The aim of this study is to investigate the feasibility and efficacy of PEC nanoparticles as delivery system for cancer chemotherapy. Assembly of paclitaxel-loaded nanoparticles with high loading efficiency and narrow-size distribution is successful. For non-invasive in vivo tracing, nanoparticle blends of chelator bearing poly(lactide) with PEC and PLGA are successfully prepared. Pharmacokinetic studies in mice reveal a twofold higher circulation time of PEC as compared to PLGA. A tumor model shows an accumulation of PEC NPs in cancerous tissue and a higher anti-tumor efficiency compared to the standard Taxol?, which is reflected in a significantly slower tumor growth compared to the NaCl control group.  相似文献

Proteolysis of the basement membrane and interstitial matrix occurs early in the angiogenic process and requires matrix metalloproteinase (MMP) activity. Skeletal muscle microvascular endothelial cells exhibit robust actin stress fibers, low levels of membrane type 1 (MT1)-MMP expression, and minimal MMP-2 activation. Depolymerization of the actin cytoskeleton increases MT1-MMP expression and MMP-2 activation. Rho family GTPases are regulators of actin cytoskeleton dynamics, and their activity can be modulated in response to angiogenic stimuli such as vascular endothelial growth factor (VEGF). Therefore, we investigated their roles in MMP-2 and MT1-MMP production. Endothelial cells treated with H1152 [an inhibitor of Rho kinase (ROCK)] induced stress fiber depolymerization and an increase in cortical actin. Both MMP-2 and MT1-MMP mRNA increased, which translated into greater MMP-2 protein production and activation. ROCK inhibition rapidly increased cell surface localization of MT1-MMP and increased PI3K activity, which was required for MMP-2 activation. Constitutively active Cdc42 increased cortical actin polymerization, phosphatidylinositol 3-kinase activity, MT1-MMP cell surface localization, and MMP-2 activation similarly to inhibition of ROCK. Activation of Cdc42 was sufficient to decrease RhoA activity. Capillary sprout formation in a three-dimensional collagen matrix was increased in cultures treated with RhoAN19 or Cdc42QL and, conversely, decreased in cultures treated with dominant negative Cdc42N17. VEGF stimulation also induced activation of Cdc42 while inhibiting RhoA activity. Furthermore, VEGF-dependent activation of MMP-2 was reduced by inhibition of Cdc42. These results suggest that Cdc42 and RhoA have opposing roles in regulating cell surface localization of MT1-MMP and MMP-2 activation.  相似文献

A method for adventitious shoot regeneration from leaves of micropropagated peach shoots has been developed. Apices were excised from in vitro shoot cultures of a seed-derived (juvenile) genotype (P16Cl5) and mature genotypes (Babygold 6, 842 Standard, San Giorgio and Yumyeong). Apices were cultured 21 days in the dark on a medium supplemented with 6-benzyladenine and α-naphthaleneacetic acid and then transferred to an auxin-free medium and incubated in the light for 21 days. The first four apical leaves were excised from these apices and cultured in the same way. During the dark incubation period, leaves developed a callus at the petiolar base. Adventitious shoots appeared on this callus after transfer to auxin-free medium and culture under light conditions. The morphogenic ability of the callus was maintained for several months.  相似文献

In heterogeneous habitats with limited resources, spacing behaviour will affect individual variation in breeding success and density of populations, and is thus of general interest to ecologists. We investigated how red squirrels Sciurus vulgaris adapt their social organisation to tine‐grained heterogeneity in habitat quality, studying spacing behaviour, habitat use and population dynamics in a forest in north Italy, characterised by a mosaic of high‐quality (chestnut‐pine) and poor‐quality patches. We compared the data with those from more homogeneous broadleaf and mixed woodlands with similar overall tree seed abundance (“stable” habitats). Squirrels lived at lower densities (pre‐breeding density 0.39‐0.58 ha‐1) than in “stable” habitats, although breeding rate was not reduced. Female breeding success was related to being primiparous as yearlings, and increased with body mass and proportion high‐quality habitat in the home range. Persistency rate of females was as in stable habitats, It was higher than male persistency, but immigration and recruitment rates were male biased, resulting in even sex‐ratio. All residents occupied high‐quality patches, and no subadults established a permanent home range in poor‐quality habitat. Home range and core‐area size was typically larger in males than in females and a male's core‐area overlapped those of other males and of females. Female core‐areas were overlapped by males but not. or very little, by other females (intrasexual territoriality). Home ranges, or core‐areas, were not smaller than in “stable” habitats, nor did we find a higher degree of core‐area overlap. We conclude that in patchy habitats dominant, resident red squirrels exclude dispersing animals from preferred, high‐quality habitat, producing a spacing pattern referred to as ideal despotic distribution, and that poor patches were only used temporarily by transient individuals, resulting in a reduction of density in comparison to populations in “stable” habitats.  相似文献
