Diaphanous-related formins are eukaryotic actin nucleation factors regulated by an autoinhibitory interaction between the N-terminal RhoGTPase-binding domain (mDiaN) and the C-terminal Diaphanous-autoregulatory domain (DAD). Although the activation of formins by Rho proteins is well characterized, its inactivation is only marginally understood. Recently, liprin-α3 was shown to interact with mDia1. Overexpression of liprin-α3 resulted in a reduction of the cellular actin filament content. The molecular mechanisms of how liprin-α3 exerts this effect and counteracts mDia1 activation by RhoA are unknown. Here, we functionally and structurally define a minimal liprin-α3 core region, sufficient to recapitulate the liprin-α3 determined mDia1-respective cellular functions. We show that liprin-α3 alters the interaction kinetics and thermodynamics of mDiaN with RhoA·GTP and DAD. RhoA displaces liprin-α3 allosterically, whereas DAD competes with liprin-α3 for a highly overlapping binding site on mDiaN. Liprin-α3 regulates actin polymerization by lowering the regulatory potency of RhoA and DAD on mDiaN. We present a model of a mechanistically unexplored and new aspect of mDiaN regulation by liprin-α3.  相似文献

How increasing urbanization affects biodiversity is one of the most understudied aspects of global change biology. It is, however, known that it may negatively affect plant population genetic diversity in numerous ways, for example through its negative effects on plant population size, between‐population connectivity, and reproductive success. Therefore, it is important to investigate to what extent different levels of urbanization result in these negative phenomena. Here we used microsatellite markers to investigate urbanization effects on the population genetic structure of 23 populations of the self‐incompatible, partially clonal herb Linaria vulgaris which were sampled across a gradient of urbanization. Clonal diversity as measured by the Pareto‐parameter varied between 1.11 and 2.97 and was negatively correlated to both the degree of urbanization and to population size. Urbanization and population size were not interrelated. The least clonally rich populations also experienced significantly reduced seed set. Irrespective of the degree of urbanization, L. vulgaris populations exhibited strong genetic differentiation (FST = 0.33) and there was no significant correlation between genetic and geographic distances, suggesting low gene flow among populations. In conclusion, we showed that urbanization negatively affected fitness of L. vulgaris populations through decreasing their clonal diversity and reproductive success, an effect that may be exacerbated by the low gene flow between populations. Although the effect was modest, the results could probably be extrapolated to bigger cities where it would be considerably more pronounced.  相似文献

Plant organelle function must constantly adjust to environmental conditions, which requires dynamic coordination. Ca2+ signaling may play a central role in this process. Free Ca2+ dynamics are tightly regulated and differ markedly between the cytosol, plastid stroma, and mitochondrial matrix. The mechanistic basis of compartment-specific Ca2+ dynamics is poorly understood. Here, we studied the function of At-MICU, an EF-hand protein of Arabidopsis thaliana with homology to constituents of the mitochondrial Ca2+ uniporter machinery in mammals. MICU binds Ca2+ and localizes to the mitochondria in Arabidopsis. In vivo imaging of roots expressing a genetically encoded Ca2+ sensor in the mitochondrial matrix revealed that lack of MICU increased resting concentrations of free Ca2+ in the matrix. Furthermore, Ca2+ elevations triggered by auxin and extracellular ATP occurred more rapidly and reached higher maximal concentrations in the mitochondria of micu mutants, whereas cytosolic Ca2+ signatures remained unchanged. These findings support the idea that a conserved uniporter system, with composition and regulation distinct from the mammalian machinery, mediates mitochondrial Ca2+ uptake in plants under in vivo conditions. They further suggest that MICU acts as a throttle that controls Ca2+ uptake by moderating influx, thereby shaping Ca2+ signatures in the matrix and preserving mitochondrial homeostasis. Our results open the door to genetic dissection of mitochondrial Ca2+ signaling in plants.  相似文献

