During B- and T-cell ontogeny, extensive apoptosis occurs at distinct stages of development. Agents that increase intracellular levels of cAMP induce apoptosis in thymocytes and mature B cells, prompting us to investigate the role of cAMP signaling in human CD10+ B-precursor cells. We show for the first time that forskolin (which increases intracellular levels of cAMP) increases apoptosis in the CD10+ cells in a dose-dependent manner (19%–94% with 0–1,000 μM forskolin after 48 hours incubation, IC50 = 150 μM). High levels of apoptosis were also obtained by exposing the cells to the cAMP analogue 8-chlorophenylthio-cAMP (8-CPT-cAMP). Specific involvement of cAMP-dependent protein kinase (PKA) was demonstrated by the ability of a cAMP antagonist, Rp-isomer of 8-bromo-adenosine- 3′, 5′- monophosphorothioate (Rp-8-Br-cAMPS), to reverse the apoptosis increasing effect of the complementary cAMP agonist, Sp-8-Br-cAMPS. Furthermore, we investigated the expression of Bcl-2 family proteins. We found that treatment of the cells with forskolin or 8-CPT-cAMP for 48 hours resulted in a fourfold decline in the expression of Mcl-1 (n = 6, P = 0.002) compared to control cells. The expression of Bcl-2, Bcl-xl , or Bax was largely unaffected. Mature peripheral blood B cells showed a smaller increase in the percentage of apoptotic cells in response to 8-CPT-cAMP (1.3-fold, n = 6, P = 0.045) compared to B-precursor cells, and a smaller decrease in Mcl-1 levels (1.5-fold, n = 4, P = 0.014). Taken together, these findings show that cAMP is important in the regulation of apoptosis in B-progenitor and mature B cells and suggest that cAMP-increased apoptosis could be mediated, at least in part, by a decrease in Mcl-1 levels. The intracellular distribution of serine hydroxymethyltransferase (EC 2.1.2.1) was studied in young wheat ( Triticum aestivum L. cv. Starke II) leaves by fractionation of protoplasts and further purification of peroxisomes and chloroplasts. Essentially all of the activity in wheat leaves was located in the mitochondria. Within the mitochondria the enzyme was mainly in the matrix as shown by centrifugation of sonicated wheat mitochondria. In the C4 plants, Zea mays (L. cv. Earliking), Panicum miliaceum and Panicum maximum (cv. Australia) belonging to different C4 types, serine hydroxymethyltransferase was almost exclusively found in bundle sheath cells. The location of this enzyme in leaves is consistent with its role relative to glycine decarboxylation during photorespiration.  相似文献

A mitochondrial serine hydroxymethyltransferase (EC 2.1.2.1) has for the first time been purified close to homogeneity from a photosynthetically active tissue, spinach ( Spinacea oleracea L. cv Viking II) leaves. The specific activity of the enzyme was 7.8 μmol (mg protein)−1 min−1 using L-serine as substrate. The enzyme was stable for at least 8 weeks at 4°C in the presence of folate. The pH optimum was at pH 8.5 where the enzyme had a Km for L-serine of 0.9 m M . Carboxymethoxylamine was a strong competitive inhibitor with a K1 of 1.4 μM. An absorption spectrum taken of the enzyme in the presence of glycine and tetrahydrofolate showed a peak at 492 nm, probably originating from a substrate-enzyme complex. The molecular weight obtained by gel filtration was 209 kDa. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis of the purified enzyme showed that the apparent molecular weight of the subunit was 53 kDa, indicating four subunits.  相似文献

Physiol. 83(3): 810-816, 1997.In microgravity (µG) humans have marked changes in bodyfluids, with a combination of an overall fluid loss and aredistribution of fluids in the cranial direction. We investigatedwhether interstitial pulmonary edema develops as a result of a headwardfluid shift or whether pulmonary tissue fluid volume is reduced as aresult of the overall loss of body fluid. We measured pulmonary tissuevolume (Vti), capillary blood flow, and diffusing capacity in foursubjects before, during, and after 10 days of exposure to µG duringspaceflight. Measurements were made by rebreathing a gas mixturecontaining small amounts of acetylene, carbon monoxide, and argon.Measurements made early in flight in two subjects showed no change inVti despite large increases in stroke volume (40%) and diffusingcapacity (13%) consistent with increased pulmonary capillary bloodvolume. Late in-flight measurements in four subjects showed a 25%reduction in Vti compared with preflight controls(P < 0.001). There was aconcomittant reduction in stroke volume, to the extent that it was nolonger significantly different from preflight control. Diffusingcapacity remained elevated (11%; P < 0.05) late in flight. These findings suggest that, despiteincreased pulmonary perfusion and pulmonary capillary blood volume,interstitial pulmonary edema does not result from exposure to µG.

