Invasive species trigger biodiversity losses and alter ecosystem functioning, with life history shaping invasiveness (Sakai et al., Annu Rev Ecol Syst 32:305–332, 2001). However, pinpointing the relation of a specific life history to invasion success is difficult. One approach may be comparing congeners. The two Palearctic pavement ants, Tetramorium sp.E (widely known as T. caespitum, Schlick-Steiner et al., Mol Phylogenet Evol 40:259–273, 2006) and T. tsushimae have invaded North America (Steiner et al., Biol Invasions 8:117–123, 2006). Their life histories differ in that T. sp.E has separate single-queened colonies but T. tsushimae multi-queened colonies scattered over large areas (Sanada-Morimura et al., Insect Soc 53:141–148, 2006; Schlick-Steiner et al., Mol Phylogenet Evol 40:259–273, 2006; Steiner et al., Biol Invasions 8:117–123, 2006). Comparison of the genetic diversity in the entire native and non-native ranges will elucidate the invasion histories. Here, we present 13 and 11 microsatellites, developed for T. sp.E and T. tsushimae, respectively, and characterize all for both species. Florian M. Steiner, Wolfgang Arthofer and Birgit C. Mutualisms are interspecific interactions in which both players benefit. Explaining their maintenance is problematic, because cheaters should outcompete cooperative conspecifics, leading to mutualism instability. Monoecious figs (Ficus) are pollinated by host-specific wasps (Agaonidae), whose larvae gall ovules in their “fruits” (syconia). Female pollinating wasps oviposit directly into Ficus ovules from inside the receptive syconium. Across Ficus species, there is a widely documented segregation of pollinator galls in inner ovules and seeds in outer ovules. This pattern suggests that wasps avoid, or are prevented from ovipositing into, outer ovules, and this results in mutualism stability. However, the mechanisms preventing wasps from exploiting outer ovules remain unknown. We report that in Ficus rubiginosa, offspring in outer ovules are vulnerable to attack by parasitic wasps that oviposit from outside the syconium. Parasitism risk decreases towards the centre of the syconium, where inner ovules provide enemy-free space for pollinator offspring. We suggest that the resulting gradient in offspring viability is likely to contribute to selection on pollinators to avoid outer ovules, and by forcing wasps to focus on a subset of ovules, reduces their galling rates. This previously unidentified mechanism may therefore contribute to mutualism persistence independent of additional factors that invoke plant defences against pollinator oviposition, or physiological constraints on pollinators that prevent oviposition in all available ovules.  相似文献

The Polistine wasps include both independent‐founding species, with small, single‐queen colonies founded by one or a few potential queens, and swarm‐founding species, which have larger societies, many queens and initiate colonies as a swarm of queens and workers. Swarm‐founding evolved from independent‐founding and Ropalidia is the only genus with both types, making it an excellent model system for understanding this dramatic shift in colony organization. We have isolated 18 polymorphic microsatellite loci from three species of Ropalidia, including two independent‐founding and one swarm‐founding species. These loci will allow us to reconstruct colony social and genetic structures in this important genus.  相似文献

CDK4/6 inhibitors arrest the cell cycle in G1‐phase. They are approved to treat breast cancer and are also undergoing clinical trials against a range of other tumour types. To facilitate these efforts, it is important to understand why a cytostatic arrest in G1 causes long‐lasting effects on tumour growth. Here, we demonstrate that a prolonged G1 arrest following CDK4/6 inhibition downregulates replisome components and impairs origin licencing. Upon release from that arrest, many cells fail to complete DNA replication and exit the cell cycle in a p53‐dependent manner. If cells fail to withdraw from the cell cycle following DNA replication problems, they enter mitosis and missegregate chromosomes causing excessive DNA damage, which further limits their proliferative potential. These effects are observed in a range of tumour types, including breast cancer, implying that genotoxic stress is a common outcome of CDK4/6 inhibition. This unanticipated ability of CDK4/6 inhibitors to induce DNA damage now provides a rationale to better predict responsive tumour types and effective combination therapies, as demonstrated by the fact that CDK4/6 inhibition induces sensitivity to chemotherapeutics that also cause replication stress.  相似文献

Abstract  The red imported fire ant, Solenopsis invicta , a damaging invasive pest, was discovered in February 2001 in Brisbane, Australia at two sites, Fisherman Islands and suburban Richlands-Wacol. Using four microsatellite loci and the protein marker Gp-9 , we compared the two infestations with each other, and with potential source populations in North and South America to better understand the history of their introduction to Brisbane. Based on an analysis of molecular variance, as well as a maximum likelihood tree of colonies from the two Australian sites, we found that the two sites were genetically distinct and were almost certainly introduced separately. All of the colonies at Fisherman Islands were monogynous, headed by a single queen, while the Richlands-Wacol site had a mixture of single-queen monogynous and multiple-queen polygynous colonies. However, the monogynous and polygynous colonies at the Richlands-Wacol site were not genetically distinct from each other, and probably constitute a single, mixed introduction. Based on allele frequencies at the microsatellite loci, and Gp-9 , both Australian infestations were more similar to North American populations than to South American, though the Fisherman Islands infestation was intermediate, making it difficult to assign. Thus, there has been one introduction from either a North or South American monogynous population at Fisherman Islands, and one introduction from a mixed monogynous/polygynous North American population at Richlands-Wacol. These findings have implications for the control of the current infestations, as well as for the quarantine regulations necessary to prevent additional introductions to Australia.  相似文献

