Electron microscopic examinations of Glugea hertwigi and Spraguea lophii spores indicated the presence of a single plasma membrane; however, this membrane remained in the spore during the discharge of the sporoplasm from the spore. Although discharged spores retained the old plasma membrane, the extruded sporoplasms acquired a new plasma membrane. In order to determine where the new plasma membrane came from, we used two fluorescent probes with membrane affinities. The markers were tested on unfired and discharged spores. The probe, N-phenyl-1-naphthylamine (NPN), labeled the polaroplast membrane in addition to the apolar groups in the posterior vacuoles of unfired spores. After spore discharge, NPN label disappeared from the spore ghosts except for a slight fluorescence on residual plasma membranes. Much of the NPN-labeled membrane reappeared after spore discharge on the outer envelope of discharged sporoplasms. The probe chlorotetracycline (CTC) labeled calcium-associated membranes of spore polaroplasts. During spore discharge, the CTC fluorescence shifted from the polaroplast organelle of unfired spores to the outer envelope of discharged sporoplasms. These results indicate that the polaroplast organelle may provide the new plasma membrane for discharged microsporidian sporoplasms.  相似文献

1. This paper introduces a new approach for setting streamflow-based river ecosystem management targets and this method is called the 'Range of Variability Approach' (RVA). The proposed approach derives from aquatic ecology theory concerning the critical role of hydrological variability, and associated characteristics of timing, frequency, duration, and rates of change, in sustaining aquatic ecosystems. The method is intended for application on rivers wherein the conservation of native aquatic biodiversity and protection of natural ecosystem functions are primary river management objectives.
2. The RVA uses as its starting point either measured or synthesized daily streamflow values from a period during which human perturbations to the hydrological regime were negligible. This streamflow record is then characterized using thirty-two different hydrological parameters, using methods defined in Richter et al . (1996). Using the RVA, a range of variation in each of the thirty-two parameters, e.g. the values at ± 1 standard deviation from the mean or the twenty-fifth to seventy-fifth percentile range, are selected as initial flow management targets.
3. The RVA targets are intended to guide the design of river management strategies (e.g. reservoir operations rules, catchment restoration) that will lead to attainment of these targets on an annual basis. The RVA will enable river managers to define and adopt readily interim management targets before conclusive, long-term ecosystem research results are available. The RVA targets and management strategies should be adaptively refined as suggested by research results and as needed to sustain native aquatic ecosystem biodiversity and integrity.  相似文献

1. Recent increases in phytoplankton biomass and the recurrence of cyanobacterial blooms in western Lake Erie, concomitant with a shift from a community dominated by zebra mussels (Dreissena polymorpha) to one dominated by quagga mussels (D. bugensis), led us to test for differences in ammonia‐nitrogen and phosphate‐phosphorus excretion rates of these two species of invasive molluscs. 2. We found significant differences in excretion rate both between size classes within a taxon and between taxa, with zebra mussels generally having greater nutrient excretion rates than quagga mussels. Combining measured excretion rates with measurements of mussel soft‐tissue dry weight and shell length, we developed nutrient excretion equations allowing estimation of nutrient excretion by dreissenids. 3. Comparing dreissenid ammonia and phosphate excretion with that of the crustacean zooplankton, we demonstrated that the mussels add to nitrogen and phosphorus remineralisation, shortening nitrogen and phosphorus turnover times, and, importantly, modify the nitrogen and phosphorus cycles in Lake Erie. The increased nutrient flux from dreissenids may facilitate phytoplankton growth and cyanobacterial blooms in well‐mixed and/or shallow areas of western Lake Erie.  相似文献

1. Through analyses of a 34‐year record of phytoplankton, zooplankton and physicochemical parameters from Lake Kinneret, Israel, we show that distinct and persistent phytoplankton assemblage states occurred from winter to summer. 2. The most obvious characteristic of these states was the presence or absence of a spring bloom of the dinoflagellate, Peridinium gatunense. 3. Analyses of the data within the framework of the alternative states model revealed a possible complex triggering mechanism, and system hysteresis. 4. A change in zooplankton biomass and body size coincident with changes in predation pressure associated with the collapse of the Kinneret Bleak, Acanthobrama terraesanctae, fishery appeared to be the ‘slow changing’ variable in the context of the alternative states model. Alternative phytoplankton states were only possible after this variable crossed a threshold in 1993–94, following the collapse of the fishery. 5. When alternative states were possible, some physicochemical parameters and the structure of the zooplankton assemblage appeared to control which phytoplankton state emerged in a given year. In years without a P. gatunense bloom, important physicochemical parameters in winter included low NO3 loading, high water temperature, high water level, a deeper thermocline, low transparency, high concentrations of NO3 and Cl in the epilimnion, and low concentration of epilimnetic total phosphorus. In addition, the cladoceran Chydorus sphaericus and adults of the copepod Mesocyclops ogunnus were observed in winter in years without a bloom. 6. Zooplankton biomass and body size of some taxa have recovered since the 1993–94 collapse of the fishery, yet incidence of both phytoplankton states in Lake Kinneret was still possible. Within the framework of the alternative states model, this suggests that the slow changing variable threshold where alternative states became possible is different from the threshold where alternative states will no longer be possible.   • 1 Greenhouse experiments tested the effect of pH 5 v pH 7.5 on reproductive success for the freshwater macrophytes Najas flexilis, an annual, and Vallisneria americana, which perennates as a tuber.
  • 2 Seed production by the small Najas plants grown at pH 5 averaged 0.25 seeds/plant, in contrast to 95.5 seeds/plant at pH 7.5. At low pH, Vallisneria grown from seeds produced no flowers and too few tubers to replace themselves, so that sexual reproduction failed nearly completely for both species.
  • 3 Vallisneria grown from tubers produced 97% less total tuber mass at pH 5 (0.4 v 14.9 g), the compounded result of producing, on average, 89% fewer and 82% smaller tubers. The smaller tubers developed at pH 5 were less likely to overwinter in the field, and those surviving tubers subsequently grew into smaller plants.
  • 4 These findings generated the hypothesis of a closing spiral: growth at low pH (and relatively low CO2 concentrations) results in small plants that produce a smaller number of progressively smaller tubers each autumn, which in turn develop into progressively smaller plants each summer. This hypothesis was supported by field transplant experiments in two acidic Adirondack Mountain (NY) lakes.

The efficiencies of incomplete block designs were investigated by comparing two hundred and twenty eight analyses from eleven trials using hedonic scales with corresponding randomized complete block analyses. Of the ten explanatory factors examined, only the panelist, the product type, the number of samples per session and the average score of the data had an effect on the efficiency of incomplete block designs. The effect of product type was attributed to influences of produce consumed outside the trial, and the effect of the data mean reflected decreased conscientiousness with products the panelists disliked. With three and four samples per session, incomplete block designs were 31 % and 2 % more efficient, respectively, than randomized complete block designs. When five or more samples were tested, the incomplete block designs were markedly less efficient. The practical implications of all these effects on experimental design are discussed.  相似文献
