FLUID COMPARTMENTATION AND ELECTROLYTES OF CAT CEREBRAL CORTEX IN VITRO–II SODIUM, POTASSIUM AND CHLORIDE OF MATURE CEREBRAL CORTEX 总被引:1,自引:0,他引:1
The contents of K+, Na+ and Cl? in various incubation media and in slices of adult cat cerebral cortex incubated in vitro under a variety of conditions have been determined in conjunction with studies on slice swelling and fluid compartmentation reported in the preceding paper (Bourke and Tower , 1966). Cortical slices incubated in media containing 16 Or 27 mm-K+ exhibit contents of K+ and Na+ most nearly comparable to those found in viuo. Substitution of isethionate? For Cl? or omission of Ca2+ in such media have little effect on slice cation composition. Rb+ can effectively substitute for K+, but substitution of Li+ or choline+ for most of the naf in incubation media is associated with accumulation of these cations in slices at the expense of both K+ and Na+. Compared to values in vivo for net contents and/or concentrations of electrolytes in the non-sucrose spaces of cortical slices, conditions yielding most favourable data in vitro appeared to be incubation of cortical slices in 16 mm -K+ medium or in 27 mm -K+ medium with either omission of Ca2+ or replacement of Cl? by isethionate. Essentially complete inhibition of maintenance of K+ and extrusion of Na+ in slices of cat cerebral cortex occurs upon incubation with 10?5 or 10?4m -ouabain, with 50 per cent inhibition of cortical slice electrolyte metabolism occurring at about 8 × 10?7m -ouabain. Cortical slices incubated in 27 mm -K+ medium in the presence of 42K exhibited rates of exchange and turnover of slice K+ (in non-sucrose spaces) of 0·7 μequiv./min and 6.45 per cent respectively. In the presence of 10?5m -ouabain, a maximal ratio of slice specific activity/medium specific activity is attained within about 5 min after 42K addition, compared to >20 min for control slices. In neither case does the maximal specific activity ratio exceed about 0.85; this suggests that some 10-15 per cent of total cortical K+ comprises a “slowly exchangeable” fraction. In the presence of Ca2+ (1.3 mm ) slice oxygen consumption is markedly stimulated (39 per cent) and aerobic glycolysis is markedly depressed (54 per cent) in the presence of 10?5m -ouabain; whereas on omission of Ca2+ from incubation media, both respiration and glycolysis are normally stimulated but, with 10?5m -ouabain present, both are significantly depressed (20 per cent and 37 per cent respectively). Possible relevance of these effects to mobilization of tissue Ca2+ by ouabain and to effects of intracellular Ca2+ on mitochondrial respiratory metabolism is discussed. 相似文献

SVXOPSIS. Parasites belonging to the genus Cytamoeba have been found in amphibians from central and northern California. The infected animals and their numbers were (URODELA) Aneides flavipunctatus (2), A. lugubris (29), Batrachoseps a. atttnuatus (81). Dicamptodon ensatus (2), and (SALIENTIA) Rana b. boyli (1). The rates of infection varied between 0.6% and 29.3%, and the number of parasites per infected cell ranged from one to seven.

ABSTRACT. Paramecium calkinsi from tidal marshes survive a wide salinity range. Fluid output of contractile vacuoles of these cells decreased as salinity of the medium to which they were acclimated increased, and both pulse rate and vacuole volume were used to regulate output. When cells were first exposed to more dilute medium, contractile vacuoles greatly increased volume so that fluid output increased even though pulse rate decreased. In cells shifted to a more concentrated medium, contractile vacuole output decreased by decreasing pulse rate. The contractile vacuole is surrounded by a set of collecting structures which change form as the salinity changes. Distensible ampullae are found in media of low salinity and collecting canals are found in media of high salinity. When cells are shifted from high salinity to low, the number of ampullae increases and the number of canals decreases. When cells are shifted from low salinity to high, the number of ampullae decreases and the number of canals decreases. Other non-contracting vacuoles also appear in response to a hypoosmotic shock. These include vacuoles within the cell as well as "blisters" on the surface. The number and frequency of blisters increases with the size of the hypoosmotic shock. They detach from cells without resulting in any visible loss of cytoplasm. Non-contractile vacuoles may play a role in sequestering and removing excess water that the contractile vacuoles cannot handle. 相似文献

