Induction of growth-related metabolism in human vascular smooth muscle cells by low density lipoprotein 总被引:11,自引:0,他引:11

Human vascular smooth muscle cells (hVSMC) rendered quiescent by maintenance under serum-free culture conditions for 48 h exhibited several metabolic responses, normally associated with proliferation, following exposure to low density lipoprotein (LDL). LDL induced a time- and dose- (half-maximally effective concentration, ED50 25.0 +/- 8 nM) dependent activation of S6 kinase which could be negated following pretreatment of hVSMC with 12-O-tetradecanoylphorbol-13-acetate (TPA) for 48 h. In myo-[3H]inositol-prelabeled hVSMC, LDL caused a rapid (maximum within 1 min) decrease in phosphatidylinositol 4,5-bisphosphate (35% p less than 0.001) and phosphatidylinositol 4-phosphate (20%, p less than 0.01) with a return to prestimulated levels within 5-10 min. LDL induced a concomitant increase in [3H]inositol phosphates for which the order of generation was inositol-tris greater than -bis greater than -mono phosphate and which reached threshold levels of significance (p less than 0.05) above control values within 1, 2, and 10 min, respectively. The effect of LDL on hVSMC phosphoinositide metabolism was dose-dependent (half-maximally effective concentration, ED50 32.1 +/- 5.0 nM). This concentration, like that for S6 kinase, approximates with the KD (5-21 nM) for high affinity binding of 125I-LDL to specific receptors (1.5 x 10(4) sites/cell) on hVSMC. LDL induced a rapid but transient translocation of protein kinase C from the cytosol to membranes as assessed using both immunoblotting and [3H] 4-beta-phorbol-12-13-dibutyrate-binding procedures. Exposure of quiescent hVSMC to LDL elevated intracellular pH (delta pH 0.30 +/- 0.03, p less than 0.001). Such alkalinization was prevented in the presence of Na+/K+ exchange inhibitors such as amiloride, dimethylamiloride, and ethylisopropylamiloride. In an investigation of the nuclear action of LDL, a time-dependent induction of both c-myc and c-fos was observed. Such LDL-induced expression of these nuclear proto-onco-genes was not detectable in protein kinase C down-regulated hVSMC. Nevertheless, in spite of the cascade of "growth-promotional" responses elicited by LDL in quiescent hVSMC, this lipoprotein alone (under serum-free conditions) was neither mitogenic in nuclear labeling experiments, nor could it support growth of hVSMC in culture. We demonstrate that LDL might function in a complementary/synergistic fashion with other weakly mitogenic (to VSMC) growth factors and suggest that activation of protein kinase C (vis à vis intrinsic tyrosine kinase characteristic of other growth factor receptors) may be crucial to the signal transduction pathway for LDL. 相似文献

Acelylcholinesterase was purified from the electric organs of Torpediniforms nacline timelci by affinity chromatography. "Pure" enzyme preparation as identified by PAGE analysis was obtained. The specific activity is 78 mmol ACh/h/mg protein. Sedimentation coefficient is 11.86 S. Molecular weight is 236000. Acidic amino acid and basic amino acid contents are 22% and 12% respectively. Circular dichroism pattern shows two negative valleys at 209 and 220 nm.Optimum substrate concentration is 1-3 mM ACh with Km around 0.18 rnM. Excess substrate causes inhibition of enzyme activity. Optimum pH lies between 7.5-8.0. Bovine serum albumin and phosphatidylcholine benefits the stability of enzyme activity. The pure enzyme can be stored at - 20癈. Cationic liposomes enhanced the rate of transduction of target cells with retroviral vectors. The greatest effect was seen with the formulation DC-Chol/DOPE, which gave a 20-fold increase in initial transduction rate. This allowed an efficiency of transduction after brief exposure of target cells to virus plus liposome that could be achieved only after extensive exposure to virus alone. Enhancement with DC-Chol/DOPE was optimal when stable virion-liposome complexes were preformed. The transduction rate for complexed virus, as for virus used alone or with the polycation Polybrene, showed first-order dependence on virus concentration. Cationic liposomes, but not Polybrene, were able to mediate envelope-independent transduction, but optimal efficiency required envelope-receptor interaction. When virus complexed with DC-Chol/DOPE was used to transduce human mesothelioma xenografts, transduction was enhanced four- to fivefold compared to that for virus alone. Since the efficacy of gene therapy is dependent on the number of cells modified, which is in turn dependent upon the balance between transduction and biological clearance of the vector, the ability of cationic liposomes to form stable complexes with retroviral vectors and enhance their rate of infection is likely to be important for in vivo application. 相似文献

