Phytoplankton acclimates to irradiance by regulating the cellular content of light‐harvesting complexes, which are nitrogen (N) rich and phosphorus (P) poor. Irradiance is thus hypothesised to influence the cellular N : P ratio and the N : P defining the threshold between N and P limitation (the ‘optimal’ N : P). We tested this hypothesis by first addressing the response of the optimal N : P to irradiance in a controlled experiment with Chlamydomonas reinhardtii. Then, we did a meta‐analysis of experimental data on optimal and cellular N : P ratios across light gradients to test the generality of an N : P to light response within species. In both the experiment and the meta‐analysis, N : P ratios decreased with irradiance, indicating that factors affecting underwater irradiance, like depth and the composition of the water, may influence the relative N : P requirement. The effect of irradiance did not differ between optimal and cellular N : P ratios, but observations of optimal N : P were on average 2.8 times higher than observations of cellular N : P.  相似文献

Gold nanoparticle mediated (GNOME) laser transfection is a powerful technique to deliver small biologically relevant molecules into cells. However, the transfection of larger and especially negatively charged DNA remains challenging. The efficiency for pDNA was 0.57% using parameter that does not influence the endo‐ and exogenous DNA. In order to gain a deeper understanding of the actual molecule uptake process, the uptake efficiency was determined using molecules of different sizes. It was evaluated that uncharged dextran molecules (2000 kDa) were delivered with an efficiency of 68%. The intracellular distribution of injected molecules was visualized and larger molecules were primary found in the cytoplasm. Patch clamp measurements suggested a permeabilization time up to 15 minutes. The uptake efficiency depended on the size and charge of the molecule to deliver as well as the cell size. A minor role for transfection plays the cell type since primary stem cells were successfully transfected.

Coccolithus huxleyi propagates vegetatively by simple constriction of the cell including the cover. Possibly a sexual life cycle also exists consisting of an alternation between a coccolith-covered cell and a scaly flagellate, the structure of which is presented in this paper. Abstract

In a cross-sectional study, the serum concentrations of inhibin B and prolactin of 96 male current welders were compared with the concentrations measured in 96 age-matched referents. Also, 23 patients who were all former welders diagnosed as having welding-related manganism were studied. The current welders’ geometric mean (GM) airborne exposure to manganese (Mn) was 121 µg m?3 (range 7–2320). The serum concentrations of prolactin adjusted for age and smoking habits (GM 193 mIU l?1 vs. 166 mIU l?1; p=0.047) and inhibin B adjusted for alcohol consumption (arithmetic mean (AM) 151 ng l?1 vs. 123 ng l?1; p=0.001) were higher in the welders compared with the referents. The whole blood Mn concentration was associated with the serum prolactin concentrations. Tobacco smoking resulted in lower serum prolactin concentrations. The GM serum prolactin concentrations of the patients did not significantly differ from that of the referents, but their AM serum inhibin B concentration was statistically significantly lower. The results may suggest an effect of Mn on the pituitary that is reversible upon cessation of exposure. Lower inhibin B concentrations in the patients could point to a functional impairment of the testicular Sertoli cells, that may be caused by a welding fume component or other factors in their work environment.  相似文献

Summary Three cellulose-negative (Cel-) mutants of Acetobacter xylinum strain ATCC 23768 were complemented by a cloned 2.8 kb DNA fragment from the wild type. Biochemical analysis of the mutants showed that they were deficient in the enzyme uridine 5-diphosphoglucose (UDPG) pyrophosphorylase. The analysis also showed that the mutants could synthesize (1-4)-glucan in vitro from UDPG, but not in vivo from glucose. This result was expected, since UDPG is known to be the precursor for cellulose synthesis in A. xylinum. In order to analyze the function of the cloned gene in more detail, its biological activity in Escherichia coli was studied. These experiments showed that the cloned fragment could be used to complement an E. coli mutant deficient in the structural gene for UDPG pyrophosphorylase. It is therefore clear that the cloned fragment must contain this gene from A. xylinum. This is to our knowledge the first example of the cloning of a gene with a known function in cellulose biosynthesis from any organism, and we suggest the gene be designated celA.  相似文献

