Translation of psbA mRNA in Chlamydomonas reinhardtii chloroplasts is regulated by a redox signal(s). RB60 is a member of a protein complex that binds with high affinity to the 5'-untranslated region of psbA mRNA. RB60 has been suggested to act as a redox-sensor subunit of the protein complex regulating translation of chloroplast psbA mRNA. Surprisingly, cloning of RB60 identified high homology to the endoplasmic reticulum-localized protein disulfide isomerase, including an endoplasmic reticulum-retention signal at its carboxyl terminus. Here we show, by in vitro import studies, that the recombinant RB60 is imported into isolated chloroplasts of C. reinhardtii and pea in a transit peptide-dependent manner. Subfractionation of C. reinhardtii chloroplasts revealed that the native RB60 is partitioned between the stroma and the thylakoids. The nature of association of native RB60, and imported recombinant RB60, with thylakoids is similar and suggests that RB60 is tightly bound to thylakoids. The targeting characteristics of RB60 and the potential implications of the association of RB60 with thylakoids are discussed.  相似文献

Community structure has been widely identified as a feature of many real-world networks. It has been shown that the antigenic diversity of a pathogen population can be significantly affected by the contact network of its hosts; however, the effects of community structure have not yet been explored. Here, we examine the congruence between patterns of antigenic diversity in pathogen populations in neighbouring communities, using both a deterministic metapopulation model and individual-based formulations. We show that the spatial differentiation of the pathogen population can only be maintained at levels of coupling far lower than that necessary for the host populations to remain distinct. Therefore, identifiable community structure in host networks may not reflect differentiation of the processes occurring upon them and, conversely, a lack of genetic differentiation between pathogens from different host communities may not reflect strong mixing between them.  相似文献

Antibodies present in sera of patients with auto immune diseases (systemic Lupus erythematosus, mixed connective tissue disease) were used to react with nuclear ribonucleoproteins (HnRNPs) from normal cells and cells infected with retroviruses. Only antibodies directed against Sm and RNP antigens precipitated particles with definite spectra of small nuclear RNAs (SnRNA) and proteins. No difference could be found between infected and uninfected cells, suggesting that virus replication is dependent on normal cellular fonctions.  相似文献

Hot dry winds (Sharav in Israel) are notorious for causing controversial symptoms such as depression, discomfort, headaches, irritability and exacerbation of respiratory ailments. Daily urinalysis combined with study of the complaints of two hundred weather-sensitive patients suffering from Sharav have shown that there exist at least three different patterns of reaction: serotonin hypersecretion causing a general "Irritation Syndrome", catecholamine deficiency resulting in the "Exhaustion Syndrome", histamine and creatinine overproduction combined with clinical "Hyperthyroidism - Forme Fruste" and typical thyroid complaints. These syndromes are amenable to appropriate treatment controlled by urinalysis.
Zusammenfassung Trockene heisse Wüstenwinde (Sharav in Israel) rufen eine seltsame Mischung von Symptomen hervor wie Depression, Unwohlsein, Kopfschmerzen, Reizbarkeit und Verschlimmerung respiratorischer Beschwerden. Tägliche Urinuntersuchungen kombiniert mit dem Studium der verschiedenen Beschwerden von 200 Sharav-gefühligen Personen haben gezeigt, dass drei verschiedene Reaktionsformen vorliegen: Serotonin-Ausschüttung, die zu einem allgemeinen Irritationssyndrom führt; Katecholamin-Mangel, der zu einem typischen Erschöpfungssyndrom führt; Histamin- und Kreatinin-Ausschüttung, die mit leichtem Hyperthyreoidismus einhergehen. Letzteres Syndrom bereitet dem Patienten nur an Sharav-Tagen Beschwerden. Die Syndrome sind durch gezielte und Urin-kontrollierte Behandlung heilbar.

Resume Des vents chauds et secs provenant du désert (le Charav en Israël par exemple) sont connus pour provoquer un ensemble de symptômes particuliers tels que dépression nerveuse, indisposition, céphalées, irritabilité et aggravation des troubles respiratoires. L'analyse journalière de l'urine et l'étude des divers symptômes que présentaient 200 personnes sensibles au Charav ont montré la présence de trois formes différentes de réactions: sécrétion accrue de sérotonine qui conduit à un syndrome d'irritabilité générale; une carence en catécholamine qui se traduit par un syndrome typique d'épuisement; une sécrétion d'histamine et de créatinine parallèle à un faible hyperthyréoïdisme. Ce dernier syndrome ne provoque des troubles aux patients que durant les jours où le Charav souffle. Ces trois syndromes sont guérissables par une médication appropriée et dont l'effet est contrôlé par des analyses d'urine.

Summary With use of the positively charged, colloidal ferric oxide labelling technique for electron microscopy of sections of rabbit marrow, a reduction in labelling density on the surface of differentiating red cell precursors was demonstrated. The number of iron particles per unit length of membrane was counted. A progressive diminution in labelling density follows cell division, reaching a minimum in the orthochromatic erythroblast, from which the nucleus is expelled. A slight increase in charge density is noted in the reticulocyte, and further increase is observed with its maturation to the erythrocyte. The results indicate that biosynthesis of n-acetyl neuraminic acid stops at the earliest recognizable stage of erythroid differentiation.The Patrick E. During the course of a mild chemical peritonitis, new skeletal muscle fibers develop and persist over a twelve-month interval in the diaphragmatic peritoneum. Light and electron microscopic studies revealed that the ectopic fibers developed from myoblasts and myotubes to fully differentiated muscle cells in the same manner as normally situated skeletal muscle. The ectopic fibers were separated from the intrinsic muscle by dense connective tissue and an elastic lamina. Diaphragms taken from normal rats and transplanted to the omentum of isogeneic recipients also developed skeletal muscle neogenesis in the same ectopic location as in the normal diaphragm. Satellite cells, reactive fibroblasts in the peritoneum, mesenchymal stem cells or blood-borne myoblast precursor cells could be the source of these ectopic muscle fibers. The results of the present studies, however, cannot provide conclusive evidence for the origin of the new muscle fibers. Regardless of their source, the methods employed may represent a unique model for the development and prolonged maintenance of skeletal muscle fibers in a heterotopic location in vivo.  相似文献

A chloroplast protein disulfide isomerase (PDI) was previously proposed to regulate translation of the unicellular green alga Chlamydomonas reinhardtii chloroplast psbA mRNA, encoding the D1 protein, in response to light. Here we show that AtPDI6, one of 13 Arabidopsis thaliana PDI genes, also plays a role in the chloroplast. We found that AtPDI6 is targeted and localized to the chloroplast. Interestingly, AtPDI6 knockdown plants displayed higher resistance to photoinhibition than wild‐type plants when exposed to a tenfold increase in light intensity. The AtPDI6 knockdown plants also displayed a higher rate of D1 synthesis under a similar light intensity. The increased resistance to photoinhibition may not be rationalized by changes in antenna or non‐photochemical quenching. Thus, the increased D1 synthesis rate, which may result in a larger proportion of active D1 under light stress, may led to the decrease in photoinhibition. These results suggest that, although the D1 synthesis rates observed in wild‐type plants under high light intensities are elevated, repair can potentially occur faster. The findings implicate AtPDI6 as an attenuator of D1 synthesis, modulating photoinhibition in a light‐regulated manner.  相似文献
