Previous work had indicated that the molecular weight calculated from the equilibrium distribution of DNA in a CsCl density gradient appears to be one half the estimated value of the true molecular weight. In the present study, the sedimentation equilibrium of a polyclectrolyte in a density gradient generated by a low molecular weight electrolyte is treated in terms of the representation according to which a constant fraction of the counterions is considered permanently immobilized and the remaining fraction completely free. Equations for the preferential hydration and the molecular weight are obtained. The validity of the treatment is tested by applying it to experimental data on the equilibrium sedimentation of ?X 174 DNA in CsCl, and it is found that the molecular weight calculated in this way is in agreement with the accepted value for this molecule. Also, recalculation of published data on density gradient centrifugation of T2 DNA in CsCl according to the present treatment brings up the molecular weight to within the range of values given by other methods. 相似文献

Abstract— —High circulating levels of l -methionine produced by inclusion in the diet or parenteral injection of the amino acid caused alterations in the free amino acid pattern of liver and brain tissues. Acute effects following l -methionine injection were more pronounced than those following long term feeding where adaptation played a role. The net effect following parenteral injection was to increase the total free amino acids of liver while decreasing those of brain. Individually, hepatic levels of aspartic acid, threonine, serine, glutamine, glutamic acid, glycine, and alanine were depressed while levels of taurine, cystathionine, methionine, lysine, and ornithine were markedly elevated. Brain levels of aspartic acid, threonine, serine, glutamic acid, glycine, alanine, and γ-aminobutyric acid were markedly depressed and increased levels of cystathionine, methionine, lysine, and glutamine were observed. A generalized aminoaciduria occurred shortly after excessive methionine intake. Disruption of the free amino acid pools was of two kinds. The influence of gibberellic acid over a wide range of concentrations on the rate of elongation of root hairs of redtop grass was investigated. The rate of root hair elongation was increased by GA over the concentration range of 10?7 to 10?12 M inclusive, with peak stimulation occurring at 10?6 M. Although root hair growth was slightly accelerated by 10?6 M GA, this concentration damaged many root hairs and caused some to stop growing altogether. The identification of prompt lactose-fermenting gram-negative rods has generally relied heavily upon colonial morphology coupled with one or more indole, methyl red, Voges-Proskauer, citrate (IMViC) parameters, hydrogen sulfide, and motility. Studies were undertaken to compare diagnoses dependent solely upon the more orthodox criteria to a system for identification based upon hydrogen sulfide, ornithine decarboxylase, and citrate utilization (HOC). The results suggest that the IMViC scheme of identification is neither consistent nor applicable when applied to the current nomenclature of the above group of organisms and should be discarded, whereas the HOC system may prove to be of significant value to clinical microbiologists. 相似文献

A model of the peripheral auditory system responding to low-frequency tone stimulation is given. The model is of the type previously introduced by Weiss (1966). It includes three interconnected parts: a linear model of the ear's mechanical system, a model of the cochlear transducer, and a stochastic model of an auditory nerve fiber. The output of the model accurately mimics many characteristics of the output of some auditory nerve neurons responding to sinusoidal stimuli but is unable to successfully match all reported aspects of data obtained from other of these neurons.
