SYNOPSIS. Methylglyoxal bis(guanylhydrazone) (MGBG) at 0.5 mm had little effect in vitro on Blastocrithidia culicis, Crithidia oncopelti, and Leishmania spp., but completely inhibited growth of Trypanosoma brucei. Inhibition became irreversible after a 3-h exposure of T. brucei culture procyclics. Treated organisms remained motile, but failed to divide. Polyamines, spermidine, and spermine, did not reverse the anti-trypanosome action of MGBG (preloading of cells or concurrent administration). Two intraperitoneal injections of the drug at a concentration of 50 mg kg body weight at a 1-day interval greatly reduced the parasitemia of T. brucei and T. congolense in rats. Trypanosome infections, however, relapsed and killed the animals in 6 days after treatment. It was evident from the results of tracer experiments with T. brucei that MGBG significantly lowered incorporation of [3H]thymidine by culture procyclics and of [3H]uridine by bloodstream forms; in both stages [3H]leucine incorporation was only slightly inhibited. It is suggested that MGBG interferes with nucleoside incorporation by Trypanosoma and that its mode of action is different in bloodstream and culture procyclics.  相似文献

The reasons for female desertion of offspring and the evolution of predominantly male care among monogamous bird species are not clearly understood. We studied parental effort during the incubation and chick rearing periods in the Eurasian Curlew Numenius arquata in western Finland, and compared timing of brood desertion with other populations in Europe. Males and females contributed equally to incubation and showed no differences in the intensity of mobbing behaviour towards a potential nest predator (stuffed crow) shortly after hatching. However, females deserted their offspring approximately halfway through the brooding period ( c. 16 d after hatching), while males remained with chicks until independence ( c. 35 d). Females with late-laid clutches deserted their offspring sooner after hatching than those with clutches produced earlier in the season. Curlew females deserted younger chicks in northeast Europe, where laying dates were later, breeding seasons shorter and migration distances were longer, than in western and central Europe. We suggest that the most likely reasons for offspring desertion by females may be associated with increased female survivorship and maintenance of pairbond between years.  相似文献

Mitochondrial and nuclear DNA information was analysed among four subspecies of the African Queen butterfly, Danaus (Anosia) chrysippus sensu lato (s.l.), along with four other Danaus species drawn from all three subgenera (D. (Danaus) plexippus, D. (Salatura) genutia, D. (A.) gilippus, D. (A.) eresimus) and two outgroup species from the same tribe, Tirumala septentrionis and Amauris niavius. A mitochondrial phylogeny derived from the 12S rRNA (347 bp) and COI (537 bp) loci indicates two very distinct haplotypes for subspecies D. (A.) c. dorippus, dorippus‐1 and dorippus‐2. Interestingly, dorippus‐1, on the one hand, and all other D. (A.) chrysippus haplotypes, on the other, are the most distantly related clades within the genus and have different most recent ancestors from different subgenera, though sharing the common ancestor of the monophyletic genus. A phylogeny based on the EF1‐α nuclear locus (400 bp) shows that the two well‐separated mitochondrial lineages of dorippus are identical for this gene and reciprocally monophyletic to the other D. (A.) chrysippus lineages. Thus, nuclear and cytoplasmic phylogenies are not only discordant, but also suggest that both D. (A.) chrysippus s.l. and subspecies dorippus are polyphyletic. Paradoxically, four African subspecies, chrysippus‐orange, chrysippus‐brown, alcippus and dorippus, though substantially vicariant, hybridize extensively in East Africa wherever the ranges of two or more of them overlap. Linkage disequilibrium, and hence sexual isolation, in sympatry between colour (nuclear) genes and unlinked mitochondrial (cytoplasmic) loci is consistent across populations and therefore indicates the operation of positive natural selection. Together with data from previous experimental and field work, our results suggest that extensive hybridization occurs among once allopatric or parapatric lineages, that are now nascent species. We deduce that hybridism among lineages in sympatry is currently enforced, in the face of assortative mate choice, by a bacterial symbiont, Spiroplasma, a male‐killer that forces females in female‐biased populations to pair with heterotypic males. In discussion we emphasize that neither D. 1. We investigate long‐term (>200 years) changes to the composition and spatial structure of macrophyte communities in a shallow, eutrophic lake (Barton Broad, eastern England) and consider the implications for lake restoration. 2. Historical macrophyte data were assembled from a variety of sources: existing plant databases, museum herbaria, journal articles, old photographs and eyewitness accounts. Additionally, two types of sediment core sample were analysed for plant macro‐remains and pollen; bulk basal samples from multiple core sites analysed to provide information on ‘pre‐disturbance’ macrophyte communities and two whole cores analysed to determine historical change. 3. Prior to the late 1800s, macrophyte communities were diverse and included a multilayered mosaic of short‐stature submerged taxa and taller submerged and floating‐leaved species. With the progression of eutrophication after around 1900, the former community was displaced by the latter. Diversity was maintained, however, since an encroaching Schoenoplectus–nymphaeid swamp generated extensive patches of low‐energy habitat affording refugia for several macrophytes otherwise unable to withstand the hydraulic forces associated with open water conditions. When this swamp vegetation disappeared in the 1950s, many of the ‘dependent’ aquatic macrophytes also declined leaving behind a sparse, species‐poor community (as today) resilient to both eutrophication and turbulent open waters. 4. The combination of historical and palaeolimnological data sources offers considerable benefits for reconstructing past changes to the aquatic vegetation of lakes and for setting restoration goals. In this respect, our study suggests that successful restoration might often be better judged by reinstatement of the characteristic structure of plant communities than the fine detail of species lists; when nutrients are low and the structure is right, the right species will follow.  相似文献

