Covalent dimers of alamethicin form conducting structures with gating properties that permit measurement of current-voltage (I-V) relationships during the lifetime of a single channel. These I-V curves demonstrate that the alamethicin channel is a rectifier that passes current preferentially, with voltages of the same sign as that of the voltage that induced opening of the channel. The degree of rectification depends on the salt concentration; single-channel I-V relationships become almost linear in 3 M potassium chloride. These properties may be qualitatively understood by using Poisson-Nernst-Planck theory and a modeled structure of the alamethicin pore.  相似文献

We report complete sequences of the cytochrome b, 12S rRNA, and protamine P1 genes from 18 of the 21 extant species of dasyurine marsupials (family Dasyuridae). Partial sequences are included for Pseudantechinus ningbing, but no data are available for Ps. mimulus or Phascolosorex doriae. Phylogenetic analyses of these sequences yield compatible gene trees with limited resolution. Simultaneous parsimony analysis of all three genes suggests the following: (a) a basal polytomy of eight lineages; (b) a sister-group relationship between phascolosoricine genera (Neophascogale and Phascolosorex, which are monophyletic) and Dasyums + Sarcophilus; (c) monophyly of Dasyurus viverrinus, D. albopunctatus, D. geoffroii, and D. spartacus apart from other quolls and Sarcophilus; and (d) sister-pairing of D. geoffroii and D. spartacus Previous attempts at unraveling the phylogenetic history of dasyurines have produced strikingly inconsistent results, due in part to differences in character systems examined, interpretations of character homology and independence, and analytical methods employed. We provide some evidence that the basal polytomy is the result of rapid cladogenesis and suggest that this episode of dasyurine evolution is temporally correlated with the onset of aridification following the New Guinean uplift of 15 million years ago.  相似文献

Adrenomedullin (AM) is a peptide hormone with numerous effects in the vascular systems. AM signals through the AM1 and AM2 receptors formed by the obligate heterodimerization of a G protein-coupled receptor, the calcitonin receptor-like receptor (CLR), and receptor activity-modifying proteins 2 and 3 (RAMP2 and RAMP3), respectively. These different CLR-RAMP interactions yield discrete receptor pharmacology and physiological effects. The effective design of therapeutics that target the individual AM receptors is dependent on understanding the molecular details of the effects of RAMPs on CLR. To understand the role of RAMP2 and -3 on the activation and conformation of the CLR subunit of AM receptors, we mutated 68 individual amino acids in the juxtamembrane region of CLR, a key region for activation of AM receptors, and determined the effects on cAMP signaling. Sixteen CLR mutations had differential effects between the AM1 and AM2 receptors. Accompanying this, independent molecular modeling of the full-length AM-bound AM1 and AM2 receptors predicted differences in the binding pocket and differences in the electrostatic potential of the two AM receptors. Druggability analysis indicated unique features that could be used to develop selective small molecule ligands for each receptor. The interaction of RAMP2 or RAMP3 with CLR induces conformational variation in the juxtamembrane region, yielding distinct binding pockets, probably via an allosteric mechanism. These subtype-specific differences have implications for the design of therapeutics aimed at specific AM receptors and for understanding the mechanisms by which accessory proteins affect G protein-coupled receptor function.  相似文献

The coordinated regulation of protein kinases is a rapid mechanism that integrates diverse cues and swiftly determines appropriate cellular responses. However, our understanding of cellular decision‐making has been limited by the small number of simultaneously monitored phospho‐regulatory events. Here, we have estimated changes in activity in 215 human kinases in 399 conditions derived from a large compilation of phosphopeptide quantifications. This atlas identifies commonly regulated kinases as those that are central in the signaling network and defines the logic relationships between kinase pairs. Co‐regulation along the conditions predicts kinase–complex and kinase–substrate associations. Additionally, the kinase regulation profile acts as a molecular fingerprint to identify related and opposing signaling states. Using this atlas, we identified essential mediators of stem cell differentiation, modulators of Salmonella infection, and new targets of AKT1. This provides a global view of human phosphorylation‐based signaling and the necessary context to better understand kinase‐driven decision‐making.  相似文献

