Monoamine transporters (MATs) function by coupling ion gradients to the transport of dopamine, norepinephrine, or serotonin. Despite their importance in regulating neurotransmission, the exact conformational mechanism by which MATs function remains elusive. To this end, we have performed seven 250 ns accelerated molecular dynamics simulations of the leucine transporter, a model for neurotransmitter MATs. By varying the presence of binding-pocket leucine substrate and sodium ions, we have sampled plausible conformational states representative of the substrate transport cycle. The resulting trajectories were analyzed using principal component analysis of transmembrane helices 1b and 6a. This analysis revealed seven unique structures: two of the obtained conformations are similar to the currently published crystallographic structures, one conformation is similar to a proposed open inward structure, and four conformations represent novel structures of potential importance to the transport cycle. Further analysis reveals that the presence of binding-pocket sodium ions is necessary to stabilize the locked-occluded and open-inward conformations.  相似文献

The dopamine transporter (DAT) operates via facilitated diffusion, harnessing an inward Na+ gradient to drive dopamine from the extracellular synaptic cleft to the neuron interior. The DAT is relevant to central nervous system disorders such as Parkinson disease and attention‐deficit hyperactivity disorder and is the primary site of action for the abused psychostimulants cocaine and amphetamines. Crystallization of a DAT homolog, the bacterial leucine transporter LeuT, provided the first reliable 3‐D DAT template. Here, the LeuT crystal structure and the DAT molecular model have been combined with their respective substrates, leucine and dopamine, in lipid bilayer molecular dynamics simulations toward tracking substrate movement along the protein's substrate/ion permeation pathway. Specifically, movement of residue pairs that comprise the “external gate” was followed as a function of substrate presence. The transmembrane (TM) 1 arginine‐TM 10 aspartate strut formed less readily in DAT compared with LeuT, with or without substrate present. For LeuT but not DAT, the addition of substrate enhanced the chances of forming the TM 1‐10 bridge. Also, movement of the fourth extracellular loop EL‐4 in the presence of substrate was more pronounced for DAT, the EL‐4 unwinding to a degree. The overall similarity between the LeuT and DAT molecular dynamics simulations indicated that LeuT was a legitimate model to guide DAT structure‐function predictions. There were, nevertheless, differences significant enough to allow for DAT‐unique insights, which may include how cocaine, methylphenidate (Ritalin, NIDA Drug Supply, Rockville, MD), and other DAT blockers are not recognized as substrates even though they can access the primary substrate binding pocket. The seasonal change, i.e. the marked differences between seasons of low and high productivity, in the abundance of ectosymbionts and the defence intensity of the host against pathogens is a well defined characteristic of temperate zone organisms. Here we investigate the seasonal variation in the uropygial gland size and the abundance of Proctophyllodes feather mites on the wing feathers of house sparrows Passer domesticus in two breeding populations. The size of the uropygial gland varied significantly in male and female house sparrows over the annual cycle. The gland was small during the non‐breeding and mating season, after that it started to grow sharply, reaching its maximum size during breeding. Females had larger gland volumes than males during breeding, and the increase in gland size during breeding was more pronounced in females than in males. The number of feather mites was the lowest during breeding, followed by an increase during moult, and reaching its maximum between the wintering and mating seasons. The absence of a significant relationship between the uropygial gland size and the abundance of feather mites, after controlling for potential confounding variables, supports the view that gland oils do not regulate the number of mites. To investigate further this hypothesis, through a full factorial experimental design we tested the effect of uropygial gland and photoperiod manipulation on the population size and population dynamics of feather mites. The manipulation of uropygial gland had no effect on mites, supporting our observational results. As a result of the experimentally increased day‐length, the abundance of feather mites on wing feathers decreased significantly and more sharply than in the control group, supporting the previous anecdotal evidence about the photosensitivity of these organisms. Using photoperiodic cues, feather mites may respond to seasonal changes that affect their life‐history and population dynamics.  相似文献

Social groups often consist of diverse phenotypes, including personality types, and this diversity is known to affect the functioning of the group as a whole. Social selection theory proposes that group composition (i.e. social environment) also influences the performance of individual group members. However, the effect of group behavioural composition on group members remains largely unexplored, and it is still contentious whether individuals benefit more in a social environment with homogeneous or diverse behavioural composition. We experimentally formed groups of house sparrows Passer domesticus with high and low diversity of personality (exploratory behaviour), and found that their physiological state (body condition, physiological stress and oxidative damage) improved with increasing group-level diversity of personality. These findings demonstrate that group personality composition affects the condition of group members and individuals benefit from social heterosis (i.e. associating with a diverse set of behavioural types). This aspect of the social life can play a key role in affiliation rules of social animals and might explain the evolutionary coexistence of different personalities in nature.  相似文献

A novel multiscale model and simulation has been developed by combining the algorithms from a course grained 2D lattice model with Brownian dynamics (BD). The lattice model incorporates Lennard-Jones and coulombic interactions between particles, as well as nearest-neighbor interactions. The BD simulation allows the particles to move in solution before they adsorb to the surface. This hybrid simulation is used to study the behavior of nanoparticles in the presence of a surface and evaporation of solvent. The simulation is able to produce amorphous topologies of nanoparticles on the surface.  相似文献

Emotions are parts of organizational reality to an ever increasing extent. Importantly, they are not just tools in the hand of healthcare workers to achieve better physician / healthcare professional-to-patient interactions but intrinsic processes and characteristics with psychic, cognitive and somatic actions. For a thorough investigation of the issue, a PANAS-X questionnaire was used to examine the emotions of 187 physicians and other healthcare professionals, all engaged in oncology, in 2009. The research succeeded in exploring the overall emotional state oncology professionals had assumed in relation with their job as well as enabled the authors of this study to draw the respondents' emotional map and assess their fundamental emotional attitudes. Furthermore, the authors managed to identify groups of respondents that had felt more intense positive, and/or less intense negative emotions that are socially accepted than others. They included those of senior experienced oncologists, males, individuals with families, childless individuals, ward workers, and skilled professionals. According to the findings, the range of emotions an oncologist experiences / feels intently during his everyday work is dependent upon a great number of factors.  相似文献

Human serum transferrin has a potential for drug-delivery systems. Oxalate and aziridine-carboxylate was conjugated to serum transferrin in order to transport into the targeted cancer cells via transferrin-receptor mediated endocytosis. Capillary zone electrophoresis and capillary isoelectric focusing were used to analyze the effectiveness of complexation reactions. The electropherograms show the differences between iron-free- and iron-complexed molecular forms of human serum transferrin. The iron-complexed transferrin sample containing the different anions as synergistic complexing agents were characterized by different electrophoretic parameters.  相似文献

Polysaccharide films containing chitosan, methylcellulose, and a mixture of these polysaccharides in various ratios were prepared and modified with meso-tetrakis(4-sulfonatophenyl)porphyrin in an aqueous medium at pH 7. The modified films were compared with the initial films using spectroscopic methods and microscopic imaging. Electronic (UV-vis absorption, electronic circular dichroism (ECD)) and vibrational (FTIR and Raman) spectra showed that the porphyrin macrocycles had a strong affinity toward chitosan and did not interact with the methylcellulose. The total porphyrin uptake depended on the chitosan: methylcellulose ratio and pure methylcellulose films did not retain porphyrin macrocycles. ECD measurements detected the presence of optically active porphyrin species bound to the films. SEM and AFM images confirmed that the porphyrin macrocycles caused structural changes on the film surface and within the film layer.  相似文献