Atlantic cod and spotted wolffish fry were fed high-M alginate containing feed for 59 and 55 days, respectively. During this period the fry showed a higher specific growth rate compared to controls. Uptake and distribution of alginate was studied by inclusion of the (125)I-labelled molecule in the feed. The stomach and intestine contained the highest amount while the kidney, liver and spleen contained some, indicating that the alginate was taken up by the gut and transported to internal organs. Cod fry fed 0.06% and 0.1% high-M alginate showed a death rate of 51.4% and 53.3%, respectively. The lowest mortality, 48.1%, was found in fry fed 0.01% high-M alginate. Controls showed a mortality rate of 49.0%. Differences were, however, not statistically significant. Challenge of the immunostimulated fry (fed 0.02% and 0.06% alginate for 62 days) with atypical Aeromonas salmonicida bacteria resulted in accumulated mortalities of 56% and 49%, respectively, 47 days after infection. The group that received 0.06% alginate for a shorter period (47 days) and then control feed until challenged, and the group that received alginate by bath reached a cumulative mortality of 59% and 60%, respectively. Lowest mortality (44%) was seen in the control group. Numerous microabscesses were found in both immunostimulated and control fish in secondary lamellae of the gills, haematopoietic tissues of the kidneys, the submucosa and mucosa of the intestine, the spleen, the liver and the myocardium of the heart.  相似文献

The dynamics of seston and dissolved elements in a meromictic lake with high concentrations of manganese and iron in the monimolimnion were studied through an annual cycle. This publication presents results for assimilation, sedimentation and recovery of nutrients (C, N, P, and Si) in the trophogenic zone. Phosphorus deficiency kept the productivity of the diatom dominated phytoplankton at an oligotrophic level. High concentrations of iron in influent streams and redistribution followed by precipitation of iron during periods of partial turnover removed phosphorus from the water. High concentrations of manganese and sulfate did not have the anticipated fertilizing effect, and recovery of nutrients from the depth of the lake was negligible. Mass balance calculations indicate that liberation of phosphorus from the sediments in the trophogenic zone was most important for the maintenance of primary production. 75% of carbon, 80% of nitrogen and 25% of phosphorus assimilated by the phytoplankton was mineralized in the trophogenic zone. Silica was effectively regenerated from the littoral zone during the decline of diatom blooms. Nitrogen and silica retention was 45% of the external load compared to 66% for phosphorus.Dept. of Limnology University of Oslo  相似文献

Physiol. 83(3): 810-816, 1997.In microgravity (µG) humans have marked changes in bodyfluids, with a combination of an overall fluid loss and aredistribution of fluids in the cranial direction. We investigatedwhether interstitial pulmonary edema develops as a result of a headwardfluid shift or whether pulmonary tissue fluid volume is reduced as aresult of the overall loss of body fluid. We measured pulmonary tissuevolume (Vti), capillary blood flow, and diffusing capacity in foursubjects before, during, and after 10 days of exposure to µG duringspaceflight. Measurements were made by rebreathing a gas mixturecontaining small amounts of acetylene, carbon monoxide, and argon.Measurements made early in flight in two subjects showed no change inVti despite large increases in stroke volume (40%) and diffusingcapacity (13%) consistent with increased pulmonary capillary bloodvolume. Late in-flight measurements in four subjects showed a 25%reduction in Vti compared with preflight controls(P < 0.001). There was aconcomittant reduction in stroke volume, to the extent that it was nolonger significantly different from preflight control. Diffusingcapacity remained elevated (11%; P < 0.05) late in flight. These findings suggest that, despiteincreased pulmonary perfusion and pulmonary capillary blood volume,interstitial pulmonary edema does not result from exposure to µG.
