In plants, the enzymes for cysteine synthesis serine acetyltransferase (SAT) and O-acetylserine-(thiol)-lyase (OASTL) are present in the cytosol, plastids and mitochondria. However, it is still not clearly resolved to what extent the different compartments are involved in cysteine biosynthesis and how compartmentation influences the regulation of this biosynthetic pathway. To address these questions, we analysed Arabidopsis thaliana T-DNA insertion mutants for cytosolic and plastidic SAT isoforms. In addition, the subcellular distribution of enzyme activities and metabolite concentrations implicated in cysteine and glutathione biosynthesis were revealed by non-aqueous fractionation (NAF). We demonstrate that cytosolic SERAT1.1 and plastidic SERAT2.1 do not contribute to cysteine biosynthesis to a major extent, but may function to overcome transport limitations of O-acetylserine (OAS) from mitochondria. Substantiated by predominantly cytosolic cysteine pools, considerable amounts of sulphide and presence of OAS in the cytosol, our results suggest that the cytosol is the principal site for cysteine biosynthesis. Subcellular metabolite analysis further indicated efficient transport of cysteine, γ -glutamylcysteine and glutathione between the compartments. With respect to regulation of cysteine biosynthesis, estimation of subcellular OAS and sulphide concentrations established that OAS is limiting for cysteine biosynthesis and that SAT is mainly present bound in the cysteine–synthase complex.  相似文献

The trace fossil Zoophycos is abundant in transgressive, shallow marine carbonates in the Middle Permian (Wordian) Khuff Formation of the Huqf-Haushi Uplift of Interior Oman. It often occurs as part of a complex (compound) trace fossil that comprises two integrated elements: (i) irregular galleries with straight to gently curved tunnels and interconnected shafts, and (ii) simple planar to complex spreiten structures with a marginal tube ( Zoophycos ). The galleries are characterized by irregularly winding, dichotomous branching, large variation in shape and size and circular to elliptical vertical cross-sections. Zoophycos consists of spreiten with a marginal tube, either originating as a simple lobe from the convex segment of a curved tunnel, or forming more complex, subcircular, spreiten systems parallel to bedding. The spreiten were formed by simple strip mining, where the animal defecated without producing faecal pellets. U-shaped marginal tubes indicate that the burrows were well aerated. The complex trace fossil points to combined dwelling and deposit-feeding behaviour, with irregular galleries in the firm substrate and Zoophycos spreiten in the softground below it. It can be assumed that the animal used the open tunnel system mainly for dwelling (domichnion) and possibly suspension feeding, but occasionally changed to deposit feeding while creating the spreiten (fodinichnion). The integration of the irregular galleries (tunnels and interconnected shafts) with the marginal tubes of Zoophycos suggests the same producer for this compound trace fossil. Many modern polychaetes produce very similar galleries within firm and soft substrates, and polychaetes are therefore interpreted as the most likely producers. Similarities between Permian and Triassic Zoophycos suggest comparable trace making behaviour before and after the end-Permian mass extinction.  相似文献

Abstract.  1. An organism's growth parameters are expected to depend on environmental constraints, such as predation risk and food supply. However, antipredator responses, food intake, and thus growth of an animal may be mediated by behavioural traits, which are likely to differ among developmental stages. In this study, it was investigated how the relationship between growth and behavioural antipredator responses changes during ontogeny in the time-constrained dragonfly species Libellula depressa , and which factors influenced specific behavioural decisions at different points in ontogeny.
2. The results revealed that behavioural strategies differed between larval developmental sages, depending on associations between larval growth, food supply, and predation risk. Early in ontogeny, faster development was correlated with high larval activity and high food supply. This resulted in high activity levels under high food conditions irrespectively of predator presence, and under low food supply in predator absence only. In the intermediate stage of development, all larvae displayed a high activity level, which was correlated in general with fast development. However, growth later in ontogeny was not only influenced by the activity level, but also by predator presence and food supply, with larvae reared under high food supply and/or in presence of predators attaining a higher final mass. Thus, not only the way in which larval growth parameters and behaviour are related changed during development, but also whether the factors influenced larval growth and behaviour. Once the larvae reached the ultimate stage of development, in which they overwinter, behavioural patterns observed were consistent with model predictions.
3. It is advocated that behavioural plasticity of prey organisms in different developmental stages should be analysed in the context of associated growth variables.  相似文献

