Nuclear or cell number, and the mitotic index, were recordedin endosperms of Triticum aestivum cv. Mardler to test if aparticular stage of endosperm development was critical in determiningthe final grain weight. The basal four florets of emasculatedspikelets (controls), and the third and fourth florets of spikeletswhere the two basal ovaries were removed (ovary-removed), weresampled at various times up to 360 h after hand-pollination.The coenocytic phase in endosperms ended about 84 h after pollinationregardless of both grain position and the treatment. The onsetof the cellular stage was characterized by the final large fluctuationsin the mitotic index reflecting the culmination of the synchronousnuclear division of the coenocytic stage. Thereafter, the mitoticindex fluctuated with smaller amplitudes and, by 216 h afterpollination, was < 1%. Neither floret position in the spikeletnor the treatment affected the pattern of alteration to themitotic index. However, ovary removal from first and secondflorets resulted in significantly heavier grains and higherendosperm cell number in the 3rd and 4th florets compared withthe controls. In all florets, mean endosperm cell number peakedat 280 h but decreased by 360 h after pollination. At this time,the mean cell numbers in endosperms of the 3rd and 4th floretsof ovary-removed spikelets were significantly higher than inthe corresponding endosperms in the controls. Thus, a key contributoryfactor in determining the final endosperm cell number may bethe number of cells which are lost during the late period ofthe cellular stage of endosperm development. Intact Myriophyllum spicatum plants were grown in compartmentalized containers in a growth room so that the roots were separated from the shoots by a watertight partition. Nitrogen 15N was added to the water or sediment to trace the uptake of inorganic N by the plant shoots or roots. Myriophyllum spicatum is capable of taking up inorganic N through both roots and shoots. Plant N requirements can apparently be met by root uptake alone. However, when about 0·1 mg/l of NH4-N were present in the water, foliar uptake supplied more N to the plants than did root uptake. Foliar uptake of NH4-N was found to be several times faster than that of NO3-N when both forms of N were present in the water. Only about 1% of the N taken up by the roots was subsequently released to the water through the foliage.  相似文献

SYNOPSIS. Algal symbiotes isolated from marine invertebrate hosts, and grown axenically in culture, have been successfully combined in artificial associations with chick fibroblasts and grunt (Haemulon sp.) fin explants. Growth (measured by cell numbers and chlorophyll a) is stimulated in both symbiotes and host cells. Studies of nitrogen uptake and exchange suggest that these artificial associations are similar to other, natural symbioses involving invertebrate hosts, e.g. the hermatypic corals.  相似文献

SYNOPSIS. Sporozoites of Eimeria tenella were injected into the peritoneal cavity of normal chickens and chickens immunized against E. tenella. In some experiments normal scrum and serum from resistant chickens were injected prior to the injection of sporozoites. After 15 or 30 minute periods of intraperitoneal incubation, exudates were harvested and the occurrence of intracellular sporozoites was determined. Only macrophages and degranulated granulocytes were observed to contain sporozoites. There was no significant difference between the number of macrophages obtained from normal chickens (normal macrophages) which contained sporozoites and the number of macrophages obtained from immune chickens (immune macrophages) which contained sporozoites. Significantly fewer immune macrophages treated with immune serum contained sporozoites than untreated normal or immune cells, normal macrophages treated with either serum, or immune macrophages treated with normal scrum. Sporozoites in untreated normal macrophages did not appear to be harmed by the intracellular environment, based on structural observations. The majority of sporozoites in macrophages from all other groups were difficult to distinguish within the cytoplasm and were visibly distorted. It is hypothesized that the presence of fewer infected macrophages in exudates of immune chickens and serum-treated normal chickens was caused by an enhanced ability of these cells to destroy the parasite. Similar observations were noted in the case of sporozoites within degranulated granulocytes of experimental groups. The lack of understanding of the degranulation phenomenon makes it difficult to interpret these findings.  相似文献

Methylbutenol is a 5-carbon alcohol that is produced and emitted by several species of pine in western North America, and may have important impacts on the tropospheric chemistry of this region. In the present study the response of methylbutenol basal emission rate (measured at a constant light intensity of 1500 µmol m−2 s−1 and temperature of 30 °C) to the light and temperature conditions of the growth environment was examined, using field-grown plants shielded with shade cloth of various densities. Methylbutenol basal emission rates increased linearly with the temperature of the growth environment but did not respond to the shading of foliage during growth and development. Both photosynthesis and basal methylbutenol emission rate declined in older needles; however, these declines appear to result from parallel but independent processes and not from basal MBO emission rate directly tracking photosynthetic rates. Older needles did not occupy cooler microenvironments within the canopy; and thus differing thermal microenvironment could not explain the reduced MBO emission in older needles.  相似文献

Leaf domatia, specialized chambers in the vein axils on the underside of leaves of many plant species, have remained an enigma for over a century. In this study we show a strong association between foliar domatia and mites in 37 plant species in Australasia. Overall, mites accounted for 91% of the arthropods observed in domatia. Across all species, a median of 51% of domatia were occupied and 71% of leaves showed mite evidence in domatia. The level of mite association did not depend on domatia type (pit, pouch, pocket, or tuft) or provenance (Papua New Guinea, Queensland, Victoria, or New Zealand). Mite association with domatia commonly varied between plant species, between individuals within species, and between shoots within individuals. The leaf developmental stage probably explains much of the variation in association for many of these species. The presence of a variety of life history stages of mites within domatia indicates that these structures act as shelters for development and reproduction. Furthermore, in 12 of 13 plant species examined, domatia concentrate mites in particular locations on the leaf. Mite taxa that we classify as largely predaceous (e.g. phytoseiids, stigmaeids and tydeids) or fungivorous (e.g. acarids and oribatids) were most common in domatia and dominated the association in 21 of 24 plant species in which the relative abundance of herbivorous, fungivorous and predaceous groups was quantified. We evaluate hypotheses that explain the role of leaf domatia, including non-functional hypotheses (e.g. architectural constraints), physiological function (e.g. gas exchange and water uptake), bacterial symbiosis and antagonistic and mutualistic associations with mites. Our quantitative results confirm anecdotal accounts of mite association with leaf domatia and are most consistent with Lundströem's century-old hypothesis of plant-mite mutualism in which leaf domatia billet predaceous and fungivorous mites that prey on plant enemies. Leaf domatia are widespread among woody angiosperms and abundant in many temperate and tropical regions of Australasia. Mites, an ancient group of arthropods whose diversity and abundance parallels that of insects, are likely to be important selective agents on terrestrial plants. Our results (1) indicate that mite-domatia association represents a relationship of comparable scope to plant-ant associations mediated by specialized plant structures such as extrafloral nectaries, food bodies and specialized domatia; (2) suggest that sociality is not a necessary prerequisite for widespread and diverse mutualisms between arthropods and plants; and, (3) extend the diversity of organisms that produce specialized mite ‘houses’ from lizards, and wasps and bees to woody angiosperms.  相似文献

ABSTRACT. An intronless gene encoding a protein of 674 amino acid residues with a molecular mass of 73,403 Da showing homology to the cytoplasmic form of the 70 kDa heat shock proteins has been cloned and sequenced from the intestinal pathogen Cryptosporidium parvum . Monospecific polyclonal antibodies obtained to recombinant protein recognized a single band with an approximate molecular mass of 70 kDa on a Western blot of C. parvum proteins, as well as the 70 kDa heat shock protein from bovine brain. Southern blot analysis suggested the gene was single copy in the C. parvum genome. Eleven perfect repeats of the sequence GGMP were found in the predicted protein near the carboxyl terminus.  相似文献
