A density-based load estimation method was applied to determine femoral load patterns. Two-dimensional finite element models were constructed using single energy quantitative computed tomography (QCT) data from two femora. Basic load cases included parabolic pressure joint loads and constant tractions on the greater trochanter. An optimization procedure adjusted magnitudes of the basic load cases, such that the applied mechanical stimulus approached the ideal stimulus throughout each model. Dominant estimated load directions were generally consistent with published experimental data for gait. Other estimated loads suggested that loads at extreme joint orientations may be important to maintenance of bone structure. Remodeling simulations with the estimated loads produced density distributions qualitatively similar to the QCT data sets. Average nodal density errors between QCT data and predictions were 0.24 g/cm(3) and 0.28 g/cm(3). The results indicate that density-based load estimation could improve understanding of loading patterns on bones. 相似文献

ABSTRACT. A soluble enzyme amylopectin synthase (UDP-glucose-α 1,4-glucan α-4-glucosyltransferase) which transfers glucose from uridine 5'-diphosphate glucose (UDP-glucose) to a primer to form α-I,4-glucosyl linkages has been identified in the extracts of unsporulated oocysts of Eimeria tenella . UDP-glucose and not ADP-glucose was the most active glucosyl donor. Corn amylopectin, rabbit liver glycogen, oyster glycogen and corn starch served as primers; the latter two were less efficient. The enzyme has an apparent pH optimum of 7.5 and exhibited typical Michaelis-Menten kinetics with dependence on both the primer and substrate concentrations. The Michaelis constants (Km). with respect to UDP-glucose, was 0.5 mM; and 0.25 mg/ml and 1.25 mg/ml with respect to amylopectin and rabbit liver glycogen. The product formed by the reaction was predominantly a glucan containing α-1,4 linkages. The specificity of the enzyme suggests that this enzyme is similar to glycogen synthase in eukaryotes and has been designated as amylopectin synthase (UDP-glucose-α-1,4-glucosetransferase EC 2.4.1.11). 相似文献

ABSTRACT. The clonal life history of ciliated protists is characterized by a sequence of phenotypes; sexual immaturity, maturity, and senescence. The distinctiveness of immaturity and maturity has been investigated. Standard assays of the onset of maturity of progeny clones from a cross between stocks EC1 and EC2 of Euplotes crassus demonstrated significant differences among clones and among testers within clones. They also revealed that the first positive test(s) of a progeny subclone were typically followed by at least one negative test. Special protocols were devised to investigate if maturity was reversible at the cellular level. In these experiments, the first mating pair of a progeny subclone was split before the consummation of mating. From these two cells as well as from control progeny and tester cells, subclones were established and every leftover cell was tested for maturity after each transfer. Both standard and split-pair progeny subclones had immature and slow- to-mate cells. The number of fissions before progeny exhibited sexual behavior indistinguishable from the testers was more than twice that to the first mating reaction of a subclone. At the first sign of maturity, progeny lines are a heterogeneous population of cells able and not able to mate, but remarkably, clonal descendants of those able to mate may become unable to mate. The development of maturity is progressive, quantitative and non-monotonic rather than an instantaneous switch. 相似文献

Although plant invasions are often associated with disturbance, localized disturbances can promote invasion either by: (i) creating sites where individuals establish; or (ii) enabling an invader to colonize the entire stand. The former is expected when both establishment and survival to reproductive age require disturbed conditions, whereas the latter should occur in systems when either establishment or survival are limited to disturbed sites. We investigated the role of localized disturbance, specifically treefalls, in the invasion of the Asian Rubus phoenicolasius in a deciduous forest in Maryland, USA. We investigated the density and demography of R. phoenicolasius in treefall gaps of various sizes, but identical age to non‐gap areas, using Classification and Regression Tree (CART) analyses to identify the most important predictors. To explore how the demography of established individuals responds to disturbed versus undisturbed conditions, we carried out a garden experiment with three different levels of shade (5, 12 and 22% full sun). We found vegetative and sexual reproduction, and seedling establishment, to be limited to large gaps in an old stand, but not in a stand in an earlier age of succession. However, in the garden experiment, established plants were able to survive and grow under all shade treatments. These findings indicate that R. phoenicolasius requires disturbances such as treefalls to establish in forests, but established plants will survive canopy closure, leading to stand‐wide invasion. Managers should be able to prevent invasion, however, by inspecting large gaps for new recruits every 3 years. 相似文献

Resistance to bialaphos, a non-selective herbicide, was intro duced into cotton through genetic engineering. A gene encoding phosphinothric in acetyltransferase (bar) from Streptomyces hygroscopicus was inserted into elite varieties of cotton through particle bombardment. Based on the marker gene, -glucuronidase (gus) expression, a total of 18 Pima (Gossypium barbadense), 45 DP50 (G. hirsutum L.), 20 Coker 312 (G. hirsutum) and 2 El Dorado (G. hirsutum) transgenic plants were recovered. Integration of the bar gene into cotton genomic DNA was confirmed by Southern blot analysis and gene expression was confirmed by northern blot and enzyme assays. Herbicide (Basta®) tolerance up to 15 000 ppm was demonstrated in greenhouse trials. The newly introduced herbicide tolerance trait is inherited in a Mendelian fashion in the progenies of germline transformants. This study demonstrates the potential for particle bombardment to introduce commerically important genes directly into elite varieties of cotton. SEVERAL endocrine cell lines established in recent years show a functional response to hormones in vitro1 but, except for one mammary cell line2, none of them exhibits the normal hormone requirement for growth in vivo. We have now isolated a rat ovarian cell line whose growth in vitro is markedly stimulated by bovine luteinizing hormone (LH-NIH-B7), a pituitary gonadotrophin and by dexamethasone, a synthetic glucocorticoid. Orchid seeds are minute and the first seedling stages of terrestrial species are underground, so there is little knowledge about the biology of the early life history and the size of seed/seedling populations relative to the number of plants that eventually emerge above ground. A recently developed field sowing technique makes it possible to explore a variety of topics such as the length of time that seeds spend as part of the soil seed bank, the phenology of seed germination, substrate requirements for germination, dependence of germination on fungal availability, substrate requirements and growth rate of seedlings, seedling phenology and development, and seedling-fungus specificity patterns in the soil. Several of these applications are relevant for conservation issues. Studies of Goodyera pubescens, Corallorhiza odontorhiza and Liparis lilifolia show considerable variation in germination strategies, patterns of seedling development and establishment, substrate requirements, and levels of specificity with the fungi. 相似文献
