In Vivo Nitrogenase Regulation by Ammonium and Methylamine and the Effect of MSX on Ammonium Transport in Anabaena flos-aquae 总被引:2,自引:2,他引:0
Ammonium suppresses nitrogenase activity in Anabaena flos-aquae (Lyng) Breb. at all pH values tested. l-Methionine-dl-sulfoximine at 1 millimolar totally inhibited glutamine synthetase, and 10 micromolar partially inhibited. Both concentrations protected nitrogenase activity from ammonium-induced suppression at pH 7.1 and 8.1. At pH 9.3 and 10.2, methionine sulfoximine did not alleviate the suppression of nitrogenase by ammonium. This pH-dependent protection of nitrogenase activity is a result of the noncompetitive inhibition of the ammonium transporter by methionine sulfoximine. At pH 7.1 and 8.2, ammonium is protonated and methionine sulfoximine inhibits its entry into the cell. At pH 9.3 and 10.2, unprotonated ammonia is abundant and may enter the cell independent of the transport system. The effects of ammonium are closely mimicked by the ammonium analog methylamine. These results suggest that ammonium per se is an important in vivo regulator of nitrogen fixation and its function can be mimicked by methylamine. Previous studies employing methionine sulfoximine may have to be re-evaluated in light of the inhibitory effects of methionine sulfoximine on the ammonium transporter. 相似文献

Genetic analyses of 49 duplications of the rII region of bacteriophage T4D suggests that there is a non-random relationship between the end points of duplicated segments, that relaxed packaging restrictions have little if any effect on the distribution of duplications, that segregation is 3–4 times more frequent than normal recombination for the same interval, and that non-tandem duplications are rare. Extrapolation of the r1231 x rJ101 cross data suggests that the minimum frequency of duplications/genome is 1.7 x 10-6, but possibly 3.4 x 10-4. 相似文献

Oestrus was synchronized in 46 Afrikaner and Mashona beef cows by two injections of cloprostenol 11 days apart. All cows had a history of sub-optimum reproductive performance. Cows were inseminated up to three times after the second cloprostenol injection on the basis of observed oestrus and changes in the conductivity of cervical mucus. Half the cows received daily injections of 1000 i.u. HcG from days four to 19 after their last insemination; the other half received daily injections of 2 ml saline over the same period. Concentrations of progesterone in plasma was determined from samples taken on days 6, 8 and 22 after the last insemination. Treatment did not significantly affect conception rate and overall conception rate was 39 per cent. On day 8 after insemination none of the 8 cows that had progesterone levels of less than 1 ng/ml were pregnant when examined at day 70. Mean progesterone concentrations were not significantly different between treated and control cows on days 6 and 8, but were significantly higher (P<0,05) in treated cows by day 22. The practical significance of using HcG to stimulate luteal function in the early post-inseminaion period is discussed. 相似文献

Seven congenic strains differing from C57BL/10Sn at theH-13 locus have been produced which define fourH-13 alleles. Isografting, exchanging of grafts between sublines, F(1) testing, and linkage testing demonstrate the presence of additionalH genes in four of these strains. The medial survival times (MSTs) of skin grafts fromH-13(a) to unimmunizedH-13(b) recipients ranged from 69 to 83 days. Rejection across all other barriers was extremely weak with most MSTs being > 100 days. Preinjection of donor strain thymocytes caused accelerated rejection of skin grafts fromH-13(a) toH-13(b) mice, but had only minimal effect on skin grafts across other barriers. Phytochrome A (phyA) mRNA abundance decreased rapidly in total RNA samples isolated from 4-day-old etiolated oat seedlings following a red light pulse. Putative in vivo phyA mRNA degradation products were detectable both before and after red light treatment. Cordycepin-treated coleoptiles were unable to accumulate the chlorophyll a/b-binding protein mRNA in response to red light, indicating that cordycepin effectively inhibited mRNA synthesis. In cordycepin-treated coleoptiles, phyA mRNA rapidly decreased in abundance, consistent with the hypothesis that phyA mRNA is inherently unstable, rather than being destabilized after red light treatment of etiolated oat seedlings. 相似文献

Supplying l-proline to the root system of intact soybean (Glycine max [L.] Merr.) plants stimulated acetylene reducing activity to the same extent as did supplying succinate. Feeding l-proline also caused an increase in bacteroid proline dehydrogenase activity that was highly correlated with the increase in acetylene-reducing activity. Twenty-four hours after irrigating with l-proline, endogenous proline content had increased in host cell cytoplasm and bacteroids, about three- and eightfold, respectively. In bacteroids, proline concentration was calculated to be at least 3.5 millimolar. In experiments in which [U-14C]l-proline was supplied to uprooted, intact plants incubated in aerated solution, 14C-labeled products of proline metabolism, as well as [14C]proline itself, accumulated in both host cells and bacteroids. When plants were incubated in aerated solutions containing [5-3H]l-proline, 3H-labeled proline was found in host cells and bacteroids. [3H] Pyrroline-5-carboxylate was found in bacteroids, but not host cells, after a 2-hour incubation in [5-3H]l-proline. When [U-14C]l-proline was supplied for 24 hours, a significant amount of [14C] pyrroline-5-carboxylate was found in the host cells, in contrast with the results from the shorter incubation in [5-3H]proline, although the amount in the host cells was only about half the quantity found in the bacteroids. Taken as a whole, these results indicate that proline crosses both plant and bacterial membranes under the in vivo experimental conditions utilized and are consistent with a significant role for proline as an energy source in support of bacteroid functioning. In spite of the increase in acetylene-reducing activity when proline was supplied to the root system of intact plants, proline application did not rescue stemgirdled plants from loss of acetylene-reducing activity, although succinate application did. Addition of NO3− to N-limited Selenastrum minutum during photosynthesis resulted in an immediate drop in the NADPH/NADP ratio and a slower increase of the NADH/NAD ratio. These changes were accompanied by a rapid decrease in glucose-6-phosphate and increase in 6-phosphogluconate, indicating activation of glucose-6-phosphate dehydrogenase and a role for the oxidation pentose phosphate pathway during photosynthetic NO3− assimilation. In contrast, the short-term changes in pyridine nucleotides and metabolites during photosynthetic assimilation of NH4+ were not consistent with a stimulation of the oxidative pentose phosphate pathway. 相似文献

Near-infrared interactance spectra of the vulva and vestibule of cows were analyzed throughout the estrous cycle to determine if they could be used to detect tissue changes that are associated with estrus. The spectra were collected daily from 16 cyclic Holstein cows beginning at 40 days post partum and continuing for 3 estrous cycles, or until pregnancy was verified. Spectra collected on the day when estrus was exhibited were compared with spectra collected on the nonestrous days. There were spectral differences at 1695 to 1705, at 1790 to 1800 and at 1880 to 1900 nm (P<0.05), regions that are associated with changes in carbohydrate, protein, and water content of the tissues, respectively. Discriminant analysis of interactance measurements at the significant wavelengths was used to develop an equation to predict the day of estrus. The error rate associated with the equation was 26%. Results indicate that near-infrared analysis can detect changes in vestibular and vulvar tissue during the estrous cycle. However, accurate estrus detection using near-infrared interactance techniques will require additional study to reduce variation in the results. 相似文献
