An ecotope (spatial eco-space) map that considers topography and bio-organism-relevant variables emerges as an important basic framework when landscape-scale characteristics for ecosystem management and wildlife conservation are needed. A spatio-geoecological framework based on geographic information systems (GIS) and a vegetation survey were developed for wildlife habitat evaluation of national parks and applied to a representative rugged valley area of Mt. Sorak National Park in Korea. An ecotope map was classified into hundreds of types and dozens of groups by combining biological and geophysical variables. Variables included: forest vegetation type, topographic solar radiation, normalized difference vegetation index (NDVI), elevation, and anthropogenic factors, such as, streams and roads. Layers of GIS variables were produced by field surveys, modeling, satellite images, or digitalization. Vegetation surveys were carried out to identify finer-scale distribution of vegetation types in the rugged valley area. Digital forest vegetation maps from the Forestry Administrator were then modified using the field-surveyed vegetation maps. Topographic solar radiation was predicted with a daily topographic radiation model. The NDVI was calculated from the satellite imagery of a Landsat Thematic Mapper. A digital elevation model (DEM) was used and the other layers were digitized using topographical maps with a scale of 1:25000. The aim of this study is to determine the geoecological factors relating to the spatial pattern of plant community. It was cleared by the spatial pattern of environmental variables and vegetation characteristics by detrended correspondence analysis using plant species and the environmental variables of each plot. The ordination component value of the first axis shows significant regression to some environmental variables. A case study of habitat evaluation was carried out using the resultant ecotope map. The spatial distribution of potential goral habitat and vegetation characteristics were predicted and the impact of human trails on the neighboring vegetation was also examined for restoration planning. The GIS-based framework developed for wildlife habitat evaluation is useful for natural resource management and human activity control in national parks in Korea.  相似文献

以杜仲愈伤组织为材料,研究了不同接种量及培养时间、不同来源的愈伤组织及其继代次数对愈伤组织生长和次生代谢产物含量的影响。结果表明,愈伤组织继代培养时接种量在0.35g左右比较合适。愈伤组织的生长曲线大致呈S形,在20d时达到最大值,而总黄酮和绿原酸的含量均在16d时达到最大值。在继代培养中,茎和叶愈伤组织的增长量、绿原酸和总黄酮含量均在第三代达到最大值;下胚轴诱导的愈伤组织的增长量、绿原酸和总黄酮含量均在第四代达到最大值;子叶愈伤组织的增长量和总黄酮含量在第五代达到最大值,绿原酸含量于第四代达到最大值。不同来源的愈伤组织中,叶愈伤组织中绿原酸含最最高,下胚轴愈伤组织中总黄酮含量最高。  相似文献

Selected commercial and technical grade pesticides were tested against the egg, preparasite and adult stages of Agamermis unka , a nematode parasite of the brown planthopper, Nilaparvata lugens . The commercial insecticide, diazinon (LC = 0.37 ppm), was most toxic to the 50 preparasites, followed by phenthoate (LC = 0.43 ppm), BPMC (LC = 0.44 ppm), IBP 50 50 (LC = 0.46 ppm), cartap hydrochloride (LC = 0.82 ppm) and buprofezin + isoprocarb 50 50 (LC = 1.11 ppm). The least toxic commercial pesticide tested was the fungicide, pencycuron 50 (LC = 2.19 ppm). Out of 12 technical grade insecticides tested, phenthoate, monocrotophos, 50 diazinon and carbofuran (LC = 0.37-0.46 ppm) were highly toxic to the preparasites, followed by 50 buprofezin, BPMC and fenitrothion (LC = 0.74-0.86 ppm). Fenthion, etofenprox, chlorpyrifos, 50 imidacloprid and MIPC (LC = 1.11-2.19 ppm) were the technical grade insecticides least toxic 50 to the preparasites. Most preparasites survived for up to 24 h at the low insecticide concentrations (0.63 and 0.31 ppm). Preparasites that were exposed to BPMC for 24 h at concentrations as high as 5.0 ppm and survived the treatments infected brown planthopper nymphs. Four selected insecticides-chlorpyrifos, BPMC, imidacloprid and carbofuran-had significant adverse effects on A. unka egg hatching. Eggs that were in the insecticide solution for 168 h fared poorly with imidacloprid having the best survival ( > 2% of the eggs hatching at 0.04 ppm). No eggs hatched from the other insecticide treatments. Three selected insecticides, BPMC, imidacloprid and chlorpyrifos, tested against adult A. unka showed that most adults survived the exposure to the insecticides between 0.31 and 2.5 ppm. At 5.0 ppm of BPMC or chlorpyrifos none of the adults survived, whereas with imidacloprid 70% of the adults survived. Egg deposition by the surviving adults was greatly reduced in those treated with the insecticides compared with those in the controls. Imidacloprid had some negative impact on the preparasites' ability to infect BPH nymphs, but it had the least detrimental effect of the insecticides tested on preparasite survival and on the eggs and adults of A. unka .  相似文献

