SYNOPSIS. The surface membrane potentials of suctorian genus Heliophrya were studied with intracellular electrodes. Resting membrane potentials averaged -32 mV, and spontaneous depolarizing potentials occurring at apparently random intervals and having a variety of waveforms were routinely observed. Such spontaneous potentials were correlated in time with visually monitored contractile vacuole activity. Individual contractile vacuoles had unique, although somewhat variable, electrical signatures. In the presence of an intracellular electrode all vacuoles contracted independently, but at approximately the same frequency. The amplitude of the electrical potentials increased when the membrane was hyperpolarized and decreased when it was depolarized. The sign of such potentials reversed at between -10 mV and the zero membrane potential. A 20% decrease in the membrane resistance was measured at the peak of the spontaneous depolarizing potentials. 相似文献

Abstract. A revision of the Charis cleonus group of Neotropical riodinid butterflies is presented, including discussions on the taxonomy, biogeography and biology of its species, and illustrations of the adults and male and female genitalia (where known) of all taxa. We recognize twenty-two species, including nineteen new species: C. ariquemes sp.n., C. brasilia sp.n., C. breves sp.n., C. cacaulandia sp.n., C. cuiaba sp.n., C. humaita sp.n., C. ipiranga sp.n., C. iquitos sp.n., C. ma sp.n., C. manicore sp.n., C. manu sp.n., C. matic sp.n., C. maues sp.n., C. negro sp.n., C. palcazu sp.n., C. rocana sp.n., C. santarem sp.n., C. tapajos sp.n and C. tefe sp.n. A cladistic analysis using thirty-six characters of male and female genitalia and external facies generated a single most parsimonious cladogram highlighting the existence of two clades, the major and cleonus subgroups. As each species largely occupies a parapatric range, this cladogram provides the opportunity to reassess historical relationships among areas of endemism in the Amazon basin. 相似文献

无籽西瓜(Citrullus lanatus Thunb. Mansfeld, cv. Millionaire)果实在10 μL/L 1-甲基环丙烯(1-MCP)或正常空气中预贮18 h后,切取内果皮肉柱 (直径7 mm,长40 mm),再用2% CaCl2或去离子水冲淋肉柱,然后将肉柱贮藏于10 ℃。测定贮藏过程中的果肉硬度、电导率、磷脂酶D (PLD)、磷脂酶C (PLC)和脂氧合酶 (LOX)的活性变化。结果显示,2% CaCl2刺激PLD、PLC和LOX的活性,维持果肉的硬度。单独用CaCl2处理并不足以维持切割西瓜的品质,还可能因刺激脂质水解酶而发生不良的作用。1-MCP能够抵抗CaCl2对PLD、PLC和LOX的刺激作用。与对照相比,1-MCP与CaCl2结合处理能延缓西瓜果肉的衰老过程,表现出较高的果肉硬度和较低的脂质水解酶活性。 相似文献

The use of genomic techniques to address ecological questions is emerging as the field of genomic ecology. Experimentation under environmentally realistic conditions to investigate the molecular response of plants to meaningful changes in growth conditions and ecological interactions is the defining feature of genomic ecology. Because the impact of global change factors on plant performance are mediated by direct effects at the molecular, biochemical, and physiological scales, gene expression analysis promises important advances in understanding factors that have previously been consigned to the 'black box' of unknown mechanism. Various tools and approaches are available for assessing gene expression in model and nonmodel species as part of global change biology studies. Each approach has its own unique advantages and constraints. A first generation of genomic ecology studies in managed ecosystems and mesocosms have provided a testbed for the approach and have begun to reveal how the experimental design and data analysis of gene expression studies can be tailored for use in an ecological context. 相似文献

The lipid-linked precursor ofN-type glycoprotein oligosaccharides was isolated from porcine thyroid microsomes after in cubation with UDP[3H] Glucose. The carbohydrate was released from dolichol pyrophosphate by mild acid hydrolysis, purified by gel filtration and characterized by 500-MHz1H-NMR spectroscopy in combination with enzymatic degradation. The parent oligosaccharide was found to be Glc3Man9Glc-NAc2. The three glucose residues are present in the linear sequence Glcα1-2Glα1-3 Glc, the latter being α(1-3)-linked to one of the mannose residues. In order to establish the branch location of the triglucosyl unit, the parent compound was digested with jack-bean α-mannosidase. The oligosaccharide product was purified by gel filtration, and identified by1H-NMR as Glc3Man5GlcNAc2 lacking the mannose residues A, D2, B and D3. In this article, I explore the characteristics of global-change archaeology as an emerging field of research. Global-change archaeology seeks to document and apply historical knowledge of past human–environmental interactions to the understanding of contemporary environmental problems and management and planning for future sustainability. It takes place within an interdisciplinary research structure and is situated within the explanatory contexts of historical science and humanistic history with close links to historical and political ecology. Both history and agency play important roles in the practice of global-change archaeology. Lipids were extracted from fresh, field-grown coastal bermudagrass (Cynodon dactylon (L.) Pers.) and the fatty acids determined by gas chromatography. Total fatty acid levels (dry weight basis) increased during the day and reached a single maximum at sunset in 2-week-old grass; whereas, in older grass, the fluctuations in fatty acid levels showed two maxima. The first maximum occurred 4 h after sunrise and the second maximum occurred at sunset. Total fatty acid levels, based on dry weight, decreased during the first 6 weeks of growth and changed very little after an additional 4 weeks' growth in bermudagrass leaf blades. Chlorophyll levels (dry weight basis) continuously decreased during the entire growth period (10 weeks). Chlorophyll alb ratios increased at sunset in 2- and 6-week-old grass, but this ratio did not change during the day in subsequent growth stages. The results of these experiments show that stages of maturity affected fatty acid fluctuations during the day as well as total fatty acid and chlorophyll levels in Coastal bermudagrass leaf blades. Chlorophyll alb ratios varied independently of fatty acid levels. 相似文献