Daphnia magna was fed the green alga Selenastrum capricornutum, cultured under four different growth conditions: (1) phosphorus limitation, (2) nitrogen limitation, (3) UV-B irradiation, and (4) no nutrient limitation, no UV-B irradiation. Contrary to non-limited algal cells, nutrient-limited cells were not efficiently assimilated. Especially, P-limited cells passed through the gut mostly intact, while N-limited cells were partly assimilated. Also, algae exposed to moderate doses of UV-B radiation (0.3 mW cm–2 of UV312) were less efficiently assimilated after being grazed. Digestibility of the algae decreased with increased UV-B exposure time. Nutrient-limited and UV-B stressed algal cells increased in volume and became granular in appearance. These changes in the algal cells, combined with changed cell wall properties, most probably reduced their digestibility.  相似文献

Hot water-soluble polysaccharides were extracted from field colonies and suspension cultures of Nostoc commune Vaucher, Nostoc flagelliforme Berkeley et Curtis, and Nostoc sphaeroides Kützing. Excreted extracellular polymeric substances (EPS) were isolated from the media in which the suspension cultures were grown. The main monosaccharides of the field colony polysaccharides from the three species were glucose, xylose, and galactose, with an approximate ratio of 2:1:1. Mannose was also present, but the levels varied among the species, and arabinose appeared only in N. flagelliforme. The compositions of the cellular polysaccharides and EPS from suspension cultures were more complicated than those of the field samples and varied among the different species. The polysaccharides from the cultures of N. flagelliforme had a relatively simple composition consisting of mannose, galactose, glucose, and glucuronic acid, but no xylose, as was found in the field colony polysaccharides. The polysaccharides from cultures of N. sphaeroides contained glucose (the major component), rhamnose, fucose, xylose, mannose, and galactose. These same sugars were present in the polysaccharides from cultures of N. commune, with xylose as the major component. Combined nitrogen in the media had no qualitative influence on the compositions of the cellular polysaccharides but affected those of the EPS of N. commune and N. flagelliforme. The EPS of N. sphaeroides had a very low total carbohydrate content and thus was not considered to be polysaccharide in nature. The field colony polysaccharides could be separated by anion exchange chromatography into neutral and acidic fractions having similar sugar compositions. Preliminary linkage analysis showed that 1) xylose, glucose, and galactose were 1→4 linked, 2) mannose, galactose, and xylose occurred as terminal residues, and 3) branch points occurred in glucose as 1→3,4 and 1→3,6 linkages and in xylose as a 1→3,4 linkage. The polymer preparations from field colonies had higher kinematic viscosities than those from correspondingsuspension cultures. The high viscosities of the polymers suggested that they might be suitable for industrial uses.  相似文献

Summary
Pili of Neisseria gonorrhoeae are correlated with Increased bacterial attachment to epithelial cells and undergo both phase and antigenic variation. Phase variation of gonococcal pili can be brought about by recombination events in the pilin structural gene, pilE , or by the on/off switch in expression of PilC, a pilus biogenesis protein for which two loci exist. We have studied the binding to epithelial cell lines and to fixed tissue sections of N. gonorrhoeae MS11 derivatives and mutants carrying structurally defined PilE and PilC proteins, in situ binding studies of N. gonorrhoeae to formalin-fixed tissue sections resulted in a binding pattern similar to that obtained using viable epithelial cell lines of different origin. Piliated gonococcal clones, containing different pilE sequences, varied dramatically from one another in their efficiencies at binding to corneal and conjunctival tissue, but bound equally well to cervical and endometrial tissues. Further, the binding data suggested that PJIC expression by itself, i.e. without pili, cannot confer bacterial binding and that expression of either PilC1 or PiiC2 does not confer different binding properties to the bacterial cells. Possible receptors for piliated gonococci were expressed in human tissues, such as cervix, endometrium, cornea, intestine, stomach, mid-brain and meninges, but not in human kidney. Pretreatment of the target tissues with Proteinase K decreased the gonococcal binding dramatically, whereas pretreatment with neuraminidase and meta-periodate, which cleave carbon-carbon linkages between vicinal hydroxyl groups in carbohydrates, did not affect attachment of gonococci. These data argue that pilus-dependent attachment of N. gonorrhoeae to human tissue may be mediated by a eukaryotic receptor having protein characteristics, and that the pilus subunit sequence may play an important role in the interaction with human cornea.  相似文献

In spite of resource limitation, five abundant species of herbivorous metazoan zooplankton in a humic lake exhibited extensive niche overlap both with regard to seasonal and spatial occurrence, time of reproductive maxima, juvenile release and food choice. Their coexistence could not be explained by modifying predation, environmental oscillations or recolonization.Laboratory bottle experiments indicated only weak interspecific interactions between the three tested species at low food levels, but negative interactions were induced at elevated food levels. Bosmina appeared as competitively inferior during enrichment with cultured algae, but as the superior species during starvation. At low nutrient levels, all species coexisted for several generations with low reproduction, in accordance with the lake situation. It was concluded that the observed niche overlaps would be promoted if; 1) Intraspecific competition is more important than interspecific competition. 2) All species are co-adapted to low nutrient availability, food is quantitatively in surplus, but qualitatively deficient (mainly recycled detritus). During such conditions, no species would be capable of obtaining a population increase until extinction of the other species. This situation may be typical of oligotrophic humic lakes, and of other localities with low predation pressure and high inputs of allochthonous particulate carbon.  相似文献