The floodplains of the West‐African Sahel region have experienced extensive habitat transformation during the past four decades, coinciding with an impoverishment of raptor populations. We investigated foraging patterns of Palaearctic migratory Eurasian Marsh Harriers Circus aeruginosus, Pallid Harriers C. macrourus and Montagu’s Harriers C. pygargus on a floodplain system in northern Cameroon to assess species, sex‐ and age‐related habitat preferences. Sex and age have rarely been incorporated into general studies of raptor habitat associations, despite clear evidence of intrasexual and age‐related differences in foraging strategies and diet composition, potentially carrying strong conservation implications. We found evidence of sexual differences in foraging preference related to land use, particularly in the most sexually dimorphic Pallid Harrier, and evidence that juveniles used different habitats to adults. This constitutes the first quantitative documentation of such differentiation by Palaearctic raptors on African wintering grounds, indicating that general patterns of habitat use in wintering raptors may obscure sex‐ and age‐specific preferences. Contrary to expectations, we found limited evidence for interspecific foraging segregation. Food partitioning by prey mass was related to harrier body mass and facilitated by a diverse availability of prey on human‐transformed floodplains. Anticipated further large‐scale conversion of floodplain habitat into predominantly desiccated grasslands raises concerns about the survival of wintering harriers.  相似文献

This paper describes the strategies of resource utilization in the course of the breeding season by five radio-tagged Grey Herons Ardea cinerea. The seasonal changes in exploitation of the environment by two breeding adults, one non-breeding adult and two non-breeding first-year birds were studied from March to August 1982, near Zonhoven in Belgium. Two adult breeding birds could be followed continuously from the end of March until the middle of June. During the first month they explored an extended area all around the colony, but each concentrated its search in a specific direction. From the end of April until the beginning of June, most probably from egg-hatching until the end of breeding activities, each bird spent a very large proportion of its time at a particular feeding site, from which other herons were actively excluded. In the first part of June they again visited different sites, each maintaining its preferred direction. From the middle of June onwards they seemed to have left the fish-pond area. The pattern of movements of the first-year birds differed markedly from that of the breeding adults. In April, although both non-breeding and breeding birds explored large areas, only the areas used by non-breeders were centred on the colony. From the end of April onwards, probably after general egg-hatching in the colony, the non-breeders very rarely revisited the colony, and from May till August their ranges became more and more restricted to very small areas at an increasing distance from the colony. They were never observed defending particular sites. The results are discussed with regard to recent speculations about the evolution of colonies as an adaptation for the exploitation of food resources. Breeding herons seem to explore a large part of the environment during incubation and defend a particular site while feeding their young. Choice of feeding site by non-breeding birds may be influenced by the site defence of the breeding birds. Non-breeding birds exploit a large area when breeding birds occupy feeding territories. Perhaps they are forced to forage in less suitable places at this time. Colonies might have evolved as a strategy to minimize effort in resource esploitation as, especially at the beginning of the breeding season, the colony could act as an assembly point in the exploration of the environment. However, its importance as an assembly point diminishes in the course of the season, as non-breeding birds no longer visit the colony and adults defend territories.  相似文献

In vivo, melanocytes were detected in epidermis from human tissue of 6.5 weeks estimated gestinational age (EGA) and older. We have successfully established melanocyte monocultures from tissue of 9 to 10 weeks EGA. To our knowledge, this is the first report on physiology of human foetal melanocytes in monoculture. In culture, such melanocytes retained foetal characteristics. Proliferation rates noted were markedly higher (approximately 2.7-fold) when compared to those in cultures of neonatal melanocytes. Moreover, when analyzing cellular phenotypes by markers for cells of the melanocytic lineage, foetal cells isolated from tissue of 9 weeks EGA reproducibly showed expression of the high molecular weight (HMW) antigen and c-kit to an extent intermediate to that found in neonatal melanocytes and M14 melanoma cells. Such differential expression was not observed if cells were isolated from tissue of 10 weeks EGA, indicating that the foetal environment provides essential differentiation stimuli during the 10th week of gestation. Moreover, these results are supportive of the theory that malignant transformation involves a process of dedifferentiation. In all, human foetal melanocyte culture provides a useful model to investigate pigment cell differentiation.  相似文献

Intact rich fen soil cores with controlled water levels near thesurface were installed in an open greenhouse. To simulate short termsummer drought, water levels were lowered (20 cm) after two weeks inhalf of the cores (experimental cores) and remained near the surface inthe other half (blanks). After two more weeks, the water levels werebrought back to the surface in the experimental cores and remainedthere for another two weeks. In the blanks, reduction and alkalinizationof the top peat layer occurred. In the experimental soil cores oxidationand acidification started within one week after drawdown. An indicationfor a drought induced rise in soluble reactive phosphorus has beenfound. The velocity of the acidification process illustrates the dynamicnature of the hydrochemical conditions in fen soils during drought. Theprocesses controlling the acid/base status of rich fen, the effect ofdrought induced acidification on P availability and the significance forthe vegetation are discussed.  相似文献
