? The exploitation of natural variation in Arabidopsis thaliana (Arabidopsis) provides a huge potential for the identification of the molecular mechanisms underlying this variation as a result of the availability of a vast array of genetic and genomic resources for this species. Eighty-six Arabidopsis accessions were screened for natural variation in flooding tolerance. This forms the first step towards the identification and characterization of the role of candidate genes contributing to flooding tolerance. ? Arabidopsis accessions at the 10-leaf stage were subjected to complete submergence in the dark. Survival curves were plotted to estimate median lethal times as a measure of tolerance. Flooding-associated survival parameters, such as root and shoot oxygen content, initial carbohydrate content and petiole elongation under water, were also measured. ? There was a significant variation in submergence tolerance among Arabidopsis accessions. However, the order of tolerance did not correlate with root and shoot oxygen content or initial amounts of shoot starch and total soluble sugars. A negative correlation was observed between submergence tolerance and underwater petiole elongation. ? Arabidopsis accessions show considerable variation in the ability to tolerate complete submergence, making it a good species in which to identify and characterize genes and to study mechanisms that contribute to survival under water. 相似文献

A genetic analysis of the Triticum aestivum/Aegilops umbellulata introgressive line for the ear awnedness trait was carried out. Based on the results obtained in the study of awnedness and microsatellite components in lines and hybrids, a digenic model was proposed for the control of this trait by two genes??inhibitor of awnedness B1 (5AL) and promoter of awnedness Awn1, which are both located on chromosome 6U of Aegilops umbellulata. Chromosome 5A of the awned line was rearranged with respect to the same chromosome of the Aurora genotype. The genetic distance between the SSR-locus Xwmc705-5AL and the rearranged fragment or locus B1/b1 was determined to be 15.5 cM. 相似文献

Cytosolic protein phosphotyrosine phosphatases from rabbit kidney. Purification of two distinct enzymes that bind to Zn2+-iminodiacetate agarose 总被引:14,自引:0,他引:14
Cytosolic protein phosphotyrosine (PPT) phosphatase was measured using a new substrate, Tyr(32P)-labeled bovine serum albumin. Kidney was found as a particularly rich tissue source of PPT-phosphatase activity, containing twice as much as liver and over 10-fold more than brain, heart, lung, or skeletal muscle. An affinity column of Zn2+-iminodiacetate agarose adsorbed up to 60% of the PPT-phosphatase present in kidney extracts. Subsequent chromatography on DEAE-Sepharose separated the phosphatase into two peaks, labeled I and II, that had Mr = 34,000 and 37,000, respectively, upon gel filtration with Sephadex G-75 Superfine. Overall purification of 850- and 1100-fold was achieved with a net 4% yield. Both phosphatases hydrolyzed p-nitrophenylphosphate as well as the protein substrate in the presence of EDTA. Peak I phosphatase activity displayed a neutral pH optimum, had an absolute requirement for sulfhydryl compounds, and was sensitive to trypsin, whereas Peak II activity had an acidic pH optimum and was active without mercaptans. The two proteins also gave different fragmentation patterns by gel electrophoresis after digestion with S. aureus V8 protease. The results show that multiple forms of PPT phosphatase specifically interact with Zn2+ and provide a basis for further structural and functional comparisons among different members of the phosphoprotein phosphatase family. 相似文献

Pressure distributions were measured along a hollow vascular axisymmetric replica of a segment of the left circumflex coronary artery of man with mildly atherosclerotic diffuse disease. A large range of physiological Reynolds numbers from about 60 to 500, including hyperemic response, was spanned in the flow investigation using a fluid simulating blood kinematic viscosity. Predicted pressure distributions from the numerical solution of the Navier-Stokes equations were similar in trend and magnitude to the measurements. Large variations in the predicted velocity profiles occurred along the lumen. The influence of the smaller scale multiple flow obstacles along the wall (lesion variations) led to sharp spikes in the predicted wall shear stresses. Reynolds number similarity was discussed, and estimates of what time averaged in vivo pressure drop and shear stress might be were given for a vessel segment. 相似文献

Somatostatin (SS) inhibits secretion from many cells, including clonal GH3 pituitary cells, by a complex mechanism that involves a pertussis toxin (PTX)-sensitive step and is not limited to its cAMP lowering effect, since secretion induced by cAMP analogs and K+ depolarization are also inhibited. SS also causes membrane hyperpolarization which may lead to decreases in intracellular Ca2+ need for secretion. Using patch clamp techniques we now demonstrate: 1) that both (SS) and acetylcholine applied through the patch pipette to the extracellular face of a patch activate a 55-picosiemens K+ channel without using a soluble second messenger; 2) that, after patch excision, the active state of the ligand-stimulated channel is dependent on GTP in the bath, is abolished by treatment of the cytoplasmic face of the patch with activated PTX and NAD+, and after inactivation by PTX, is restored in a GTP-dependent manner by addition of a nonactivated human erythrocyte PTX-sensitive G protein, and 3) that the 55-picosiemens K+ channel can also be activated in a ligand-independent manner with guanosine [gamma-thio] triphosphate (GTP gamma S) or with Mg2+/GTP gamma S-activated erythrocyte G protein. We call this protein GK. It is an alpha-beta-gamma trimer of which we have previously shown that the alpha-subunit is the substrate for PTX and that it dissociates on activation with Mg2+/GTP gamma S into alpha-GTP gamma S plus beta-gamma. Yellow perch (YP) are metal tolerant fish that form large populations in many metal-impacted regions across Canada. While they are able to survive and reproduce successfully in environments with water and sediment metal concentrations that are toxic to many invertebrates, perch experience a suite of direct and indirect impacts. YP were studied in a series of Canadian Precambrian Shield lakes near Sudbury, Ontario, along a gradient of metal exposure downwind from metal smelters. In lakes at the high end of our exposure gradient, concentrations of metals (Cu, Ni, and Cd) in YP liver and kidney were well above levels seen in fish from reference lakes. Direct effects linked to metal-exposure were observed, ranging from effects at the cellular level, to effects in organs and tissues, individuals, and populations. In addition to direct or physiological effects, we also documented indirect, foodweb-mediated effects of metals on YP in the most contaminated lakes resulting from the elimination of metal-sensitive large benthic invertebrates and their replacement by small metal-tolerant taxa. The most common indication of such indirect effects on YP is severely stunted growth coupled with a high degree of zooplankton dependence throughout their life. Such indirect effects have important implications for ecological risk assessment (ERA) because they indicate that higher trophic levels may be functionally altered even though the functional groups that they depend on are present and abundant. Although the functional groups important to yellow perch can be abundant in metal-impacted lakes, their benthic communities are impoverished and this is strongly reflected in their size structure upon which energy transfer to higher trophic levels depends. Thus indirect (foodweb-mediated) effects can be important in situations where invertebrate size structure is impacted in such a way as to reduce the efficiency of energy transfer to higher trophic levels, and therefore measures should be taken to protect and/or restore large sensitive benthic species. 相似文献
