with normal color vision and 20/20 visual acuity participated in this study. The circadian pattern was assessed using the Karolinska Sleepiness Scale (KSS), the Pittsburgh Sleep Quality Index (PSQI), and a sleep diary. To measure the luminance contrast, we used version 11.0 of the Metropsis software with sine-element frequency stimuli for spatial frequencies of 0.2, 0.6, 1, 3.1, 6.1, 8.8, 13.2, and 15.6 cycles per degree of visual angle (cpd). The stimuli were presented on a 19-inch color cathode ray tube (CRT) video monitor with a resolution of 1024 × 786 pixels, an update rate of 100 Hz, and a photopic luminance of 39.6 cd/m2. There was a significant difference in KSS on the weekdays [χ2(2) = 20.27; p = .001] and in the luminance contrast for frequencies of 13.2 cpd [χ2(2) = 8.27; p = .001] and 15.6 cpd [χ2(2) = 13.72; p = .041]. The results showed greater stability of the measurement during the afternoon and a reduction in the visual sensitivity in the high spatial frequencies during the night.  相似文献

Agomelatine is a novel antidepressant drug with melatonin receptor agonist and 5-HT(2C) receptor antagonist properties. We analyzed whether agomelatine has antioxidant properties. Antioxidant activity of agomelatine (25, 50, or 75 mg/kg, i.p.) or melatonin (50 mg/kg) was investigated by measuring lipid peroxidation levels, nitrite content, and catalase activities in the prefrontal cortex, striatum, and hippocampus of Swiss mice pentylenetetrazole (PTZ) (85 mg/kg, i.p.), pilocarpine (400 mg/kg, i.p.), picrotoxin (PTX) (7 mg/kg, i.p.), or strychnine (75 mg/kg, i.p.) induced seizure models. In the pilocarpine-induced seizure model, all dosages of agomelatine or melatonin showed a significant decrease in TBARS levels and nitrite content in all brain areas when compared to controls. In the strychnine-induced seizure model, all dosages of agomelatine and melatonin decreased TBARS levels in all brain areas, and agomelatine at low doses (25 or 50 mg/kg) and melatonin decreased nitrite contents, but only agomelatine at 25 or 50 mg/kg showed a significant increase in catalase activity in three brain areas when compared to controls. Neither melatonin nor agomelatine at any dose have shown no antioxidant effects on parameters of oxidative stress produced by PTX- or PTZ-induced seizure models when compared to controls. Our results suggest that agomelatine has antioxidant activity as shown in strychnine- or pilocarpine-induced seizure models.  相似文献

The association of metazoan, protist, and microbial communities with Scleractinian corals forms the basis of the coral holobiont. Coral bleaching events have been occurring around the world, introducing changes in the delicate balance of the holobiont symbiotic interactions. In this study, Archaea, bacteria, and eukaryotic phototrophic plastids of bleached colonies of the Brazilian coral Siderastrea stellata were analyzed for the first time, using 16S rRNA gene libraries. Prokaryotic communities were slightly more diverse in healthy than in bleached corals. However, the eukaryotic phototrophic plastids community was more diverse in bleached corals. Archaea phylogenetic analyses revealed a high percentage of Crenarchaeota sequences, mainly related to Nitrosopumilus maritimus and Cenarchaeum symbiosum. Dramatic changes in bacterial community composition were observed in this bleaching episode. The dominant bacterial group was Alphaproteobacteria followed by Gammaproteobacteria in bleached and Betaproteobacteria in healthy samples. Plastid operational taxonomic units (OTUs) from both coral samples were mainly related to red algae chloroplasts (Florideophycea), but we also observed some OTUs related to green algae chloroplasts (Chlorophyta). There seems to be a strong relationship between the Bacillariophyta phylum and our bleached coral samples as clones related to members of the diatom genera Amphora and Nitzschia were detected. The present study reveals information from a poorly investigated coral species and improves the knowledge of coral microbial community shifts that could occur during bleaching episodes.  相似文献

Very few South American avian superspecies or species groups are composed of both forest and non‐forest taxa. The genus Lepidocolaptes comprises 8–9 species of woodcreepers, most of which are forest birds, but two species, L. angustirostris and L. souleyetii, inhabit open vegetations. Therefore, this genus should play an important role in the discussion about the relationships between forest and non‐forest South American avifaunas. Nucleotide sequences from two mitochondrial genes, cytochrome b and ND2, suggest that: (i) L. fuscus should be removed from the genus since its association with other members of this genus is poorly supported. This view has been pointed out also by morphological and behavioural data; (ii) the phylogenetic position of the open‐vegetation species within the Lepidocolaptes radiation indicate that the split between forest and non‐forest elements within this genus took place as recently as two million years ago. This result suggests that the evolutionary relationships between forest and non‐forest biotas in South America may have been more dynamic than previously thought.  相似文献
