Hormonal imprinting is transmitted from imprinted to virgin cells of the same cell line. No transmission of imprinting occurred between Chinese hamster ovary (CHO) and human Chang liver cells, and the presence of the latter reduced rather than enhanced the thyrotropic hormone (TSH) binding capacity of the CHO cells. While hormone binding capacity was relatively homogeneous in the control and the mixed cell cultures, it was not homogeneous in the homologous imprinted plus virgin cell population, indicating a continuous transmission of imprinting from the former to the latter. 相似文献

The recently synthesized isocyanonaphtalene derivatives ACAIN and CACAIN are fluorochromes excitable at wavelengths of around 366 nm and bind cysteine-rich proteins with hydrophobic motifs. We show that these compounds preferentially label tonoplasts in living Arabidopsis and tobacco (Nicotiana tabacum SR1) cells. ACAIN-labeled membranes co-localized with the GFP signal in plants expressing GFP-δ-TIP (TIP2;1) (a tonoplast aquaporin) fusion protein. ACAIN preserved the dynamics of vacuolar structures. tip2;1 and triple tip1;1-tip1;2-tip2;1 knockout mutants showed weaker ACAIN signal in tonoplasts. The fluorochrome is also suitable for the labeling and detection of specific (cysteine-rich, hydrophobic) proteins from crude cell protein extracts following SDS-PAGE and TIP mutants show altered labeling patterns; however, it appears that ACAIN labels a large variety of tonoplast proteins. ACAIN/CACAIN could be used for the detection of altered vacuolar organization induced by the heptapeptide natural toxin microcystin-LR (MCY-LR), a potent inhibitor of both type 1 and 2A protein phosphatases and a ROS inducer. As revealed both in plants with GFP-TIP2;1 fusions and in wild-type (Columbia) plants labeled with ACAIN/CACAIN, MCY-LR induces the formation of small vesicles, concomitantly with the absence of the large vegetative vacuoles characteristic for differentiated cells. TEM studies of MCY-LR-treated Arabidopsis cells proved the presence of multimembrane vesicles, with characteristics of lytic vacuoles or autophagosomes. Moreover, MCY-LR is a stronger inducer of small vesicle formation than okadaic acid (which inhibits preferentially PP2A) and tautomycin (which inhibits preferentially PP1). The analysis of contacts is a powerful tool to understand biomolecular function in a series of contexts, from the investigation of dynamical behavior at equilibrium to the study of nonequilibrium dynamics in which the system moves between multiple states. We thus propose a tool called CONtact ANalysis (CONAN) that, from molecular dynamics (MD) trajectories, analyzes interresidue contacts, creates videos of time-resolved contact maps, and performs correlation, principal component, and cluster analysis, revealing how specific contacts relate to functionally relevant states sampled by MD. We present how CONAN can identify features describing the dynamics of ubiquitin both at equilibrium and during mechanical unfolding. Additionally, we show the analysis of MD trajectories of an α-synuclein mutant peptide that undergoes an α-β conformational transition that can be easily monitored using CONAN, which identifies the multiple states that the peptide explores along its conformational dynamics. The high versatility and ease of use of the software make CONAN a tool that can significantly facilitate the understanding of the complex dynamical behavior of proteins or other biomolecules. L-alanine and its peptides (L-Ala-2–6) do not attract or repulse Tetrahymena in a 10–8M concentration. In 10–10M concentration there is a consistent repellent effect. Twenty four hours after L-alanine or L-alanine-peptides' pretreatment (imprinting) the progeny generation of the cells react differently to the same materials. L-Alanine, L-alanine penta- and hexapeptide in both concentrations are chemoattractant, while L-alanine tetrapeptide is repellent. L-Alanine dipeptide is inert in 10–10M and repellent at 10–8M concentrations, while L-alanine tripeptide is strongly repellent at 10–10M and attractant at 10–8M concentrations. This means, that the first encounter (imprinting) with an exogeneous amino acid or peptide is decisive to the later reaction of the protozoan cell. The chain length is important in the imprinting, however the reaction is not consistent. Insulin dimers deprived of biological activity by linking with suberic acid symmetrically in position B29 or B1 were not able to induce imprinting. Lack of N-terminal phenylalanine or even of five C-terminal amino acids did not interfere with imprinting, regardless of whether or not it was associated with an activity loss. It appears that while hormonal imprinting is closely associated with the hormone's ability to bind to the receptor, it may be related as well as unrelated to the hormone's biological activity. From the aspect of its mucopolysaccharide content the mast cell population is not homogeneous. The pulmonary and heart muscle mast cells of the rat are alcian blue positive, the mast cells of the thyroid gland, lymph nodes, subcutaneous connective tissue, mesentery and peripheral nerve are safranin positive, whereas among the mast cells of the peritoneal cavity and the thymus there are both alcian blue and saffranin positive forms. The least acid mucopolysaccharides are in the mast cells of the peritoneal fluid, the mesentery and the lungs, whereas the most acid ones are in the mast cells of the lymph nodes, the subcutaneous connective tissue and the thyroid gland. There is a considerable difference between the two last mentioned organs. The mast cells of the subcutaneous connective tissue are end-product cells without amine or precursor turnover, whereas the mast cells of the thyroid gland incorporate and deliver amines, which may participate in the regulation of the host gland. 相似文献

In adult rats the ouabain-sensitive ATP-ase activity of the heart muscle as well as the amount of specifically bound 3H-ouabain decreased by 1/3 as a result of a single triamcinolone treatment undertaken in the neonatal period. Neonatal ouabain treatment did not alter the triamcinolone binding of the thymus of adult animals. The experiments again call attention to the phenomenon that the steroid hormones, even though specific for their target cells, are able to induce "alien" imprinting when administered in the neonatal period. Newborn rats were treated once with 2 Units of vasopressin (VP) or oxytocin, respectively. At the age of 2 months and 1 year, respectively, the reactivity of isolated aortic strips to vasopressin and noradrenaline was tested. In young adults animals vasopressin pretreatment enhanced whilst oxytocin pretreatment decreased the responsiveness of aortic strips to VP. In elder specimens both types of pretreatment resulted in an increased late sensitivity to VP. The norepinephrine-sensitivity was higher in the young adults pretreated with oxytocin, whilst in the elder ones vasopressin-retreatment gave the same late effect. It could be established that the hormone excess produced in the neonatal age led to the "amplification" of hormone receptors and this alteration provided to be permanent. A compound which is similar to but not identical with the hormone--in our case, oxytocin--would elicit an alteration of a less unindirectional and permanent type; late sensitivity changes in both directions may occur. 相似文献
