Typical preparation of seed samples for infrared (IR) microspectroscopy involves imbibition of the seed for varying time periods followed by cryosectioning. Imbibition, however, may initiate germination even at 4° C with associated changes in the chemistry of the sample. We have found that it is possible to section seeds that are sufficiently hard, such as soybeans, on a standard laboratory microtome without imbibition. The use of dry sectioning of unimbibed seeds is reported here, as well as a comparison of different mounting media and modes of analysis. Glycerol, Tissue-Tek, and ethanol were used as mounting media, and the quality of the resulting spectra was assessed. Ethanol was the preferred mountant, because it dried quickly with no residue and thus did not interfere with the spectrum of interest. Analysis in transmission mode using barium fluoride windows to hold the samples was compared with transmission-reflection analysis with sections mounted on special infrared-reflecting slides. The two modes of analysis performed well in different regions of the spectrum. The mode of analysis (transmission vs. transmission-reflection) should be based on the components of greatest interest in the sample.  相似文献

Summary Flesh flies (Sarcophagidae) collected in Costa Rica and Panama lack the pupal diapause that is characteristic of flesh flies from the temperate zone and tropical Africa. The absence of a diapause capacity in the neotropical species correlates with several other life history traits: in most species the post feeding wandering phase of the third larval instar is longer and duration is more variable, adult life is longer, clutch size is smaller, and more clutches are produced. Among species that have the capacity for diapause, risk is invested primarily in the diapausing stage and other life stages are brief. Though diapausing species are short-lived, they produce as many or more progeny than nondiapausing species by increasing clutch size. The slower and more variable developmental rate and increased adult longevity desynchronizes development and permits the nondiapausing species to spread an environmental risk over different stages of the life cycle, thus offering an alternative to diapause. Other traits such as body size, developmental velocity, thermal constant thresholds, thermal constants, age at first reproduction, and the interval between clutches do not appear related to the capacity for diapause.  相似文献

Protein synthesis is cyclic during pupal diapause in Sarcophaga crassipalpis. These cycles are in phase with infradian MO2 cycles, which have a periodicity of about 4 days at 25°C. Mean incorporation of [35S]methionine by diapausing pupae was 5.4% during the 2 days of highest MO2 but dropped to 1.7% during the 2 days of low MO2. Diapausing pupae treated with a juvenile hormone analog prior to pupariation had a constant high MO2 similar to peak values observed in untreated pupae, and such pupae consistently incorporated [35S]methionine at a high rate (7.7%). [35S]Methionine incorporation by nondiapausing pupae and pharate adults was eightfold higher than the peak rates observed during diapause. Autoradiography of in vivo labeled proteins indicated quantitative and qualitative changes in the synthesis of proteins by diapausing pupae during different phases of the MO2 cycle. Brains from diapausing pupae labeled in vitro showed higher incorporation at the peak of the MO2 cycle than at the nadir of the cycle, but no such differences were detected for integument, fat body, or fat body supernatant. Theses differences in tissue response indicate that control of protein synthesis during diapause is not cell autonomous, but is a function of the metabolism of the intact organism.  相似文献

Gross ultrastructural and histochemical details of the integumental milk glands of the tsetse fly Glossina morsitans have been examined during the pregnancy cycle. Structural evidence for protein secretion is found between Days 3-8 of the nine-day cycle: termination of activity is completed on the day of parturition. Onset of lactation is synchronized with the eclosion of the first instar larva. The changes in cell volume (notably in the extracellular reservoir) occurring throughout the pregnancy cycle are illustrated in electron micrographs, and a one hundred-fold volume increase in the reservoir volume between the inactive phase and the active period is illustrated and discussed in terms of membrane modulation of the limiting membrane of the reservoir. Intracellular membrane changes during the cycle, particularly the development of extensive ER arrays in the actively secreting cell, are illustrated and discussed. It is suggested that cytoplasmic microtubules play a part in maintaining the form of the distended secretory cell, at the height of secretory release and storage. Histochemical observations on the milk secretion, and the contents of the larval gut are presented.  相似文献

Failure of the brain to stimulate the prothoracic gland to release ecdysone has been widely regarded as the basis for diapause in insect pupae. In diapause-destined flesh flies, the absence of a peak of moulting hormone around the time of pupal head eversion supports this contention, but in addition, major pulses of juvenile hormone (JH) activity with a rhythmicity of 24 hr are unique to flesh flies destined for pupal diapause. JH activity persists during diapause, and a pulse of JH precedes the rise of moulting hormone that initiates adult development.  相似文献

A normal pregnancy in tsetse involves the successful integration of larval development with maternal activity. At 25°C, ovulation in Glossina morsitans occurs 1 hr after the previous larviposition, the egg hatches on day 3·8 (1·57 mm length, 0·09 mg dry wt.), ecdysis to second instar occurs on day 4·9 (2·3 mm, 0·30 mg), the third instar cuticle is formed on day 6·8 (4·5 mm, 5·0 mg), and parturition occurs on day 9·0 (6·0 mm, 10·0 mg). Melanization of the in utero third instar follows a regular sequence over a 2 day period. Parturition follows a circadian pattern with a peak 9 hr after lights on (12 hr daily photophase). All instars receive nutriment from the female's milk gland. During early pregnancy the rate of milk synthesis is greater than rate of uptake by the larva, thus causing expansion of the secretory reservoirs. After day 6, the volume of the secretory reservoirs decreases, but as is indicated by nuclear volume and larval growth the rate of synthesis remains high until day 8. Feeding activity of the adult female is maximal on day 1, levels off at 60 per cent up to day 6, and then declines sharply towards the end of pregnancy. Oöcyte development proceeds in phase with larval development and thus minimizes a lag period between successive pregnancies.  相似文献

The Asian tiger mosquito, Aedes albopictus, is a medically important invasive species whose geographic distribution has expanded dramatically during the past 20 years, and one of the key elements of its success is its capacity to survive long distance transport as a diapausing pharate first instar larva, encased within the chorion of the egg. We report that pharate larvae entering diapause are larger and contain 30% more lipid than their nondiapausing counterparts. To improve our understanding of the molecular regulation of lipid metabolism during diapause, we assessed the relative mRNA abundance of 21 genes using qRT-PCR. Elevated expression of lipid storage droplet protein 2 during embryonic development likely contributes to the higher amounts of lipid we noted in diapausing individuals. The conservation of lipids during diapause is reflected in downregulation of genes involved in lipid catabolism, including lipase 2, lipase 3, lipase 4, acyl-CoA dehydrogenase 4, and isovaleryl-CoA dehydrogenase. Two genes involved in fatty acid synthesis and modification, Δ(9)-desaturase, and fatty acyl-CoA elongase, were both upregulated in diapausing pharate larvae, suggesting roles for their gene products in generating unsaturated fatty acids to enhance membrane fluidity at low temperatures and generating precursors to the surface hydrocarbons needed to resist desiccation, respectively. Together, the results point to substantial distinctions in lipid metabolism within the embryo as a consequence of the diapause program, and these differences occur both before the actual onset of diapause as well as during the diapause state.  相似文献
