随着转基因技术在植物中的广泛应用,转基因沉默受到越来越多的重视。转基因沉默可发生在转录和转录后两种水平,其基本特征就是依赖于同源的重复序列。转基因的重复拷贝间,转基因与同源的内源基因间及RNA病毒与同源转基因间都会发生基因沉默。可能有不同的机制导致转基因沉默,本文综述了转基因沉默的机理研究及转基因沉默在植物抗病基因工程和植物功能基因组学方面的应用 。  相似文献

Paclitaxel is a widely used microtubule drug and cancer medicine. Here we report that by short exposure to paclitaxel at a low dose, multipolar spindles were induced in mitotic cells without centrosome amplification. Both TPX2 depletion and Aurora-A overexpression antagonized the multipolarity. Live cell imaging showed that some paclitaxel-treated cells accomplished multipolar cell division and a portion of the daughter cells went on to the next round of mitosis. The surviving cells grew into clones with varied genome content. The results indicated that an aneuploidy population could be induced by short exposure to paclitaxel at a low dose, implicating potential side effects of paclitaxel.  相似文献

Indigenous ecological knowledge (IEK) can contribute to the management of local ecosystems and landscapes. Cultural landscapes are produced by and reflect the long-term interactions between humans and nature in indigenous societies. Yunnan Province, located in southwestern China, is the homeland of many ethnic groups, and is also a refuge for numerous species of wild plants and animals. Indigenous people in Yunnan, who have rich ecological knowledge, play an important role in the conservation of local biodiversity and the region’s unique terraced agricultural landscapes. We used the Hani people and their outstanding cultural landscape of rice terraces in Yuanyang County of Yunnan Province as a case study to describe their worldview and discuss their formation of IEK; their roles in the preservation of rice landraces and in pest regulation; their management of water, forest, and soil resources; and the vertical landscape pattern and resource-circulation system that has evolved in the areas managed by the Hani. We also discuss the challenges and threats facing the Hani, their IEK, and their cultural landscape, as well as discuss the potential for integration of the Hani’s IEK with modern conservation efforts.  相似文献

The study of protein function usually requires the use of a cloned version of the gene for protein expression and functional assays. This strategy is particularly important when the information available regarding function is limited. The functional characterization of the thousands of newly identified proteins revealed by genomics requires faster methods than traditional single‐gene experiments, creating the need for fast, flexible, and reliable cloning systems. These collections of ORF clones can be coupled with high‐throughput proteomics platforms, such as protein microarrays and cell‐based assays, to answer biological questions. In this tutorial, we provide the background for DNA cloning, discuss the major high‐throughput cloning systems (Gateway® Technology, Flexi® Vector Systems, and CreatorTM DNA Cloning System) and compare them side‐by‐side. We also report an example of high‐throughput cloning study and its application in functional proteomics. This tutorial is part of the International Proteomics Tutorial Programme (IPTP12).  相似文献

The PROMOTION OF CELL SURVIVAL 1 (PCS1) gene, encoding an aspartic protease, has an important role in determining the fate of cells in embryonic development and reproduction processes in Arabidopsis. To explore the potential function of the PCS1 gene in generating reproductive sterility, we placed the PCS1 gene under the control of an 1,869-bp nucleotide sequence from the 3′ end of the second intron (AG-I) of Arabidopsis AGAMOUS and CaMV 35S (–60) minimal promoter [AG-I-35S (60)::PCS1], and introduced it into tobacco. RT–PCR results demonstrated that the PCS1 gene driven by AG-I-35S (60) chimeric promoter was expressed only in anthers and carpels in the reproductive tissues of transgenic tobacco. Compared to wild-type plants, all AG-I-35S (60) and AG-I-35S (60)::PCS1 transgenic lines showed a normal phenotype throughout the vegetative growth phase. However, during the reproductive stage, most AG-I-35S (60)::PCS1 transgenic plant anthers displayed delayed dehiscence, failed dehiscence, petalody and hypoplasia, and the pollen grains had different shapes and sizes with a distorted, shrunken, or collapsed morphology. Moreover, three transgenic lines, PCS1-1, PCS1-3 and PCS1-4, showed higher sterility than wild-type and AG-I-35S (60) transgenic plants, respectively. These results showed that the construct of AG-I-35S (60)::PCS1 was partially effective at preventing seed set and provided a novel sterility strategy.  相似文献