Social experiences can profoundly shape social behavior and the underlying neural circuits. Across species, the formation of enduring social relationships is associated with both neural and behavioral changes. However, it remains unclear how longer‐term relationships between individuals influence brain and behavior. Here, we investigated how variation in social relationships relates to variation in female preferences for and neural responses to song in a pair‐bonding songbird. We assessed variation in the interactions between individuals in male‐female zebra finch pairs and found that female preferences for their mate's song were correlated with the degree of affiliation and amount of socially modulated singing, but not with the frequency of aggressive interactions. Moreover, variation in measures of pair quality and preference correlated with variation in the song‐induced expression of EGR1, an immediate early gene related to neural activity and plasticity, in brain regions important for auditory processing and social behavior. For example, females with weaker preferences for their mate's song had greater EGR1 expression in the nucleus Taeniae, the avian homologue of the mammalian medial amygdala, in response to playback of their mate's courtship song. Our data indicate that the quality of social interactions within pairs relates to variation in song preferences and neural responses to ethologically relevant stimuli and lend insight into neural circuits sensitive to social information. © 2016 Wiley Periodicals, Inc. An extension of the control equations discussed by Goodwin is proposed which allows for arbitrary strong coupling and for arbitrary parallel coupling of metabolic pools and genetic loci. It is demonstrated that these generalized control equations can be put into canonical form and further that Liouville's theorem applies. In addition, it is demonstrated that after a suitable canonical transformation the resulting partition function can be solved in closed form, and this result, as well as that for the mean energy, is exhibited. An actin-like protein has been isolated and purified from amoebae of Dictyostelium discoideum. The 3.7S protein polymerizes upon addition of 0.1 m KCl to a polymer of 26S. An increase in viscosity accompanies this polymerization and electron micrographs have revealed beaded, helical filaments with a diameter of 60–75 Å and an axial periodicity of 350 Å. These F-actin-like filaments produced a 5-fold activation of muscle myosin Mg-ATPase at low ionic strength. When incubated with rabbit muscle heavy meromyosin (HMM) the amoeba F-actin-like protein formed typical “arrowhead” structures with polarized binding of HMM and arrowhead spacings of 350 Å. In SDS polyacrylamide disc gel electrophoresis the purified amoeba protein migrates as a single band corresponding to a molecular weight of 48,000 daltons. The amino acid composition is very similar to that of muscle actin and includes the unusual amino acid 3-methylhistidine.  相似文献

The rates of inactivation of human rotavirus type 2 (strain Wa) (HRV-Wa) and poliovirus type 1 (strain CHAT) were compared in polluted waters (creek water and secondary effluent before chlorination) and nonpolluted waters (lake water, groundwater, and chlorinated tap water). Viral infectivity titers were determined by plaque assays, while HRV-Wa antigenicity also was monitored by an enzyme-linked immunosorbent assay. Both viruses persisted longest in lake water and shortest in tap water. The actual inactivation times (i.e., times required for two-log10 reductions of initial viral titers) for the two viruses were significantly different in all waters except tap water. With the exception of the groundwater and secondary effluent results, the HRV-Wa inactivation times in the fresh waters tested were significantly different. Owing perhaps to aggregation, HRV-Wa appeared less susceptible to the effects of chlorine than previously reported for this virus and for the simian rotavirus SA11. HRV-Wa displayed prolonged survival in lake water and groundwater exceeding that previously reported for the SA11 virus. The HRV-Wa infectivity reduction rate (ki) was significantly correlated with the water pH (i.e., as pH increased, ki increased). The water pH may have influenced viral aggregation and thereby HRV-Wa susceptibility to other virucidal factors in the water. Enzyme-linked immunosorbent assay results showed similar inactivation patterns with the most significant reduction in HRV-Wa antigenicity occurring in polluted waters and tap water. In all waters, particularly tap water, infectivity declined at a faster rate than antigenicity. It is proposed that HRV-Wa can be used as a model for future studies of rotaviral persistence in the aquatic environment.  相似文献

Datura innoxia plants were wick fed with angelic acid-[1-14C] and l-isoleucine-[U-14C] to act as a positive control. After 7 days the root alkaloids 3α-tigloyloxytropane, 3α,6β-ditigloyloxytropane, and 3α,6β-ditigloyloxytropan-7β-ol were isolated and it was determined that angelic acid is not a precursor for the tigloyl moiety of these alkaloids. Tiglic acid-[1-14C] which was fed via the roots to hydroponic cultures of Datura innoxia, was incorporated to a considerable degree after 8 days.  相似文献
