The GTP-binding protein Septin 7 is critical for dendrite branching and dendritic-spine morphology 总被引:3,自引:0,他引:3
Septins, a highly conserved family of GTP-binding proteins, were originally identified in a genetic screen for S. cerevisiae mutants defective in cytokinesis [1, 2]. In yeast, septins maintain the compartmentalization of the yeast plasma membrane during cell division by forming rings at the cortex of the bud neck, and these rings establish a lateral diffusion barrier. In contrast, very little is known about the functions of septins in mammalian cells [3, 4] including postmitotic neurons [5-7]. Here, we show that Septin 7 (Sept7) localizes at the bases of filopodia and at branch points in developing hippocampal neurons. Upon downregulation of Sept7, dendritic branching is impaired. In mature neurons, Sept7 is found at the bases of dendritic spines where it associates with the plasma membrane. Mature Sept7-deficient neurons display elongated spines. Furthermore, Sept5 and Sept11 colocalize with and coimmunoprecipitate with Sept7, thereby arguing for the existence of a Septin5/7/11 complex. Taken together, our findings show an important role for Sept7 in regulating dendritic branching and dendritic-spine morphology. Our observations concur with data from yeast, in which downregulation of septins yields elongated buds, suggesting a conserved function for septins from yeast to mammals. 相似文献

Influence of autoclaved saprotrophic fungal mycelia on proteolytic activity in ectomycorrhizal fungi
The production of proteolytic enzymes by several strains of ectomycorrhizal fungi i.e., Amanita muscaria (16-3), Laccaria laccata (9-12), L. laccata (9-1), Suillus bovinus (15-4), Suillus bovinus (15-3), Suillus luteus (14-7) on mycelia of Trichoderma harzianum, Trichoderma virens and Mucor hiemalis and sodium caseinate, yeast extract was evaluated. The strains of A. muscaria (16-3) and L. laccata (9-12) were characterized by the highest activity of the acidic and neutral proteases. Taking the mycelia of saprotrophic
fungi into consideration, the mycelium of M. hiemalis was the best inductor for proteolytic activity. The examined ectomycorrhizal fungi exhibited higher activity of acidic proteases

The localisation of specific RNAs is a widely employed mechanism to generate asymmetry in various biological systems, e.g. during embryonic development and cellular differentiation. Here, we highlight the importance of RNA localisation in mature neurons. Specific examples of mRNAs localised in neurons are those encoding Arc, beta-actin, CaMKIIalpha and MAP2. Moreover, non-coding RNAs, such as BC1/BC200 and microRNAs (miRNAs), which play important roles in the translational regulation of localised mRNAs, receive increasing attention. The process of RNA localisation, including RNP biogenesis, transport, anchoring and translational control, and the importance of RNA localisation for the function of the nervous system are discussed. 相似文献

Without a scale-down model for perfusion, high resource demand makes cell line screening or process development challenging, therefore, potentially successful cell lines or perfusion processes are unrealized and their ability untapped. We present here the refunctioning of a high-capacity microscale system that is typically used in fed-batch process development to allow perfusion operation utilizing in situ gravity settling and automated sampling. In this low resource setting, which involved routine perturbations in mixing, pH and dissolved oxygen concentrations, the specific productivity and the maximum cell concentration were higher than 3.0 × 106 mg/cell/day and 7 × 10 7 cells/ml, respectively, across replicate microscale perfusion runs conducted at one vessel volume exchange per day. A comparative analysis was conducted at bench scale with vessels operated in perfusion mode utilizing a cell retention device. Neither specific productivity nor product quality indicated by product aggregation (6%) was significantly different across scales 19 days after inoculation, thus demonstrating this setup to be a suitable and reliable platform for evaluating the performance of cell lines and the effect of process parameters, relevant to perfusion mode of culturing. 相似文献

