Plants of Lupinus albus were grown for 51 d under control (1.1mol m–3 NaCl) and saline (40 mol m–3 NaCl) conditions.Plants were harvested and changes of carbon, nitrogen and abscisicacid (ABA) contents of individual organs were determined 41d and 51 d after germination. In the period between the twoharvests xylem and phloem saps were collected and respirationand photosynthesis of individual organs were measured. Usingflows of carbon, C/ABA ratios and increments of ABA flows ofABA in phloem and xylem and rates of biosynthesis and degradationof ABA were calculated. Both under control and saline conditionsnet biosynthesis occurred in the root, the basal strata of leavesand in the inflorescence. Metabolic degradation of ABA tookplace in the stem internodes and apical leaf strata. Salt stress increased xylem transport of ABA up to 10-fold andphloem transport to the root up to 5-fold relative to that ofthe controls. A considerable amount of ABA in the xylem saporiginated from biosynthesis in the roots, i.e. 55% in salt-treatedand smaller than 28% in control plants. The remaining part ofABA in the xylem sap originated from the shoot: it was translocatedin the phloem from fully differentiated leaves towards the rootand from there it was recirculated back to the aerial partsof the plant. Hordeum vulgare cv. California Mariout was established in sandculture at two different NaCl concentrations (0.5 mol m–3‘control’ and 100 mol m–3) in the presenceof 6.5 mol m–3 K +. Between 16 and 31 d after germination,before stem elongation started, xylem sap was collected by useof a pressure chamber. Collections were made at three differentsites on leaves 1 and 3: at the base of the sheath, at the baseof the blade, i.e. above the ligule, and at the tip of the blade.Phloem sap was collected from leaf 3 at similar sites throughaphid stylets. The concentrations of K +, Na+, Mg2+ and Ca2+were measured. Ion concentrations in xylem sap collected at the base of leaves1 and 3 were identical, indicating there was no preferentialdelivery of specific ions to older leaves. All ion concentrationsin the xylem decreased from the base of the leaf towards thetip; these gradients were remarkably steep for young leaves,indicating high rates of ion uptake from the xylem. The gradientsdecreased with leaf age, but did not disappear completely. In phloem sap, concentrations of K+ and total osmolality declinedslightly from the tip to the base of leaves of both controland salt-treated plants. By contrast, Na+ concentrations inphloem sap collected from salt-treated plants decreased drasticallyfrom 21 mol m–3 at the tip to 7.5 mol m–3 at thebase. Data of K/Na ratios in xylem and phloem sap were used to constructan empirical model of Na+ and K+ flows within xylem and phloemduring the life cycle of a leaf, indicating recirculation ofNa+ within the leaf. In recent years, NO, a gas previously considered a potentially toxic chemical, has become established as a diffusible universal messenger mediating cell—cell communication throughout the body. In mammals, NO is a recognized mediator of blood vessel relaxation that helps to maintain blood pressure. In the central nervous system NO acts as a non-conventional neurotransmitter and participates in the establishment of long-term plasticity required for memory formation. In addition, NO is responsible for some parts of the host response to sepsis and inflammation and contributes to certain disease states. A number of strategies have emerged with regard to a pharmacological control of pathological NO overproductions. This review will discuss these novel therapeutic approaches that may provide new means for clinical medicine.  相似文献

Based on field measurements in two agriculturalecosystems, soil respiration and long-term response ofsoil organic carbon content (SOC) was modelled. Themodel predicts the influence of temperature increaseas well as the effects of land-use over a period ofthirty years in a northern German glacial morainelandscape. One of the fields carried a maizemonoculture treated with cattle slurry in addition tomineral fertilizer (maize monoculture), the otherwas managed by crop rotation and recieved organicmanure (crop rotation). The soils of both fieldswere classified as cambic Arenosols. The soilrespiration was measured in the fields by means of theopen dynamic inverted-box method and an infrared gasanalyser. The mean annual soil respiration rates were 268 (maizemonoculture) and 287 mg CO2 m-2 h-1(crop rotation). Factors controlling soil respirationwere soil temperature, soil moisture, root respirationand carbon input into the soil. Q10-valuesof soil respiration were generally higher in winterthan in summer. This trend is interpreted as anadaptive response of the soil microbial communities.In the model a novel mathematical approach withvariable Q10-values as a result oftemperature and moisture adjustment is proposed. Withthe calibrated model soil respiration and SOC werecalculated for both fields and simulations over aperiod of thirty years were established. Simulationswere based on (1) local climatic data, 1961 until1990, and (2) a regional climate scenario for northernGermany with an average temperature increase of 2.1 K.Over the thirty years period with present climateconditions, the SOC pool under crop rotation wasnearly stable due to the higher carbon inputs, whereasabout 16 t C ha-1 were lost under maizemonoculture. Under global warming the mean annualsoil respiration for both fields increased and SOCdecreased by ca. 10 t C ha-1 under croprotation and by more than 20 t C ha-1 undermaize monoculture. It was shown that overestimationof carbon losses in long-term prognoses can be avoidedby including a Q10-adjustment in soilrespiration models.  相似文献

