The perennial shrub Siam Weed (Chromolaena odorata (L.) R. M. King & H. Rob.), a native of the rainforests of central and southern America, is predicted to invade most countries between the Tropics of Cancer and Capricorn. Subsequent to its discovery in northern Queensland, Australia in 1994, it was unsuccessfully targeted for eradication, with early control efforts restricted to manual removal and foliar spraying with triclopyr/picloram. A chemical trial involving five herbicides and three application methods (foliar, basal bark and cut stump) was undertaken in northern Queensland to identify effective chemicals to control this weed. Of the foliar herbicides tested, fluroxypyr (70 g/100 L) provided 99% mortality, triclopyr/picloram (105/35 g/100 L) 95%, while metsulfuron‐methyl (9 g/100 L) killed 85% of the treated plants. The herbicides fluroxypyr (300 g/100 L), picloram (43 g/1 kg) and triclopyr/picloram (400/200 g/100 L) killed greater than 98% of the plants when applied as basal bark or cut stump. The integration of fire into the management strategy either as a primary or secondary control option could have a major role, given that in this study, 99% of the Siam Weed soil seed bank was located on the soil surface. 相似文献

In immature dogs after pneumonectomy (PNX), pulmonary viscous resistance is persistently elevated predominantly as a result of a high airway resistance (Raw). We examined the anatomical basis for this observation by using computerized tomography scans obtained from foxhounds 4-10 mo after right PNX. Airways of the left lower lobe were followed from generations z = 0 (trachea) to z = 12. By 4 mo post-PNX, airway length increased significantly relative to sham-operated dogs, but airway cross-sectional area (CSA) did not. By 10 mo post-PNX, average airway CSA was 24% above that in controls. Theoretically, the increased airway length and CSA should reduce lobar Raw by 50%. However, post-PNX airway dilatation did not normalize total CSA, and estimated resistance due to turbulence and convective acceleration increased threefold; i.e., the 50% reduction in lobar Raw would be offset by the loss of four of seven lobes. Thus the expected reduction in work of breathing in the whole animal is only ~30%, consistent with previously measured work of breathing in pneumonectomized dogs. The temporal dynamics of oocyte growth, plasma sex steroids and somatic energy stores were examined during a 12 month ovarian maturation cycle in captive Murray cod Maccullochella peelii peelii under simulated natural photothermal conditions. Ovarian function was found to be relatively uninhibited in captivity, with the exception that post-vitellogenic follicles failed to undergo final maturation, resulting in widespread pre-ovulatory atresia. Seasonal patterns of oocyte growth were characterised by cortical alveoli accumulation in March, deposition of lipids in April, and vitellogenesis between May and September. Two distinct batches of vitellogenic oocytes were found in Murray cod ovaries, indicating a capacity for multiple spawns. Plasma profiles of 17beta-oestradiol and testosterone were both highly variable during the maturation period suggesting that multiple roles exist for these steroids during different stages of oocyte growth. Condition factor, liver size and visceral fat stores were all found to increase prior to, or during the peak phase of vitellogenic growth. Murray cod appear to strategically utilise episodes of high feeding activity to accrue energy reserves early in the reproductive cycle prior to its deployment during periods of rapid ovarian growth. 相似文献

Oxycarotenoids, produced through the oxidation of carotenoids, play critical roles in plants. This reaction is mediated by
a specific enzyme, β;-carotene hydroxylase, which adds hydroxyl groups to the β;-rings of carotenes. To investigate the effect
of the β;-carotene hydroxylase gene (Chyb) on oxycarotenoid biosynthesis, we generated transgenicArabidopsis plants that over-expressedChyb under the control of a 35S promoter. Their levels of zeaxanthin and neoxanthin were two- to three-fold greater relative to
the WT, while that of violaxanthin, a final product in the xanlthophyll pathway, was 1.3-fold higher than the control. In
contrast, the amount of β;-carotene declined as much as 2.4-fold, depending on the particular transgenic line. Interestingly,
astaxanthin was produced in the transgenics, but not in the WT. These data suggest that, with the aid of unknown factors in
the host, carotenoids could be converted into metabolites in the astaxanthin biosynthetic pathway. Microarray analysis was
used lo identify several genes that were consistently up-or down-regulated in transgenic chyB leaves compared with the controls.
Here, we also discuss possible modifications in leaf carotenoids, and the importance of these data from a nutritional standpoint.

For over 30 years a phospholipase C enzyme called alpha-toxin was thought to be the key virulence factor in necrotic enteritis caused by Clostridium perfringens. However, using a gene knockout mutant we have recently shown that alpha-toxin is not essential for pathogenesis. We have now discovered a key virulence determinant. A novel toxin (NetB) was identified in a C. perfringens strain isolated from a chicken suffering from necrotic enteritis (NE). The toxin displayed limited amino acid sequence similarity to several pore forming toxins including beta-toxin from C. perfringens (38% identity) and alpha-toxin from Staphylococcus aureus (31% identity). NetB was only identified in C. perfringens type A strains isolated from chickens suffering NE. Both purified native NetB and recombinant NetB displayed cytotoxic activity against the chicken leghorn male hepatoma cell line LMH; inducing cell rounding and lysis. To determine the role of NetB in NE a netB mutant of a virulent C. perfringens chicken isolate was constructed by homologous recombination, and its virulence assessed in a chicken disease model. The netB mutant was unable to cause disease whereas the wild-type parent strain and the netB mutant complemented with a wild-type netB gene caused significant levels of NE. These data show unequivocally that in this isolate a functional NetB toxin is critical for the ability of C. perfringens to cause NE in chickens. This novel toxin is the first definitive virulence factor to be identified in avian C. perfringens strains capable of causing NE. Furthermore, the netB mutant is the first rationally attenuated strain obtained in an NE-causing isolate of C. perfringens; as such it has considerable vaccine potential. 相似文献

To investigate the effects of soil type on seed persistence in a manner that controlled for location and climate variables, three weed species—Gomphocarpus physocarpus (swan plant), Avena sterilis ssp. ludoviciana (wild oat) and Ligustrum lucidum (broadleaf privet)—were buried for 21 months in three contrasting soils at a single location. Soil type had a significant effect on seed persistence and seedling vigour, but soil water content and temperature varied between soils due to differences in physical and chemical properties. Warmer, wetter conditions favoured shorter persistence. A laboratory-based test was developed to accelerate the rate of seed ageing within soils, using controlled superoptimal temperature and moisture conditions (the soil-specific accelerated ageing test, SSAAT). The SSAAT demonstrated that soil type per se did not influence seed longevity. Moreover, the order in which seeds aged was the same whether aged in the field or SSAAT, with L. lucidum being shortest-lived and A. sterilis being longest-lived of the three species. 相似文献