Estuaries and coastal wetlands are critical transition zones (CTZs) that link land, freshwater habitats, and the sea. CTZs
provide essential ecological functions, including decomposition, nutrient cycling, and nutrient production, as well as regulation
of fluxes of nutrients, water, particles, and organisms to and from land, rivers, and the ocean. Sediment-associated biota
are integral to these functions. Functional groups considered essential to CTZ processes include heterotrophic bacteria and
fungi, as well as many benthic invertebrates. Key invertebrate functions include shredding, which breaks down and recycles
organic matter; suspension feeding, which collects and transports sediments across the sediment–water interface; and bioturbating,
which moves sediment into or out of the seabed. In addition, macrophytes regulate many aspects of nutrient, particle, and
organism dynamics above- and belowground. Animals moving within or through CTZs are vectors that transport nutrients and organic
matter across terrestrial, freshwater, and marine interfaces. Significant threats to biodiversity within CTZs are posed by
anthropogenic influences; eutrophication, nonnutrient pollutants, species invasions, overfishing, habitat alteration, and
climate change affect species richness or composition in many coastal environments. Because biotic diversity in marine CTZ
sediments is inherently low whereas their functional significance is great, shifts in diversity are likely to be particularly
important. Species introductions (from invasion) or loss (from overfishing or habitat alteration) provide evidence that single-species
changes can have overt, sweeping effects on CTZ structure and function. Certain species may be critically important to the
maintenance of ecosystem functions in CTZs even though at present there is limited empirical evidence that the number of species in CTZ sediments is critical. We hypothesized that diversity is indeed important to ecosystem function in marine
CTZs because high diversity maintains positive interactions among species (facilitation and mutualism), promoting stability
and resistance to invasion or other forms of disturbance. The complexity of interactions among species and feedbacks with
ecosystem functions suggests that comparative (mensurative) and manipulative approaches will be required to elucidate the
role of diversity in sustaining CTZ functions.

Ryan Lake, a 1.6-hectare basin lake near the periphery of the tree blowdown area in the blast zone 19 km north of Mount St. Helens, was studied from August to October 1980 to determine the microbial and chemical response of the lake to the eruption. Nutrient enrichment through the addition of fresh volcanic material and the organic debris from the surrounding conifer forest stimulated intense microbial activity. Concentrations of such nutrients as phosphorus, sulfur, manganese, iron, and dissolved organic carbon were markedly elevated. Nitrogen cycle activity was especially important to the lake ecosystem in regulating biogeochemical cycling owing to the limiting abundance of nitrogen compounds. Nitrogen fixation, both aerobic and anaerobic, was active from aerobic benthic and planktonic cyanobacteria with rates up to 210 nmol of N2 cm−1 h−1 and 667 nmol of N2 liter−1 h−1, respectively, and from anaerobic bacteria with rates reaching 220 nmol of N2 liter−1 h−1. Nitrification was limited to the aerobic epilimnion and littoral zones where rates were 43 and 261 nmol of NO2 liter−1 day−1, respectively. Potential denitrification rates were as high as 30 μmol of N2O liter−1 day−1 in the anaerobic hypolimnion. Total bacterial numbers ranged from 1 × 106 to 3 × 108 ml−1 with the number of viable sulfur-metal-oxidizing bacteria reaching 2 × 106 ml−1 in the hypolimnion. A general scenario for the microbial cycling of nitrogen, carbon, sulfur, and metals is presented for volcanically impacted lakes. The important role of nitrogen as these lakes recover from the cataclysmic eruption and proceed back towards their prior status as oligotrophic alpine lakes is emphasized. 相似文献

Role of divalent metal ions in the hammerhead RNA cleavage reaction. 总被引:32,自引:0,他引:32
A hammerhead self-cleaving domain composed of two oligoribonucleotides was used to study the role of divalent metal ions in the cleavage reaction. Cleavage rates were measured as a function of MgCl2, MnCl2, and CaCl2 concentration in the absence or presence of spermine. In the presence of spermine, the rate vs metal ion concentration curves are broader, and lower concentrations of divalent ions are necessary for catalytic activity. This suggests that spermine can promote proper folding of the hammerhead and one or more divalent ions are required for the reaction. Six additional divalent ions were tested for their ability to support hammerhead cleavage. In the absence of spermine, rapid cleavage was observed with Co2+ while very slow cleavage occurred with Sr2+ and Ba2+. No detectable specific cleavage was observed with Cd2+, Zn2+, or Pb2+. However, in the presence of 0.5 mM spermine, rapid cleavage was observed with Zn2+ and Cd2+, and the rate with Sr2+ was increased, indicating that while these three ions could not promote proper folding of the hammerhead they were able to stimulate cleavage. These results suggest certain divalent ions either participate directly in the cleavage mechanism or are specifically involved in stabilizing the tertiary structure of the hammerhead. Additionally, an altered divalent metal ion specificity was observed when a unique phosphorothioate linkage was inserted at the cleavage site. The substitution of a sulfur for a nonbridging oxygen atom substantially reduced the affinity of an important Mg2+ ion necessary for efficient cleavage.