Abstract: The genus Nothofagus in New Zealand and Australia exhibits strong mast seeding (i.e. highly variable seed crops between years). Seed crop variation is synchronized within and between species over large spatial scales, and results in greatly increased wind pollination efficiency which could provide a selective benefit favouring the maintenance of mast seeding. However, the null hypothesis (that plants simply match their reproductive effort to the variable resources available each year) has not been tested in Nothofagus. Here we use a 33‐year dataset on seedfall and wood ring increments for 19 individual Nothofagus truncata trees at Orongorongo, New Zealand, to test for the presence of switching (exaggeration of seedfall variability by diverting resources into, then out of, reproduction). A generalized least squares model explained 40.7% of the variance in standardized ring widths, using six weather variables (absolute minimum temperatures in March (lag 0) April (lag 0 and lag 1), May (lag 0) and rainfall in November and February (lag 0) ) and seedfall. Seedfall had a negative relationship with the current year's ring widths even after controlling for all significant weather variables. This shows that switching is occurring in N. truncata within individuals among years, and therefore that masting in this species is the result of selective forces such as increased wind pollination efficiency. As this result has been demonstrated for very few masting species, we call for this test to be applied more widely.  相似文献

The causes of fluctuating asymmetry (FA) are poorly understood, yet it has been widely used as a measure of fitness. Many studies have demonstrated that individuals with low FA are preferred as mates, and it has been argued that this is because FA provides an indicator of genetic quality. However, the relative importance of genes versus environment in determining the level of FA shown by an organism is currently the subject of much controversy. As yet there is no clear consensus as to whether FA generally has a heritable component and if so how large this might be, or indeed if it is sensible to generalize at all. In Musca domestica flies with low wing length FA have beeen found to enjoy higher mating success. In order to interpret this finding we assess whether wing length FA in this species is heritable, and also how it is influenced by environmental stresses induced by temperature and crowding. We also examine whether offspring viability is related to parental FA. We found that wing length FA in M. domestica had no detectable heritable components, and parental FA did not influence offspring viability. FA was influenced by rearing temperature, with flies exhibiting highest FA at the lowest rearing temperature (15oC). Larval survival rate was greatest, and the resulting adults largest, at the intermediate rearing temperature (25oC) compared to higher or lower temperatures, suggesting that 25oC is close to the optimum for the development of M. domestica. Adult size appears to provide a better indicator of stress during development man does wing FA. These results are discussed in relation to the utility of FA as a tool for use in evolutionary studies.  相似文献

ABSTRACT. A method is described for obtaining large numbers of telotrochs from mass cultures of Vorticella convallaria . These free-swimming cells contract slightly along the aboral-oral plane when extracted with Triton X-100, and thus appear more similar in shape to zooids than unextracted telotrochs. Cytoskeletal structures associated with feeding, such as the infudibulum and the cytopharynx, are visible in cytoskeletal preparations of these non-feeding telotrochs and thus appear not to be disassembled during telotroch formation. The telotroch to stalked-zooid transition proceeds rapidly through a set series of morphogenetic stages. After telotroch attachment, the aboral cilia cease beating and are resorbed. Within 7 min the cell is inverted bell-shaped and the zooid begins feeding. Stalk elongation begins about 15–20 min after attachment, lengthening at the rate of 0.5 μ/min for the first hour and more slowly (0.1 μm/min) after that. Interestingly, these developmental stages are essentially the same as those described for the telotroch to zooid transition in the colonial peritrich Zoothamnium . This evolutionary conservation suggests that the precise sequence and timing of these events are critical for their successful completion. Furthermore, the facts that the telotroch to zooid transition occurs very rapidly, that the feeding structures are maintained throughout the transformation, and that basic cytoskeletal architecture is relatively unchanged is consistent with the hypothesis that the transformation occurs through controlled cytoskeletal rearrangements rather than by changes in gene expression.  相似文献