The past 40 years have seen the legal and policy framework for nature conservation in Britain extend from protection and preservation to include enhancement through techniques such as ecological restoration and habitat creation. Clear objectives need to be set for ecological restoration and habitat creation schemes because the processes involve human intervention in combination with natural factors operating over time. Objectives are required for both management and monitoring in order to enable measurement of success or failure. The most effective way to achieve high-quality restoration and creation schemes is to define the output of the process—a habitat, vegetation type, or biological community. The better the definition of the output, the greater the need to define the inputs and the nature of the intervening processes. The receptor (EGFR) for epidermal growth factor (EGF) and transforming growth alpha (TGFα) is often overexpressed in certain types of human malignancy and high levels of expression of the receptor and/ or coexpression of growth factors. EGF and TGFα have also been correlated with poor prognosis in many patients. We have produced a number of rat monoclonal antibodies (MAbs) against four distinct epitopes on the external domain of the EGF receptor and are currently evaluating their potential for therapeutic use. Nine of these of MAbs were found to inhibit the binding of TGF and EGF to the receptor on tumor cells and these MAbs were able to inhibit the growth in vitro and in vivo of tumor cells that overexpress the EGF receptor. Here, we describe the results of experiments to determine the antitumor activity of combination treatment with two antibodies directed against separate epitopes on the external domain of human EGFR. Our results showed that treatment of human tumor xenografts with a combination of two anti-EGFR MAbs that bind to two distinct epitopes on the external domain of EGF receptor was not as effective as treatment with ICR62 alone, which binds to epitope C on the EGFR and is of IgG2b isotype. A phase I clinical trial with antibody ICR62 is currently underway in Royal Marseden Hospital (UK) in patients with head and neck and lung carcinomas. 相似文献

Exogenous stages of Isospora serini (Aragao) and Isospora canaria sp. n. from the canary (Serinus canarius Linnaeus) are described. Oocytes of I. serini are spheroid and average 19.2 times 20.1 mum, while those of I. canaria are larger, more ellipsoid, and average 21.8 times 24.6 mum. No oocyst residuum is present and the oocyst walls of both species are colorless, transparent, and single layered. Sporocysts average 9.4 times 15.2 mum for I. serini and 11.5 times 18.1 mum for I. canaria. The I. canaria sporocyst has a substiedal body, but none was found in I. serini sporocysts. Both species have a spherical sporocyst residuum; this was obscured in the I. serini sporocyst by scattered granules. Living sporozoites of I. canaria average 3.6 times 16.9 mum and have 1 to 3 refractile globules; those of I. serini have 2 globules and average 2.8 times 12.6 mum. A disseminated infection of the mononuclear phagocytes results from administration of I. serini while I. canaria oocysts give rise to a typical coccidian infection restricted to the intestinal epithelium. Asexual stages of I. serini in macrophages are indistinguishable from parasites previously called avian Toxoplasma, Atoxoplasma, and Lankesterella. 相似文献

The antioxidant enzyme response of the mussel Mytilus galloprovincialis to different degree of pollution was investigated. Antioxidant enzyme activities - catalase (CAT), glutathione peroxidase (GSH-PX), glutathione reductase (GR), superoxide dismutase (SOD) - and malondialdehyde (MDA) concentration were measured in gills and digestive glands of mussels. Mussels from the same origin were transplanted along the Balearic coastal waters in eight stations characterized by a different degree of contamination and human impacts. Antioxidant enzyme activities showed an adaptive response to increase the activities in the more polluted areas. CAT, GR and SOD in gills and CAT and GR in digestive gland presented significant differences between polluted and non-polluted stations. No significant differences were observed in MDA concentration indicating that the antioxidant response is capable to avoid the lipid peroxidation. The use of biomarkers such as CAT and GR in gills and digestive glands of the mussel M. galloprovincialis is a good tool to categorize differences between polluted and non-polluted areas. 相似文献
