The endothelin (Edn) system comprises three ligands (Edn1, Edn2 and Edn3) and their G-protein-coupled type A (Ednra) and type B (Ednrb) receptors. During embryogenesis, the Edn1/Ednra signaling is thought to regulate the dorsoventral axis patterning of pharyngeal arches via Dlx5/Dlx6 upregulation. To further clarify the underlying mechanism, we have established mice in which gene cassettes can be efficiently knocked-in into the Ednra locus using recombinase-mediated cassette exchange (RMCE) based on the Cre-lox system. The first homologous recombination introducing mutant lox-flanked Neo resulted in homeotic transformation of the lower jaw to an upper jaw, as expected. Subsequent RMCE-mediated knock-in of lacZ targeted its expression to the cranial/cardiac neural crest derivatives as well as in mesoderm-derived head mesenchyme. Knock-in of Ednra cDNA resulted in a complete rescue of craniofacial defects of Ednra-null mutants. By contrast, Ednrb cDNA could not rescue them except for the most distal pharyngeal structures. At early stages, the expression of Dlx5, Dlx6 and their downstream genes was downregulated and apoptotic cells distributed distally in the mandible of Ednrb-knock-in embryos. These results, together with similarity in craniofacial defects between Ednrb-knock-in mice and neural-crest-specific Galpha(q)/Galpha(11)-deficient mice, indicate that the dorsoventral axis patterning of pharyngeal arches is regulated by the Ednra-selective, G(q)/G(11)-dependent signaling, while the formation of the distal pharyngeal region is under the control of a G(q)/G(11)-independent signaling, which can be substituted by Ednrb. This RMCE-mediated knock-in system can serve as a useful tool for studies on gene functions in craniofacial development.  相似文献

Quantitative proteomics methods have emerged as powerful tools for measuring protein expression changes at the proteome level. Using MS‐based approaches, it is now possible to routinely quantify thousands of proteins. However, prefractionation of the samples at the protein or peptide level is usually necessary to go deep into the proteome, increasing both MS analysis time and technical variability. Recently, a new MS acquisition method named SWATH is introduced with the potential to provide good coverage of the proteome as well as a good measurement precision without prior sample fractionation. In contrast to shotgun‐based MS however, a library containing experimental acquired spectra is necessary for the bioinformatics analysis of SWATH data. In this study, spectral libraries for two widely used models are built to study crop ripening or animal embryogenesis, Solanum lycopersicum (tomato) and Drosophila melanogaster, respectively. The spectral libraries comprise fragments for 5197 and 6040 proteins for S. lycopersicum and D. melanogaster, respectively, and allow reproducible quantification for thousands of peptides per MS analysis. The spectral libraries and all MS data are available in the MassIVE repository with the dataset identifiers MSV000081074 and MSV000081075 and the PRIDE repository with the dataset identifiers PXD006493 and PXD006495.  相似文献

To date, 12 cases of heterozygous Ser72Leu mutations in the peripheral myelin protein 22 have been reported in patients suffering from severe demyelinating form of Charcot-Marie-Tooth disease (CMT1) and congenital hypomyelinating neuropathy (CHN) [MIM# 605253]. In the present study we report two cases of de novo S72L mutations in the PMP22 gene detected in patients of Polish origin suffering from CMT1 disease.  相似文献

The Chinese mitten crab Eriocheir sinensis is a non-native species that appeared in European waters more than 100 years ago. Size and sex structure as well as biological condition of populations of Eriocheir sinensis sampled annually between 1999–2014 from the eastern Baltic Sea coast (1999–2007 Gulf of Gdansk and 2008–2014 Vistula Lagoon, Poland) were analysed. Males were dominant over females, with a sex ratio of 1.6:1. Carapace width of all collected specimens (n?=?467) ranged from 33.3 to 89.1?mm and the mean values for specimens from Gulf of Gdansk and Vistula Lagoon were 62.4?±?8.1?mm (n?=?171) and 66.6?±?7.7?mm (n?=?296) respectively. There were no statistically significant differences in carapace width between males and females. It is important to note that there were no individuals with a carapace width less than 30?mm, which may indicate a lack of an established population of this species from the eastern Polish coast. The wet weight of all specimens ranged from 30.9 to 321.9?g and the mean values for all specimens from the Gulf of Gdansk and the Vistula Lagoon were 119.4?±?45.2?g (n?=?101) and 141.8?±?47.9?g (n?=?223), respectively. The wet weight of an individual was significantly correlated to carapace width and the exponent b reaching values close to 3 may indicate their good condition.  相似文献

A series of new 3′-O- and 5′-O-propargyl derivatives of 5-fluoro-2′-deoxyuridine (14) was synthesized by means of propargyl reaction of properly blocked nucleosides (2,4), followed by the deprotection reaction with ammonium fluoride. The synthesized propargylated 5-fluoro-2′-deoxyuridine analogues (14) were evaluated for their cytotoxic activity in three human cancer cell lines: cervical (HeLa), oral (KB) and breast (MCF-7), using the sulforhodamine B (SRB) assay. The highest activity and the best SI coefficient in all of the investigated cancer cells were displayed by 3′-O-propargyl-5-fluoro-2′-deoxyuridine (1), and its activity was higher than that of the parent nucleoside. The other new compounds exhibited moderate activity in all of the used cell lines.  相似文献

The structures of the title alpha (1) and beta (2) anomers of ethyl 3-azido-2,3-dideoxy-d-arabino-exopyranoside (C(8)H(15)N(3)O(4)) are reported. The single-crystal structures of C(8)H(15)N(3)O(4) were determined by X-ray crystallography at 293K. It has been found that both title compounds crystallize in the orthorhombic space group. In both cases, the unit cell contains four asymmetric molecules. From intensity measurements, it has been shown that each of these molecules adopts a (4)C(1) chair conformation. The packing arrangement in the unit cell displays a stratified structure. Moreover, medium strength O-H...O hydrogen bonds in both crystal lattices can be observed.  相似文献
