. Two strains of the Leishmania braziliensis complex have been adapted to grow extracellularly at elevated temperature as amastigote-like forms in a cell-free medium. These parasites can be serially cultivated and maintained at 32°C for L. panamensis (WR442; L. braziliensis panamensis ) and at 28°C for L. braziliensis (M5052; L. braziliensis braziliensis ). Several observations are presented that the forms adapted at elevated temperature are amastigote-like. Morphologically, the amastigote-like organisms appear rounded to ovoid and are immotile and smaller than promastigotes; the flagellum of the amastigote-like forms does not extend beyond the flagellar pocket. In comparison, the promastigotes are very elongated, with a nucleus at mid-cell length and a very long flagellum. By electron microscopy, the short flagellum of the amastigote-like form is within a distended flagellar pocket; the 9 + 2 axonemal configuration is present but the paraxial rod is not observed. By contrast, the flagellum of the promastigote has a paraxial rod which extends from the axosome level. In addition, these amastigote-like forms of Leishmania are able to infect, to survive and to divide within the macrophage cell line J774.  相似文献

ABSTRACT. The biochemistry and immunology of Leishmania promastigotes has been extensively studied; this is due primarily to the facility with which this stage, in contrast to the amastigotes stage, can be maintained in axenic culture. Several attempts to axenically culture lines of Leishmania amastigotes have been reported in the literature. This paper summarizes methods of adaptation (low pH, elevated temperature and culture medium) and characterization of several axenic lines of Leishmania amastigotes. Based on morphological, biological, immunological and biochemical evidence, these organisms appear to resemble amastigotes from infected macrophages or tissue. The axenically cultured amastigotes appear to be distinct from shocked (heat, serum deprivation, stressed) Leishmania promastigotes in the plethora of proteins synthesized, growth (multiplication) in culture, and developmental regulation observed. These data suggest that Leishmania organisms have a significant developmental response to certain signals (pH, temperature) mimicking their in vivo macrophage milieu. The response to other environmental parameters characteristic of the host-macrophage remain to be determined. These axenically cultured amastigotes should be of interest for further immunological, biochemical and developmental investigations of the disease-maintaining stage of this parasite.  相似文献

Community studies as a form of study in complex societies cart be unduly restrictive though this need not be the case. It depends on the criteria employed to select a field location, and on the types of social organization studied within that location. These themes are illustrated by reference to the Australian community studies of Ron Wild and Harry Oxley. Some limitations in their studies are noted only in order to illustrate how their work might have been linked profitably with debates in Australian historiography over issues of class culture, ideology and consciousness. I propose a framework for Australian community studies that would allow anthropologists to explore some of the themes introduced in this discussion.  相似文献

The botanical product RD-Repelin® was highly repellent to pea aphids at concentrations of 1%, 4% or 10%. Repellency occurred prior to leaf contact by aphids at all three concentrations. RD-Repelin® delayed symptom expression of zucchini yellow mosaic virus (ZYMV) in 81% of plants treated with 1% concentration, although virus transmission was not prevented. The effects of RD-Repelin® on aphid settling behaviour, symptom expression and potential for pest management are discussed.  相似文献

Abstract The impact of introducing animals into an established ecosystem can be directly observed through predator-prey and competition interactions. The impact of animals via more obscure relationships, such as the host-parasite relationship, are generally not considered. The cane toad Bufo marinus (Linnaeus) was introduced to Australia in 1935. Despite intensive research into many aspects of the biology of the toad, there has been no systematic survey of the parasite fauna of B. marinus in Australia. It is unknown exactly what parasites the toad may have introduced to Australia and also the range of parasites that may have adapted to the toad from native fauna since its introduction. The provisional conclusion from this review is that all the helminth parasites so far recorded from B. marmus in Australia have been acquired from local hosts. The interaction of toads and nativa fauna via their parasites remains unknown.  相似文献

1. The lycaenid butterfly Hemiargus isola associates facultatively with the ant species Formica perpilosa in arid areas of south-western North America. Ants solicit liquid food rewards from butterfly larvae as larvae feed on the host plant, Acacia constricta . Previous studies have shown that tending by F. perpilosa enhances larval growth and pupal survivorship.
2. The effects of ants and plant water content on oviposition behaviour and survivorship to the last larval instar were tested by excluding ants and supplementing water to host plants in a two-way factorial experiment.
3. Butterflies, which lay eggs singly on host plant inflorescences, laid significantly higher egg numbers and densities (eggs/inflorescence) on plants with ants than on plants without ants. This is the first report of a facultative, generalized ant-associate using ants as oviposition cues. Water supplements increased the number, but not the density, of eggs laid on plants. Therefore, it appears that egg-laying butterflies responded to number of inflorescences, rather than plant tissue water per se .
4. Plants with ants had significantly greater numbers of inflorescences during the experiment than plants without ants. Water supplements increased number of inflorescences slightly, but not significantly.
5. Ants increased larval survivorship. Twice as many fourth-instar larvae survived per egg laid on plants with ants than on plants without ants. Ants did not reduce the number of predators present on acacias, but may have reduced predator effectiveness. Ants also did not reduce the numbers of potential H. isola competitors present.
6. Water supplementation affected neither the survivorship of H. isola larvae, nor the intensity of ant tending. Water supplementation did not affect the abundance of predators on plants, but did increase the abundance of several herbivorous insect taxa.  相似文献
