Scorpion depressant beta-toxins show high preference for insect voltage-gated sodium channels (Na(v)s) and modulate their activation. Although their pharmacological and physiological effects were described, their three-dimensional structure and bioactive surface have never been determined. We utilized an efficient system for expression of the depressant toxin LqhIT2 (from Leiurus quinquestriatushebraeus), mutagenized its entire exterior, and determined its X-ray structure at 1.2 A resolution. The toxin molecule is composed of a conserved cysteine-stabilized alpha/beta-core (core-globule), and perpendicular to it an entity constituted from the N and C-terminal regions (NC-globule). The surface topology and overall hydrophobicity of the groove between the core and NC-globules (N-groove) is important for toxin activity and plays a role in selectivity to insect Na(v)s. The N-groove is flanked by Glu24 and Tyr28, which belong to the "pharmacophore" of scorpion beta-toxins, and by the side-chains of Trp53 and Asn58 that are important for receptor site recognition. Substitution of Ala13 by Trp in the N-groove uncoupled activity from binding, suggesting that this region of the molecule is also involved in "voltage-sensor trapping", the mode of action that typifies scorpion beta-toxins. The involvement of the N-groove in recognition of the receptor site, which seems to require a defined topology, as well as in sensor trapping, which involves interaction with a moving channel region, is puzzling. On the basis of the mutagenesis studies we hypothesize that following binding to the receptor site, the toxin undergoes a conformational change at the N-groove region that facilitates the trapping of the voltage-sensor in its activated position. 相似文献

Mouse mammary tumor virus (MMTV) is a milk-transmitted betaretrovirus that causes mammary tumors in mice. Although mammary epithelial cells are the ultimate targets of MMTV, the virus utilizes components of the host immune system to establish infection. Previous studies indicated that dendritic cells play a role in MMTV infection. Here we show that dendritic cells are the first cells to be infected by MMTV in vivo and that they are capable of producing infectious virus that can be transmitted to other cell types. Moreover, upon contact with the virus, dendritic cells became more mature and migrated in response to the chemokine macrophage inflammatory protein 3beta. Finally, we demonstrate that targeted ablation of dendritic cells in vivo dramatically attenuated MMTV infection. These data indicate that MMTV infection of dendritic cells is critical to initial propagation of the virus in vivo. 相似文献

The mountain gazelle (Gazella gazelle), Dorcas gazelle (Gazella Dorcas) and acacia gazelle (Gazella arabica acacia) were historically abundant in the southern Levant, and more specifically in Israel. Anthropogenic and natural changes have caused a rapid decline in gazelle populations, raising concerns about their conservation status and future survival. The genetic profile of 111 wild gazelles from Israel was determined based on three regions of mitochondrial DNA (control region, Cytochrome b and 12S ribosomal RNA) and nine nuclear microsatellite markers. Genetic analysis of the mountain gazelle population, the largest known population of this rare species, revealed adequate diversity levels and gene flow between subpopulations. Nevertheless, ongoing habitat degradation and other human effects, such as poaching, suggest the need for drastic measures to prevent species extinction. Dorcas gazelles in Israel displayed inbreeding within subpopulations while still maintaining considerable genetic diversity overall. This stable population, represented by a distinctive genetic profile, is fragmented and isolated from its relatives in neighboring localities. Based on the genetic profile of a newly sampled subpopulation in Israel, we provide an alternative hypothesis for the historic dispersal of Dorcas gazelle, from the Southern Levant to northern Africa. The small acacia gazelle population was closest to gazelles from the Farasan Islands of Saudi Arabia, based on mitochondrial markers. The two populations did not share haplotypes, suggesting that these two populations may be the last remnant wild gazelles of this species worldwide. Only a dozen acacia gazelles survive in Israel, and urgent steps are needed to ensure the survival of this genetically distinctive lineage. The genetic assessments of our study recognize new conservation priorities for each gazelle species in the Southern Levant. 相似文献

