Conformational study of native and acid-dissociated states of porcine lutropin (pLH) by vacuum circular dichroism and ultraviolet absorption spectroscopy

Porcine lutropin shares with ovine lutropin common structural features. They exhibit identical vacuum circular dichroism down to 170 nm with characteristic negative bands at 173, 194, and 210 nm. The band at 210 ± 1 nm is shifted to 201 nm upon dissociation with disappearance of the 194-nm band. For the two hormones the acid transition involves a significant loss of the three recognized periodic structures α helix, β sheet, and β turns of type II, unshields near the same number of tyrosyl residues (2.2 ± 0.7), and gives rise to an identical absorption difference at 287 nm in a two-step mechanism. However, pLH also exhibits noticeable differences: 5 to 10 times lower rates of acid transition with a lower pK_a (3.7 ± 0.1) and different transition behavior of tyrosine residues compared to ovine lutropin.     

Identification of N- and C-terminal corticotropin peptides in the Mr 80000 form of neurophysin

The 125I-labeled Mr 80000 form of neurophysin has been purified from bovine neurohypophysi. Tryptic digests of this species were analyzed, prior to or after treatment with carboxypeptidase B, by high-pressure liquid chromatography followed by isoelectric focusing and the fragments compared with those generated by a similar treatment of reference bovine 1-39 adrenocorticotropin. The ACTH peptides 22-39 and 1-8, as well as the 1-7 derivative of the latter were identified by those two independent criteria. This provides chemical evidence supporting the hypothesis [8] that high Mr neurophysin may contain the sequence of ACTH.     

Correlations between variations of electrical field at the body surface and contractions of the stomach in guinea pig.

1. The slow potentiel changes of the base line of the electrocardiogram recorded by cutaneous electrodes, here called EGEG (electrogastroenterogram) have been recorded simultaneously with the electromyogram and pressure variations of the stomach antrum. 2. Periodic fluctuations of EGEG, 400 microV amplitude, were correlated with activities of the antrum EMG consisting of 3 to 6 mV spikes superimposed on slow wave of depolarization, and with gastric intraluminal pressure of 5 to 7 cm of water. 3. An anticholinergic drug abolished all those activities. A gastric contracture flattened the EGEG. 4. Rhythmic distensions of the antrum induced a synchronous modulation of EGEG, the magnitude of which was correlated with the amplitude of distension. These result suggest that the origin of EGEG is essentially the movements of stomach.     

Association-dependent active folding of alpha and beta subunits of lutropin (luteinizing hormone).

The kinetics of the structural transition of ovine lutropin (luteinizing hormone) from a dissociated and partially unfolded state to a biologically active, folded and associated state were studied from pH 1.8 to 6.5 by difference spectroscopy, circular dichroism, light scattering and ultracentrifugation under various conditions of temperature and ionic strength. Between pH 2.8 and 5.3 there is a thermodynamically reversible equilibrium between the two states of the hormone with a half-transition pH of 4.3 ± 0.1 at 26 °C. The interconversion of native folding to dissociated state is strictly first-order, and most of the kinetic results can be described in terms of two exponential decays with lifetimes of 20 and 100 seconds at pH 2 and 26 °C with one intermediate. A second intermediate with a short lifetime (1.5 s) is detected with stopped-flow experiments; its spectroscopic contribution is small. Refolding from the dissociated state is always second-order in the concentration range studied (12 to 110 μm lutropin), with rates at 26 °C, 1.4 m⁻¹ s⁻¹ at pH 4.3 and 2.2 m⁻¹ s⁻¹ at pH 5.3. The temperature dependence of the rate constant of active folding at pH 5.3 corresponds to activation parameters $\text{ΔH}{}^1\text{= +23.5}\text{kcal mol}{}^{\mathrm{-1}}\text{and}\text{ΔS}{}^1\text{= +21.6}\text{cal deg}{}^{\mathrm{-1}}$ mol⁻¹.From these data and computer-simulation studies, a simplest possible mechanism is proposed that involves the formation of a loose complex between the α and β subunits of the hormone, followed by a slow step of formation of the active, folded state. In the pituitary gland this would be a necessary pathway towards the active form. The storage of the hormone in the gland would offer enough time for this activation step to proceed.