A new group of darkly pigmented root-infecting fungi was isolated from cereal roots obtained from six different locations in northeastern Germany. Similar random amplified polymorphic DNA(RAPD) patterns and restriction profiles of amplified rDNA were used as a basis for classifying the isolates in a separate group. The isolates demonstrating mycelial and infection characteristics typical of Gaeumannomyces graminis could be differentiated from the varieties of G. graminis as well as from Gaeumannomyces cylindrosporus / Phialophora graminicola using RAPD Polymerase chain reaction (PCR) and rDNA Restriction-fragment length polymorphism (RFLP) analysis. Phylogenetic analysis of the Internal transcribed spacer (ITS) regions suggests that the isolates form a distinct group (named group 'E') situated within the Gaeumannomyces – Phialophora complex between the branch of the G. graminis varieties and Gaeumannomyces incrustans / Magnaporthe poae . Isolates of group E produced lobed hyphopodia and were shown in biotests to be non-pathogenic to wheat, oats, Italian Ryegrass and Chewings Fescue, suggesting it is a benign parasite which colonizes cereals or grasses without destroying vascular tissue. Furthermore, curved phialospores could be found. Summarizing the results presented, this new group could be classified as a new species of Phialophora . Although isolates of group E were found at only six of the 32 investigated locations, they composed up to 50% of total isolates of the Gaeumannomyces – Phialophora complex at these sites. Because of the non-pathogenic behaviour, the new group may be of value as biological control agents for pathogenic fungi.  相似文献

The growth of garden orache, A triplex hortensis was studiedunder conditions of mild NaCl or Na2SO4 salinity. Growth, drymatter production and leaf size were substantially stimulatedat 10 mM and 50 mM Na+ salts. Increased growth, however, appearedto be due to a K+-sparing effect of Na+ rather than to salinityper se. The distribution of K+ and Na+ in the plant revealeda remarkable preference for K+ in the roots and the hypocotyl.In the shoot the K/Na ratio decreased strongly with leaf age.However, the inverse changes in K+ and Na+ content with leafage were dependent on the presence of bladder hairs, which removedalmost all of the Na+ from the young leaf lamina. Measurementsof net fluxes of K+ and Na+ into roots and shoots of growingAtriplex plants showed a higher K/Na selectivity of the netion flux to the root compared to the shoot. With increasingsalinity the selectivity ratio SK, Na* of net ion fluxes tothe roots and to the shoots was increased. The data suggestthat recirculation of K+ from leaves to roots is an importantlink in establishing the K/Na selectivity in A. hortensis plants.The importance of K+ recirculation and phloem transport forsalt tolerance is discussed. A comparison has been made of the kinetics of 22Na+ uptake intoexcised barley roots and roots pre-loaded with glycinebetaine.The elevated intracellular glycinebetaine or a metabolic consequencethereof increased the Na+ influx, and the effect was relatedto the level of internal glycinebetaine and or Na+ [Cl].The quasi-steady-state Na+ influx at the tonoplast rather thanthe plasmalemma influx was apparently influenced by glycinebetaineloading. The tonoplast fluxes and vacuolar Na+ content wereconsistently higher in glycinebetaine-loaded roots than unloadedroots. PHYTOHAEMAGGLUTININ (PHA) has many different biological activities, for example, erythroagglutination1, leucoagglutination1, stimulation of RNA and DNA synthesis1 and induction of interferon1 and lymphotoxin (LT)2.