Using terminal deoxynucleotide transferase-mediated dUTP-digoxigenin nick end labeling (TUNEL) assay and propidium iodide-DNA staining flow cytometry assay, the effects of mouse thymic dendritic cells (MTSC4) on the process of programmed cell death of thymocytesin vitro were investigated. It was noticed that thymocytes bound to MTSC4 used in this study. That the percentages of apoptotic nuclei of the bound thymocytes on MTSC4 were much higher than those of medium-cultured thymocytes, while the bound thymocytes on mouse thymic epithelial cell (MTEC1) showed much lower percentages of apoptosis. FACS analysis quantitatively confirmed the observation. Phenotype analysis showed that MTSC4 induced the deletion of CD4 + CD8 + cells and CD4 + CD8-.cells in 18 h of coculture. The results suggest that the negative selection of medullary thymocytes may be achieved by thymic dendritic cells through their enhancing effects on apoptosis. The effects of α-MSH and cAMP on melanosomes in Cloudman S91 melanoma cells were investigated by modern stereological techniques. Cells were cultured for 4 days in medium containing α-MSH or cAMP harvested at 24 hour intervals; some were frozen for melanin assay and the reminder embedded in Epon for light and electron microscopy. Cellular and melanosomal parameters were estimated by new stereological probes. We found that both stimulators induced increases in nuclear volume, cell volume, and the volume fractions and volumes of premelanosomes (VVpm,cellVpm) and mature melanosomes (VVmm,cellVmm) and the number of mature melanosomes (Nmm). Both stimulators also caused declines in the volume of individual mature melanosomes (Vimm) the melanin content per mature melanosome unit volume and the melanin content per individual mature melanosome. The increases in the volume of individual premelanosomes and the number of premelanosomes were only induced by cAME The effect cAMP on some parameters occurred 24 hours prior to α-MSH and was more marked. The response of premelanosomes to the stimulators was more sensitive than mature melanosomes. These results suggest that both stimulators enchanced melanogenesis by increasing the VVpm,cellVVmm,cellVpm, Vmm and Nmm. The melanogenic level did not depend on the Vimm and melanin concentration in melanosomes. The maturation of premelanosomes was involved in melanogenesis induced by both stimulators, but, de novo synthesis and enlargement of premelanosomes were only stimulated by cAME It imply that exogenous cAMP may affect melanosomes, and hence melanogenesis in quantitatively or qualitatively different ways to α-MSH.  相似文献

为进一步提高口蹄疫(foot-and-mouthdisease,FMD)病毒样颗粒(virus-likeparticles,VLPs)疫苗的免疫效果,本研究采用仿生矿化方法,将Zn2+和2-甲基咪唑按照不同浓度配比制备了不同粒径的FMDV VLPs-沸石咪唑骨架-8 (zeolitic imidazolate framework-8, ZIF-8)复合物,以探究尺寸效应对免疫效果的影响。结果显示,成功制备出3种不同粒径的FMDV VLPs-ZIF-8,粒径分别约为70、100、1 000 nm。细胞毒性和组织病理学试验表明,3种复合物均具有良好的生物安全性。小鼠免疫试验表明,3种复合物均能明显提高中和抗体和特异性抗体水平,并且随着复合物体积的减小,其免疫效果也随之增强。本研究表明,ZIF-8包封FMDV VLPs可显著增强其免疫效果,且具有尺寸依赖性。  相似文献