Affinity Chromatography of B-Glucosidase and Endo-B-Glucanase from Aspergillus Niger on Concanavalin A-Sepharose: Implications for Cellulase Component Purification and Immobilization

Affinity chromatography of a commercial preparation of 3-glu-cosidase from Aspergillus niger using concanavalin A-Sepharose (CAS) was employed as a means of purifying this glycoprotein. However, mannose (up to 1.08 M) was ineffective as an eluent of this enzyme from CAS, as were several other sugars and their derivatives, including 0.5 M glucose. Also, washing the CAS:8-glucosidase complex with buffer at pH 3.5 in the absence of MnCl₂ and CaCl₂ (required to preserve the binding activity of concanavalin A below pH 5.0) did not result in elution of this enzyme. On the contrary, endo-glucanase activity present in a crude cellulase complex (A. niger) which bound to CAS could be eluted by mannose (0.5–0.7 M) and was fractionated Into at least two components. The CAS:β-glucosidase complex hydrolyzed cellobiose to glucose and possessed an activity of 2, 158 units/g dry CAS. It could be used, therefore, for continuous cellobiose hydrolysis without leakage of enzyme from the support.     

Sensitivity and Specificity of a New Vertical Flow Rapid Diagnostic Test for the Serodiagnosis of Human Leptospirosis

Background: Leptospirosis is a growing public health concern in many tropical and subtropical countries. However, its diagnosis is difficult because of non-specific symptoms and concurrent other endemic febrile diseases. In many regions, the laboratory diagnosis is not available due to a lack of preparedness and simple diagnostic assay or difficult access to reference laboratories. Yet, an early antibiotic treatment is decisive to the outcome. The need for Rapid Diagnostic Tests (RDTs) for bedside diagnosis of leptospirosis has been recognized. We developed a vertical flow immunochromatography strip RDT detecting anti-Leptospira human IgM and evaluated it in patients from New Caledonia, France, and French West Indies. Methodology/Principal Findings: Whole killed Leptospira fainei cells were used as antigen for the test line and purified human IgM as the control line. The mobile phase was made of gold particles conjugated with goat anti-human IgM. Standards for Reporting of Diagnostic Accuracy criteria were used to assess the performance of this RDT. The Microscopic Agglutination Test (MAT) was used as the gold standard with a cut-off titer of ≥400. The sensitivity was 89.8% and the specificity 93.7%. Positive and negative Likelihood Ratios of 14.18 and 0.108 respectively, and a Diagnostic Odds Ratio of 130.737 confirmed its usefulness. This RDT had satisfactory reproducibility, repeatability, thermal tolerance and shelf-life. The comparison with MAT evidenced the earliness of the RDT to detect seroconversion. When compared with other RDT, the Vertical Flow RDT developed displayed good diagnostic performances.

Conclusions/Significance

This RDT might be used as a point of care diagnostic tool in limited resources countries. An evaluation in field conditions and in other epidemiological contexts should be considered to assess its validity over a wider range of serogroups or when facing different endemic pathogens. It might prove useful in endemic contexts or outbreak situations.     

A DNA Metabarcoding Study of a Primate Dietary Diversity and Plasticity across Its Entire Fragmented Range

In tropical regions, most primary ecosystems have been replaced by mosaic landscapes in which species must cope with a large shift in the distribution of their habitat and associated food resources. Primates are particularly vulnerable to habitat modifications. Most species persist in small fragments surrounded by complex human-mediated matrices whose structure and connectivity may strongly influence their dispersal and feeding behavior. Behavioral plasticity appears to be a crucial parameter governing the ability of organisms to exploit the resources offered by new matrix habitats and thus to persist in fragmented habitats. In this study, we were interested in the dietary plasticity of the golden-crowned sifaka (Propithecus tattersalli), an endangered species of lemur, found only in the Daraina region in north-eastern Madagascar. We used a DNA-based approach combining the barcoding concept and Illumina next-generation sequencing to (i) describe the species diet across its entire range and (ii) evaluate the influence of landscape heterogeneity on diet diversity and composition. Faeces from 96 individuals were sampled across the entire species range and their contents were analyzed using the trnL metabarcoding approach. In parallel, we built a large DNA reference database based on a checklist of the plant species of the Daraina region. Our results suggest that golden-crowned sifakas exhibit remarkable dietary diversity with at least 130 plant species belonging to 80 genera and 49 different families. We highlighted an influence of both habitat type and openness on diet composition suggesting a high flexibility of foraging strategies. Moreover, we observed the presence of numerous cultivated and naturalized plants in the faeces of groups living in forest edge areas. Overall, our findings support our initial expectation that P. tattersalli is able to cope with the current level of alteration of the landscape and confirm our previous results on the distribution and the dispersal ability of this species.