The somatic cortex of Spathidium spathula is described ultrastructurally. The pellicle consists of an outer membrane and an underlying alveolar system. Numerous membrane-bound mucocysts and spherical electron-opaque bodies have similar circular sites of attachment to the outer membrane. Below these are a microfibrillar zone and an underlying region of rough ER. Mitochondria are lined up under the rough ER in longitudinal cortical ridges. Parasomal sacs are found near the basal bodies and are associated with cytoplasmic membranous sacs. Various microtubular and fiber systems are associated with single basal bodies: (1) a short kinetodesmal fiber; (2) two transverse microtubular ribbons and a transverse fiber; (3) a postciliary microtubular ribbon, initially sandwiched by two fibers, which gives rise to longitudinal subpellicular microtubules extending posteriorly for a distance of some four or five basal bodies; and (4) a system of overlapping subkinetal microtubules. A three-dimensional reconstruction is included. The somatic cortex of S spathula is similar to that reported for other Haptorida of the ciliate subclass Gymnostomata. 相似文献

ABSTRACT. Moles from England were examined for coccidian oocysts and all 64 Talpa europaea were infected; of 64 infected hosts, 56 (88%) had multiple infections representing two to six coccidian species when examined. Oocysts in 31 of the 64 samples remained unsporulated. Three eimerians and one isosporan were studied from the 33 fecal samples that had sporulated oocysts and these are described as new species; Cyclospora talpae Pellérdy & Tanyi, 1968, and Isospora sofiae (Golemansky, 1978) Levine & Ivens, 1979, are redescribed; and Cyclospora sp., similar to C. talpae, is discussed. Sporulated oocysts of C. talpae are ellipsoidal, 14.3 × 9.6 (12–19 × 6–13) μm with sporocysts ovoid, 9.4 × 5.7 (6–13 × 4–8) μm; it was found in 21 of the 33 (63.6%) sporulated samples. Sporulated oocysts of Cyclospora sp. are subspheroidal to ellipsoidal, 12.5 × 8.9 (10–14 × 6–12) μm with sporocysts ovoid, 8.6 × 5.3 (6–10 × 4–6) μm; it was found in 21 of the 33 (63.6%) sporulated samples. Sporulated oocysts of Eimeria avonensis n. sp. are elongate-ellipsoidal, 15.0 × 9.6 (13–20 × 7–12) μm with sporocysts ovoid, 6.6 × 3.6 (5–9 × 3–7) μm; it was found in 15 of the 33 (45.5%) sporulated samples. Sporulated oocysts of Eimeria berea n. sp. are subspheroidal, 12.1 × 10.5 (10–15 × 8–14) μm with sporocysts ovoid, 6.3 × 3.9 (5–10 × 2–5) μm; it was found in 8 of the 33 (24.2%) sporulated samples. Sporulated oocysts of Eimeria globula n. sp. are spheroidal, 20.9 × 19.9 (19–24 × 17–21) μm with sporocysts elongate-ovoid, 11.5 × 6.9 (9–16 × 6–9) μm; it was found in 3 of the 33 (9.1%) sporulated samples. Sporulated oocysts of Isospora sporopointaea n. sp. are subellipsoidal to ellipsoidal, 17.1 × 11.4 (13–21 × 8–14) μm with sporocysts ellipsoidal with both ends pointed, 11.9 × 5.9 (9–16 × 4–8) μm; it was found in 27 of the 33 (81.8%) sporulated samples. Sporulated oocysts of I. sofiae are spheroidal to subspheroidal, 12.2 × 11.0 (9–16 × 8–15) μm with sporocysts ovoid, 9.1 × 5.2 (6–13 × 3–8) μm; it was found in 25 of the 33 (75.8%) sporulated samples. To date, the coccidian parasites of talpids include two cyclosporans, 12 eimerians, and six isosporans, exclusive of the four new species described here. 相似文献

Offspring sex ratios in wild bird populations, and the extent to which they vary from the equality expected by random genotypic sex determination, have received much recent attention. Adult sex ratios (ASRs) in wild birds, on the other hand, remain very poorly described, and many of the questions about them posed by Ernst Mayr in 1939 remain unanswered. This review assesses population-level sex ratio patterns in wild bird populations, with an emphasis on the ASR. A quantitative assessment of over 200 published estimates of ASR, covering species from a wide range of taxa, regions and habitats, supported Mayr's assertion that skewed ASRs are common in wild bird populations. On average, males outnumbered females by around 33%, and 65% of published estimates differed significantly from equality. In contrast, population-level estimates of offspring sex ratio in birds did not generally differ from equality, and mean ASR across a range of wild mammal species was strongly female-skewed. ASR distortion in birds was significantly more severe in populations of globally threatened species than in non-threatened species, a previously undescribed pattern that has profound implications for their monitoring and conservation. Higher female mortality, rather than skewed offspring sex ratio, is the main driver of male-skewed ASRs in birds, and the causes and implications of this are reviewed. While estimates of ASR in wild bird populations may be subject to a number of biases, which are discussed, there is currently no quantitative evidence that an ASR of one male to one female represents the norm in birds. A better understanding and reporting of ASRs in wild bird populations could contribute greatly to our understanding of population processes and could contribute much to theoretical and applied research and conservation. 相似文献