The morphology of neuronal dendritic spines is a critical indicator of synaptic function. It is regulated by several factors, including the intracellular actin/myosin cytoskeleton and transcellular N-cadherin adhesions. To examine the mechanical relationship between these molecular components, we performed quantitative live-imaging experiments in primary hippocampal neurons. We found that actin turnover and structural motility were lower in dendritic spines than in immature filopodia and increased upon expression of a nonadhesive N-cadherin mutant, resulting in an inverse relationship between spine motility and actin enrichment. Furthermore, the pharmacological stimulation of myosin II induced the rearward motion of actin structures in spines, showing that myosin II exerts tension on the actin network. Strikingly, the formation of stable, spine-like structures enriched in actin was induced at contacts between dendritic filopodia and N-cadherin–coated beads or micropatterns. Finally, computer simulations of actin dynamics mimicked various experimental conditions, pointing to the actin flow rate as an important parameter controlling actin enrichment in dendritic spines. Together these data demonstrate that a clutch-like mechanism between N-cadherin adhesions and the actin flow underlies the stabilization of dendritic filopodia into mature spines, a mechanism that may have important implications in synapse initiation, maturation, and plasticity in the developing brain.  相似文献

Climate induced species range shifts might create novel interactions among species that may outweigh direct climatic effects. In an agricultural context, climate change might alter the intensity of competition or facilitation interactions among pests with, potentially, negative consequences on the levels of damage to crop. This could threaten the productivity of agricultural systems and have negative impacts on food security, but has yet been poorly considered in studies. In this contribution, we constructed and evaluated process‐based species distribution models for three invasive potato pests in the Tropical Andean Region. These three species have been found to co‐occur and interact within the same potato tuber, causing different levels of damage to crop. Our models allowed us to predict the current and future distribution of the species and therefore, to assess how damage to crop might change in the future due to novel interactions. In general, our study revealed the main challenges related to distribution modeling of invasive pests in highly heterogeneous regions. It yielded different results for the three species, both in terms of accuracy and distribution, with one species surviving best at lower altitudes and the other two performing better at higher altitudes. As to future distributions our results suggested that the three species will show different responses to climate change, with one of them expanding to higher altitudes, another contracting its range and the other shifting its distribution to higher altitudes. These changes will result in novel areas of co‐occurrence and hence, interactions of the pests, which will cause different levels of damage to crop. Combining population dynamics and species distribution models that incorporate interspecific trade‐off relationships in different environments revealed a powerful approach to provide predictions about the response of an assemblage of interacting species to future environmental changes and their impact on process rates.  相似文献

Large mammals are vulnerable to extinction, and respond directly to ecological gradients within the forest and to the intensity of forest product use by humans. In this study, we evaluated the effects of differences in forest structure and human occupation history on the composition of medium and large‐sized mammal assemblages of the terra‐firme forests of the Anavilhanas National Park, one of the most oligotrophic Amazonian ecosystems. Mammal surveys were conducted along 11 linear transects of 4 km, six of which were located in areas once inhabited by people and managed until the year that the park was created (over 30 years ago), and five in areas that were not inhabited at that time. We detected 469 individuals of 26 species during diurnal and nocturnal sampling, and 11 additional species outside transects. Human occupation history was strongly related to forest structure components. Fruit biomass, canopy cover and tree size were strongly associated with uninhabited areas, and influenced the structure of mammal assemblages. A direct relationship between diet category and species size was observed. Large frugivore‐herbivores and carnivores were more closely associated with areas with more fruit, larger trees and greater canopy cover. In contrast, small arboreal frugivore‐omnivores associated more closely with open canopy and smaller trees. Our study indicated that the effects of human occupation history on forest structure are still evident three decades after the removal of local people from the park. This long‐term effect can be explained by the low resilience of the Anavilhanas environments, demonstrating the fragility of mammal assemblages in face of anthropogenic variation in forest structure in the oligotrophic ecosystems of the Negro River basin.  相似文献