Background and Aims In Cactaceae, the areole is the organ that forms the leaves, spines and buds. Apparently, the genus Echinocereus develops enclosed buds that break through the epidermis of the stem adjacent to the areole; this trait most likely represents a synapomorphy of Echinocereus. The development of the areole is investigated here in order to understand the anatomical modifications that lead to internal bud development and to supplement anatomical knowledge of plants that do not behave according to classical shoot theory.Methods The external morphology of the areole was documented and the anatomy was studied using tissue clearing, scanning electron microscopy and light microscopy for 50 species that represent the recognized clades and sections of the traditional classification of the genus, including Morangaya pensilis (Echinocereus pensilis).Key Results In Echinocereus, the areole is sealed by the periderm, and the areole meristem is moved and enclosed by the differential growth of the epidermis and surrounding cortex. The enclosed areole meristem is differentiated in a vegetative or floral bud, which develops internally and breaks through the epidermis of the stem. In Morangaya pensilis, the areole is not sealed by the periderm and the areole meristem is not enclosed.Conclusions The enclosed areole meristem and internal bud development are understood to be an adaptation to protect the meristem and the bud from low temperatures. The anatomical evidence supports the hypothesis that the enclosed bud represents one synapomorphy for Echinocereus and also supports the exclusion of Morangaya from Echinocereus. 相似文献

During the 2012 and 2013 growing seasons, a disease was detected on potted laurustinus (Viburnum tinus) plants in two nurseries located in the Catania province (eastern Sicily, Italy). ‘Cylindrocarpon’‐like species were consistently recovered from crown rot and stem rot tissues. Based on morphological characteristics, DNA sequencing and phylogenetic analysis of β‐tubulin (TUB), histone H3 (HIS3) and translation elongation factor‐1α (TEF‐1 α) gene sequences, the fungi associated with symptomatic tissues were identified as ‘Cylindrocarpon’ pauciseptatum, Ilyonectria novozelandica and I. torresensis. Koch's postulates were fulfilled by pathogenicity tests carried out on potted V. tinus cuttings. To our knowledge, this is the first report worldwide of ‘C.’ pauciseptatum, I. novozelandica and I. torresensis causing disease on V. tinus. 相似文献

The southern elephant seal (SES; Mirounga leonina) has a circumpolar distribution, breeding mainly on sub-Antarctic islands and making long trips between breeding or molting and foraging areas. Most individuals from colonies in the South Shetland Islands (western Antarctic Peninsula; WAP) are distributed in Antarctic Specially Protected Areas (ASPA). Despite these protected habitats, pollutants can reach such remote areas far away from emission sources, affecting local fauna. To assess possible mercury (Hg) contamination in SES, we analyzed skin samples collected from free-ranging molting individuals using the remote biopsy PAXARMS system in Isla 25 de Mayo/King George Island (62°15′S, 58°39′W; ASPA 132). Hairless skin samples were analyzed to determine total-Hg (THg) concentrations, which ranged between 145 ng/g and 1,915 ng/g (M = 730, SD = 388 ng/g), showing significant differences between sexes, with adult-females having higher concentrations (range = 306–1,915, M = 859, SD = 427 ng/g dw) than subadult-males (range = 145–1,645, M = 629, SD = 329 ng/g dw). These differences may be explained mainly by feeding-niche partitioning between sexes. Females prefer mesopelagic prey or prey associated with sea-ice in the WAP, which are enriched in methylmercury. These results provide insight regarding Hg contamination in top Antarctic predators like SES, and the need to monitor for potential effects of Hg contamination in Antarctic marine mammals. 相似文献

Although agriculture dominates much of Central America, little is known about the bat assemblages that occur within agricultural landscapes and how bats use different types of tree cover within these landscapes. Using mist-nets and a mark-recapture protocol, we compared bat diversity and movement across six types of tree cover within an agricultural landscape in central Nicaragua. The tree cover types surveyed included secondary forests, riparian forests, forest fallows, live fences, pastures with high tree cover and pastures with low tree cover. We captured a total of 3084 bats of 39 species, including two new species records for the country ( Lonchorhina aurita and Molossops greenhalli ). Of these, 2970 bats and 27 species were in the Phyllostomidae family. There were significant differences in mean species density, abundance and evenness of phyllostomid bats across the different types of tree cover, but not in bat diversity. Riparian forests had the highest mean species density and bat abundance per plot. In contrast, mean bat abundance and species density were lowest in pastures with low tree cover. Of the 1947 phyllostomid bats marked, a total of 64 bats of eight species were recaptured. The average linear distance between extra-site recaptures was 2227 m (± 228 SE) and the maximum distance was 10.6 km. Bats were recorded moving between almost all types of tree cover, and especially to and from riparian forests. Our study suggests that agricultural landscapes retaining a heterogeneous tree cover may maintain a diverse bat assemblage, and that bats visit and use a variety of tree cover types within the agricultural matrix. 相似文献
