Secale vavilovii PMCs have more univalents and a lower frequency of bound arms at metaphase I than other diploid Secale species. The spreading technique applied at prophase I showed that the nuclei were able to complete synapsis at pachytene. However, 25% of the nuclei analyzed, which had more than 90% of their total length paired, showed two abnormalities: long fold-back loops, which were located mainly on the nucleolar organizer bivalent, and pairing-partner switches, probably involving all the chromosome complement. These synaptic abnormalities are unusual in diploid species and give rise to a high frequency of nonhomologous pairing regions and, therefore, could produce desynapsis, which could explain the data obtained from metaphase I. The possible origin of the unusual synaptic abnormalities of S. vavilovii is discussed.  相似文献

A variety of adhering junction is found in the ependyma of the domestic cat with a coupling of mitochondria. These are symmetrically situated (in mirror form) at both sides of the intercellular cleft, which always maintain the same separating distance, thereby leaving a limiting cellular space of a constant amplitude. The hypothesis is put forward that the energy (ATP), provided by the mitochondria over adhering junctional complexes, would produce separate fields of force which would position in a lengthwise direction the molecules which give rise to the anchoring filaments. The mitochondrial energy provided and the electrostatic forces generated would produce an adhering, intercellular junction which is functionally very strong and which could be called: high-energy adhering junctional complexes or with mitochondrial coupling.  相似文献

This paper reports the glutenin diversity of a collection of 23 wild populations of the grass Brachypodium distachyon collected in the Mediterranean and southern areas of the Iberian Peninsula. The plant material studied included the three different cytotypes of this species: 2n?=?10, 2n?=?20 and 2n?=?30. A specific method of extraction was used to isolate the glutenin subunits from the caryopsis. Separation by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) showed them to correspond to wheat low-molecular-weight glutenin subunits (LMW-GS). Twenty-two LMW-GS?Clike were identified that showed great diversity within and between populations. All the populations investigated were polymorphic for the endosperm proteins studied. The 2n?=?30 forms had the largest number of subunits; these were also more diverse than those of the 2n?=?10 or 2n?=?20 forms. The 2n?=?10 forms, the most common in the higher, interior areas of the Iberian Peninsula, showed the smallest subunit variation. In fact, negative correlations were found between subunit diversity and altitude and longitude. In contrast, a positive correlation was detected with the annual average temperature and the indices of thermicity and Mediterraneity. The similarity between the populations was estimated using the Sorensen?CDice coefficient, calculated on the basis of the presence/absence of the 22 LMW-GS proteins. The similarity indices were used to produce a dendrogram using the unweighted pair-group method with arithmetic means (UPGMA). This method produced three main groups corresponding to the three cytotypes. An analysis was made of the possible correlation between eco-geographic and climatic factors and the gene diversity and polymorphism shown in the populations. The diversity correlated positively with the number of chromosomes (2n), annual mean temperature, index of thermicity and index of Mediterraneity. In contrast, diversity correlated negatively with altitude, longitude and index of precipitation during the summer. The number of chromosomes correlated negatively with altitude, longitude and precipitation during summer and positively with all the other climatic indices. These results are coherent with the fact that diploid forms were more common in areas of the interior and at higher altitude, where the climate is more extreme.  相似文献

Several labdane diterpenes exert anti-inflammatory and cytoprotective actions; therefore, we have investigated whether these molecules protect cardiomyocytes in an anoxia/reperfusion (A/R) model, establishing the molecular mechanisms involved in the process. The cardioprotective activity of three diterpenes (T1, T2 and T3) was studied in the H9c2 cell line and in isolated rat cardiomyocyte subjected to A/R injury. In both cases, treatment with diterpenes T1 and T2 protected from A/R-induced apoptosis, as deduced by a decrease in the percentage of apoptotic and caspase-3 active positive cells, a decrease in the Bcl-2/Bax ratio and an increase in the expression of antiapoptotic proteins. Analysis of cell survival signaling pathways showed that diterpenes T1 and T2 added after A/R increased phospho-AKT and phospho-ERK 1/2 levels. These cardioprotective effects were lost when AKT activity was pharmacologically inhibited. Moreover, the labdane-induced cardioprotection involves activation of AMPK, suggesting a role for energy homeostasis in their mechanism of action. Labdane diterpenes (T1 and T2) also exerted cardioprotective effects against A/R-induced injury in isolated cardiomyocytes and the mechanisms involved activation of specific survival signals (PI3K/AKT pathways, ERK1/2 and AMPK) and inhibition of apoptosis.  相似文献

Several Lupinus species, for example, Lupinus albus, Lupinus luteus, Lupinus angustifolius, and Lupinus hispanicus were used to accumulate Mn(II), Cd(II), Pb(II), Cr(III), Cr(VI), Hg2+, and CH3Hg+ from waste waters. The influence of different species concentrations (50 and 100 mg L-1) and pH on growing behavior as well as the resulting distribution of metals in the plants were investigated. The results obtained showed that lupins were able to germinate and to grow in the presence of the metals mentioned above, even when they were present at levels as high as 50 mg L-1. Accumulation of Pb(II), Cr(III), and Cd(II) was higher in roots than in shoots. As far as mercury is concerned, the highest CH3Hg and Hg2+ accumulation was detected in roots, but fast transport toward the leaves was noticed. In contrast to mercury, the uptake of chromium seems to be influenced by the chemical form of the analyte, remaining Cr(VI) in solution. No differences in growing behavior and accumulation were observed for the four Lupinus species studied. Even though plants were exposed only a relatively short time to the metal solutions, metal concentrations of approximately 2 g/kg of dry matter were detected in the young lupins plants. The feasibility of utilizing Lupinus plants for the removal of heavy metals from wastewater was also investigated. Lupins were able to grow under extreme conditions (wastewater, pH lower than 2) and to remove 98% of the initial amount of toxic metals present in the sample.  相似文献

Triticum timopheevii (2n=4x=AtAtGG) is an allotetraploid wheat which shows a diploid-like behaviour at metaphase-I. The synaptic process was analyzed in fully traced spread nuclei at mid-zygotene, late-zygotene and pachytene. The length and type of synaptonemal complexes, as well as the number of bivalent and multivalent associations, were determined in each nucleus. A high number of bivalents per nucleus was detected at all three stages. Nuclei at pachytene showed a lower frequency of multivalents than did zygotene nuclei, which suggests the existence of a pairing correction mechanism. At metaphase-I only homologous bivalents and, rarely, one pair of univalents were observed. Similarities between the diploidization mechanism of T. timopheevii and that of allohexaploid wheat, controlled by chromosome 5B, are discussed.  相似文献
