Individual variation in fatty-acid and thiamine concentrations were determined in lake trout Salvelinus namaycush eggs collected at two spawning grounds in Lake Michigan. A suite of predictor variables, including spawning location, egg fatty-acid and thiamine concentrations, were used to attempt to explain cause-and-effect in early life stage mortality among S. namaycush families. Lipid and fatty-acid composition of S. namaycush eggs differed between spawning locations. Salvelinus namaycush offspring from south-western Lake Michigan were affected by a high occurrence of yolk oedema, whereas a higher frequency of early mortality syndrome (EMS) was observed among offspring from the north-western part of the lake. Random-forest regressions revealed location as the most influential predictor of yolk oedema mortality, whereas thiamine level in eggs was the strongest predictor of EMS-related mortality. Several polyunsaturated fatty acids were also found to be predictors of both mortalities. There is evidence of spatial variability in egg fatty-acid concentration among S. namaycush in Lake Michigan that, together with diminished thiamine concentration, contribute to low survival of S. namaycush progeny. 相似文献

Protonation equilibria and Cu(II) binding processes by an antifungal agent fluconazole, α-(2,4-difluorophenyl)-α-(1H-1,2,4-triazol-1-yl-methyl)-1H-1,2,4-triazole-1-ethanol, were studied using the UV-Vis, EPR and NMR spectroscopic techniques. The protonation constant of fluconazole was determined from NMR titration and attributed to N4′ nitrogen atoms using the DFT methods. The spectroscopic data suggest that at pH as low as 0.4 the first complex is formed, in which one or two Cu(II) ions are bound to one of the nitrogen atoms (N4′) from triazole rings. Above pH 1.5 each Cu(II) ion is surrounded by two nitrogen atoms (also N4′) from two different ligand molecules, forming primary monomeric complexes and above pH = 5, both dimeric or oligomeric species occur, which is well registered by the EPR technique.The mixture of Cu(NO3)2 with fluconazole in a 1:1 molar ratio in a water (pH = 4.5)/ethanol solution gave crystals of [Cu2(H2O){(C6H3-2,4-F2)(CH2N3C2H2)2C-OH}{(C6H3-2,4-F2)(CH2N3C2H2)2C-O}(NO3)](NO3)2·9(H2O). This complex is the first example of a cupric 3D polymeric structure with a fluconazole ligand coordinated via both N2′ and N4′ atoms from the same triazole rings. At higher pH values, we obtained a binuclear complex [Cu2(L)2(H2O)2(NO3)2], in which the copper(II) atoms were bridged by the oxygen atoms of the deprotonated OH group of fluconazole.The hypothetical oxidative properties of this system were also examined, however it failed to generate either reactive oxygen species or DNA scission products. 相似文献

High density lipoprotein (HDL) has many properties, which contribute to its atheroprotective role. However, some recent clinical trials have identified subjects with the progression of atherosclerosis despite normal levels of HDL cholesterol. This raises the question if all subfractions of HDL have the same properties. Moreover, recent investigations have shown that both acute and chronic inflammation may lead to structural and functional changes of HDL, which render the particles proinflammatory. Although therapeutic agents that increase HDL levels are now quite well established it is not clear whether they influence HDL quality. We review the current state of knowledge on the properties of HDL and factors/therapeutic agents which may restrain the transformation of normal HDL into dysfunctional HDL. 相似文献

The effects of drought stress on seedlings?? growth and grain yield of 13 single cross maize hybrids and 11 breeding lines and cultivars of spring triticale were studied in greenhouse and field experiments. In the field experiment, the drought susceptibility index (DSIGY) was calculated by determining the change in grain yield (GY) in conditions with two soil moisture levels (IR, irrigated; D, drought). In the greenhouse experiment the response to soil drought was evaluated using DSIDW, by determining changes in the dry weight (DW) of vegetative plant parts. Marked variations in GY and DW were observed among the studied genotypes. In control conditions, the GY and DW in drought-sensitive genotypes were higher compared to the drought-resistant ones; but in drought conditions, the decreases in GY and DW in resistant genotypes were smaller than in drought-sensitive ones. DSIGY and DSIDW revealed variations in the degree of drought tolerance among the examined maize and triticale genotypes. The values of DSIGY in the field experiment and DSIDW in the greenhouse experiment enabled a division of the studied genotypes into drought-resistant or -sensitive groups. A close correlation between DSIGY and DSIDW was found. The positive linear correlation and determination coefficients between DSIGY and DSIDW were statistically significant (P?=?0.05), being equal to R 2?=?0.614 (maize) and R 2?=?0.535 (triticale). The ranking of the studied genotypes based on DSIGY was in most cases consistent with the ranking based on DSIDW, which indicates that genetically conditioned drought tolerance is similar for plants in the seedling and reproductive growth stages or may at least partly have a common genetic background. 相似文献

Although cadmium is a well-established human carcinogen, the mechanisms by which it induces cancer are poorly understood. It is suggested that cadmium-mediated carcinogenesis may include the modulation of gene expression and signal-transduction pathways, interference with antioxidant enzymes, inhibition of DNA repair and DNA methylation, and induction of apoptosis. Nevertheless, no predominant mechanism playing a role in metal-induced carcinogenesis has been reported. In the present study, we used a pig Robertsonian translocation model, which is a cross between a wild boar and domestic pig resulting in Robertsonian translocation (37,XX,der15;17 or 37,XY,der15;17), to determine the role of cadmium sulfate in the modulation of genomic DNA-methylation status and the induction of aneuploidy. We found a cadmium-mediated increase in aneuploidy within chromosome group A and C, but not within chromosome group D containing the translocated chromosome der15,17 which indicates that translocated chromosome is not more prone to chromosomal aberrations than are other chromosomes. We suggest that cadmium-induced aneuploidy (up to 5-μM concentration) may be mediated by global DNA hypermethylation as monitored with HPLC and 5-mdC immunostaining. In addition, the cyto- and genotoxic potential of cadmium was evaluated. Cadmium sulfate was able to induce apoptosis, inhibit cell-proliferative status and expression of nucleolar organizer regions (NORs), and increase oxidative DNA damage (8-oxoG content). 相似文献
