We have characterised the interaction of the Aeropyrum pernix origin recognition complex proteins (ORC1 and ORC2) with DNA using DNase I footprinting. Each protein binds upstream of its respective gene. However, ORC1 protein alone interacts more tightly with an additional region containing multiple origin recognition box (ORB) sites that we show to be a replication origin. At this origin, there are four ORB elements disposed either side of an A+T-rich region. An ORC1 protein dimer binds at each of these ORB sites. Once all four ORB sites have bound ORC1 protein, there is a transition to a higher-order assembly with a defined alteration in topology and superhelicity. Furthermore, after this transition, the A+T-rich region becomes sensitive to digestion by DNase I and P1 nuclease, revealing that the transition promotes distortion of the DNA in this region, presumably as a prelude to loading of MCM helicase. 相似文献

We examined the phylogenetic relationships among five heterothallic species of Neurospora using restriction fragment polymorphisms derived from cosmid probes and sequence data from the upstream regions of two genes, al-1 and frq. Distance, maximum likelihood, and parsimony trees derived from the data support the hypothesis that strains assigned to N. sitophila, N. discreta, and N. tetrasperma form respective monophyletic groups. Strains assigned to N. intermedia and N. crassa, however, did not form two respective monophyletic groups, consistent with a previous suggestion based on analysis of mitochondrial DNAs that N. crassa and N. intermedia may be incompletely resolved sister taxa. Trees derived from restriction fragments and the al-1 sequence position N. tetrasperma as the sister species of N. sitophila. None of the trees produced by our data supported a previous analysis of sequences in the region of the mating type idiomorph that grouped N. crassa and N. sitophila as sister taxa, as well as N. intermedia and N. tetrasperma as sister taxa. Moreover, sequences from al-1, frq, and the mating-type region produced different trees when analyzed separately. The lack of consensus obtained with different sequences could result from the sorting of ancestral polymorphism during speciation or gene flow across species boundaries, or both. 相似文献

Callus has been induced on needle and bud explants from 2, 6 and 26 year old Picea abies (L.) Karst, trees. Seventy seven percent of the needle explants from the youngest material was productive compared to 0.2% for the old tree. New embryogenic sections appeared in the non-embryogenic callus derived from 2, 6 and 26 year old trees after prolonged culture and was promoted by the use of an established embryogenic callus line in a nurse culture system.Abbreviations EDTA

Peptide maps of Form A and Form B of porcine submaxillary gland beta Gal alpha 2----3 sialyltransferase were essentially identical, consistent with the view that the two forms are not different enzyme species but that one, the B form (Mr = 44,000) is derived from the A form (Mr = 49,000). Analysis of the sialyltransferase activity in subcellular fractions from homogenates of porcine submaxillary glands reveals that 85% of the total activity of the transferase is bound to membranes, mostly in the Golgi apparatus, and that the remainder is soluble. The relative amounts of the membrane-bound and soluble forms as well as their response to detergents suggests that they are the cellular counterparts to the A and B forms of the transferase. The activity of Form A and the membrane-bound enzyme is stimulated to similar extents by various detergents. Triton-type detergents are more effective than Brij-type. Lysophosphatidylcholine is a potent stimulator of the activity of Form A but lysophosphatidylethanolamine is without effect and lysophosphatidylserine and lysophosphatidylglycerol are inhibitory. C16-18 acyl derivatives of lysophosphatidylcholine stimulate the activity more extensively than the C14 acyl derivative, and the C12 acyl derivative is without effect. In contrast, Form B is fully active in the absence of all detergents tested although it is inactivated just as Form A by lysophosphatidylglycerol and octylglucoside. Kinetic analysis of Forms A and B reveal that detergents stimulate the activity of Form A by lowering the KD and KM of CMP-NeuAc and increasing the Vmax of the reaction. Form B in contrast, which is fully active in the absence of detergents, has kinetic parameters like those of Form A in the presence of detergent. Taken together, these results suggest that Form A of the sialyltransferase, but not Form B, contains a lipid-binding domain, and that binding of detergents or lipids to the domain modulates the activity of the enzyme. 相似文献

Summary Under hypoxia (10 and 5% partial oxygen tension) meristematic cells ofAllium cepa L. roots acquired new cycle kinetics, characterized by reduced but constant rates of root growth. Under these conditions, there was preferential lengthening of G1 and of the last third of the S period, S3. Since hyperoxygenation shortened S3 but not G1 in these cells, the high sensitivity of late replication to environmental oxygen is demonstrated. The preferential depression of the replication rate when those cells replicated the last third of their DNA was not associated with diminished cell size. Rather, the lower the oxygen level the larger the mean size of the cycling cells. Under anoxia (0% oxygen tension) the rate of growth slowed, accompanied by preferential accumulation of cells in G1. ABSTRACT. The microsporidium, Enterocytozoon bieneusi , causes a severe, debilitating, chronic diarrhea in patients with the acquired immunodeficiency syndrome. Specific diagnosis of intestinal microsporidiosis, especially due to Enterocytozoon , is difficult and there is no known therapy that can completely eradicate this parasite. Preliminary studies indicate that a short term (about 6 months) in vitro culture of this parasite yielding low numbers of spores, may be established by inoculating human lung fibroblasts and/or monkey kidney cell cultures with duodenal aspirates and or biopsy from infected patients. The cultures may subsequently be used for the isolation and molecular analysis of parasite DNA. 相似文献

The present studies were undertaken to examine the hypothesis that ethanol could affect cellular biosynthesis in the murine mastocytoma cell of prostaglandins and leukotrienes, oxidative metabolites of arachidonic acid, at concentrations that could be encountered in vivo as well as during in vitro experiments. The effects of ethanol which encompass these concentration ranges (200-1000 mg%) can be summarized as follows: first in the absence of exogenous arachidonic acid, ethanol caused a dose dependent decrease in the production of leukotrienes which was statistically significant at 200 mg%. At 1000 mg%, ethanol caused a 20-50% decrease in leukotrienes and a 21% decrease in the amount of prostaglandin D2 (PGD2) formed in these cells. Secondly, when cells were incubated with exogenous arachidonic acid (14 micrograms/ml), large increases in both PGD2 and leukotrienes occurred. Under these conditions, ethanol caused a further increase in the amount of leukotrienes and a small increase in the amount of PGD2 formed. This stimulatory effect was specific for ethanol since neither t-butanol nor n-butanol caused the enhanced production of leukotrienes with exogenous arachidonic acid. Thus, these experiments suggest that ethanol affects metabolism of arachidonic acid at reasonably low doses (200-400 mg%) of ethanol in a manner dependent on the free arachidonic acid in the tissue. Also, in vitro experiments in which ethanol is used as a solvent for arachidonic acid could be greatly affected by high levels of ethanol (500-1000 mg%) which are frequently utilized. 相似文献

We combine two techniques in order to discuss the time-varying elastic properties of the left ventricular muscle. An analytic
model for the shape and forces in the left ventricle is combined with the Fourier series representations of certain of the
ventricular dimensions and pressure to derive expressions for the stress and strain in the left ventricle. The strain is thus
a function of the elastic material properties, which are then expressed as functions of time by using Fourier series. The
only data needed for a numerical study using these techniques are closed-chest determinations of the ventricular dimensions