【目的】沿海滩涂耐盐植物重金属抗性内生细菌的筛选及其促生长潜在能力的研究有助于我们获得一些能够耐受并促进耐盐植物在被Cd2+、Pb2+、Hg2+、Cu2+,Zn2+等重金属离子污染的贫瘠的沿海滩涂上正常生长的菌株,达到既能够利用广袤的滩涂生物资源产生经济价值又能够净化生态环境的目的。【方法】以江苏南通沿海滩涂地区的4种耐盐植物为材料,采用稀释平板涂布法从中分离得到45株内生细菌,从中挑取23株代表性的菌株,对其进行抗重金属Cu2+、Pb2+、Cd2+、Zn2+,Hg2+的活性筛选;固氮、解磷、吲哚乙酸(IAA)的产生、1-氨基环丙烷-1-羧酸(ACC)脱氨酶活性等促生指标以及NaCl耐受能力的筛选。【结果】发现分离所得的大多数细菌能够耐受高浓度的Cu2+以及Pb2+,但是对Cd2+、Zn2+,Hg2+的耐受能力则较弱;26.1%的细菌具有固氮能力,21.7%的细菌具有解磷能力,60.9%的细菌能够产生IAA,39.1%的细菌含有ACC脱氨酶。对他们进行16S rRNA基因鉴定后发现,他们分属于芽胞杆菌属(Bacillus)、喜盐芽胞杆菌属(Halobacillus)、海洋芽胞杆菌属(Oceanobacillus)、微小杆菌属(Exiguobacterium)、沙雷氏菌属(Serratia)、短波单胞菌属(Brevundimonas)、弧菌属(Vibrio)、葡萄球菌属(Staphylococcus)共8个属,显示了丰富的多样性。其中菌株KLBMP 2432以及菌株KLBMP 2447为潜在的新种。【结论】沿海滩涂地区的耐盐植物内生细菌具有丰富多样的生物多样性以及促生长能力,且存在潜在的新种资源,并对重金属Cu2+,Pb2+具有较强的抗性。  相似文献

The phytohormone auxin is important in various aspects of organism growth and development. Aux/IAA genes encoding short-lived nuclear proteins are responsive primarily to auxin induction. Despite their physiological importance, systematic analysis of Aux/IAA genes in maize have not yet been reported. In this paper, we presented the isolation and characterization of maize Aux/IAA genes in whole-genome scale. A total of 31 maize Aux/IAA genes (ZmIAA1 to ZmIAA31) were identified. ZmIAA genes are distributed in all the maize chromosomes except chromosome 2. Aux/IAA genes expand in the maize genome partly due to tandem and segmental duplication events. Multiple alignment and motif display results revealed major maize Aux/IAA proteins share all the four conserved domains. Phylogenetic analysis indicated Aux/IAA family can be divided into seven subfamilies. Putative cis-acting regulatory DNA elements involved in auxin response, light signaling transduction and abiotic stress adaption were observed in the promoters of ZmIAA genes. Expression data mining suggested maize Aux/IAA genes have temporal and spatial expression pattern. Collectively, these results will provide molecular insights into the auxin metabolism, transport and signaling research.  相似文献

Most aerobic granule cultivation has been based on the sequencing batch reactor (SBR) and then the factors that affect aerobic granulations were developed in the SBR. However, little work has been done to cultivate aerobic granules in a continuous-flow bioreactor with simple structure that is realistic for engineering. This work is the first to cultivate aerobic granules in a continuous flow airlift fluidized bed reactor (CAFB) possesses a very simple structure and without settling time and starvation time controlling. The configuration of CAFB was the simplest continuous-flow aerobic granular bioreactor reported by now. The majority of granules could be formatted in the CAFB after 12 days cultivation. The effluent COD concentration maintained at 50 ± 10 mg/L for the variable COD loading rate of 3.5 g COD/L/d and 4.8 g COD/L/d, which confirmed that the CAFB performed good anti-shock abilities. CAFB performed good nitrification ability, however, little denitrification was found under the operating conditions of this study. The shear stress acting on the solid phase were hundreds of times stronger in the CAFB than in the SBR at the same aeration strength. It seems CAFB is very efficient for granulation due to the strong shear-force exertion, which is promising for continuous-flow aerobic granular bioreactor. Protein, positive to the hydrophobicity, was predominant in extracellular polymeric substances in the granules, and favored the granules formation in the CAFB combined with the polysaccharides. However, filamentous bulking always happened in 35 days operation of the CAFB, thus further study on the stability of this bioreactor is urgently necessary.  相似文献

干旱是限制向日葵生长发育的重要因素之一。为探究向日葵苗期抗旱性分子机制,该研究以向日葵K55与K58杂交构建的150个F7重组自交系群体为材料,对其在正常浇水和干旱胁迫两种水分处理条件下的叶片相对电导率、叶绿素含量、叶面积、叶片相对含水量、根长进行表型测定,利用前期建立的SNP、SSR分子标记遗传连锁图谱,通过复合区间作图法对5个抗旱相关的性状进行QTL定位。结果表明:(1)共定位到向日葵QTL位点11个,其中正常浇水条件下5个,干旱胁迫条件下6个,表型贡献率为0.768%~7.547%,且5号连锁群上定位到的QTL位点最多(3个)。(2)QTL置信区间内共筛选到62个与干旱相关的候选基因,包括位于qLA 8 1上的rna23019、rna23004、rna22661、rna22193、rna23294、rna22783和位于qCC 13 1上的rna40140,这些基因可作为后续基因克隆及功能研究的重点候选基因。该研究结果为向日葵抗旱性研究及其遗传改良奠定了基础。  相似文献