The effects of tributyltin (TBT) compounds on gill morphology were examined in the mummichog, Fundulus heteroclitus , in 96-h LC50 and 6-week sublethal exposures. Morphometry was used for the identification and quantification of effects with the light microscope. A 96-h LC50 of 17.2 μg 1 1 was determined. Morphometric analysis of gill tissues revealed hypertrophy of the lamellar epithelium in fish exposed to 17.2 μg 1–1. Relative diffusing capacity was significantly decreased (−41 %); ( P <0.05, ANOVA, Bonferroni t -test). At 35.6 μg 1−1, TBT exposure resulted in a significant reduction (− 40%) in the volume of the lamellar blood channels. Both of these observations occurred in fish that showed signs of acute poisoning including loss of equilibrium. In fish exposed to sublethal concentrations of 0.105–2.000 μg TBT 1−1 for 6 weeks, there were no pathological changes in the gill. There were no treatment-related changes in the surface morphology of the gills of fish from both experiments upon scanning electron microscopic examination. Although gill pathology was observed in acutely toxic exposures, it does not appear to be a major mechanism of TBT toxicity.  相似文献

Gibberellin biosynthesis pathways were investigated using isotopically-labelled C19- and C20-gibberellins and cell-free preparations from immature seed of Phaseous coccineus cv. Prizewinner. The initial steps in an early 13-hydroxylation pathway involved the conversion gibberellin A12-aldehyde (GA12-aldehyde) to GA12 which was 13-hydroxylated to yield GA53, Metabolism of GA53 yielded GA44. In contrast to other cell-free systems, GA44 was not further converted, either as a δ-lactone or an open-lactone structure, to the C-20 aldehyde GA19. GA19 was, however, metabolised to GA20, GA5 and GA1. GA20 represented a branch point in the pathway as it was converted both to GA1, which was an end product, and GA5 which was further converted to GA6. Like GA1, GA6 was also an end-product of the early 13-hydroxylation pathway.
A non-13-hydroxylation pathway involving GA4, GA15, GA24 GA37 and GA36 also originated from GA12. The terminal product of this pathway was the 3β-hydroxy C19-gibberellin, GA4.  相似文献

The rolB gene of the plant pathogen Agrobacterium rhizogenes has an important role in the establishment of hairy root disease in infected plant tissues. When expressed as a single gene in transgenic plants the RolB protein gives rise to effects indicative of increased auxin activity. It has been reported that the RolB product is a β-glucosidase and proposed that the physiological and developmental alterations in transgenic plants expressing the rolB gene are the result of this enzyme hydrolysing bound auxins, in particular (indole-3-acetyl)-β-D-glucoside (IAGluc), and thereby bringing about an increase in the intracellular concentration of indole-3-acetic acid (IAA). Using tobacco plants as a test system, this proposal has been investigated in detail. Comparisons have been made between the RolB phenotype and that of IaaM/iaaH transformed plants overproducing IAA. In addition, the levels of IAA and IAA amide and IAA ester conjugates were determined in wild-type and transgenic 35S-rolB tobacco plants and metabolic studies were carried out with [13C6]IAA [2′-14C]IAA, [14C]IAGluc, [5-3H]-2-o-(indole-3-acetyl)-myo-inositol and [14C]indole-3-acetylaspartic acid. The data obtained demonstrate that expression of the rolB encoded protein in transgenic tobacco does not produce a phenotype that resembles that of IAA over producing plants, does not alter the size of the free IAA pool, has no significant effect on the rate of IAA metabolism, and, by implication, appears not to influence the overall rate of IAA biosynthesis. Furthermore, the in vivo hydrolysis of IAGluc, and that of the other IAA conjugates that were tested, is not affected. On the basis of these findings, it is concluded that the RolB phenotype is not the consequence of an increase in the size of the free IAA pool mediated by an enhanced rate of hydrolysis of IAA conjugates.  相似文献

Floral onset in soybean (Glycine max cv. Ransom) is characterized by precocious initiation of axillary meristems in the axils of the most recently initiated leaf primordium. During floral transition, leaf morphology changes from trifoliolate leaf with stipules, to a three-lobed bract, to an unlobed bract. Soybean flowers initiated at 26/22 C day/night temperatures are normal, papilionaceous, and pentamerous. Sepal, petal, and stamen whorls are initiated unidirectionally from the abaxial to adaxial side of the floral apex. The median sepal is located abaxially and the median petal adaxially on the meristem. The organogeny of ‘Ransom’ flowers was found to be: sepals, petals, outer stamens plus carpel, inner stamens; or, sepals, petals, carpel, outer stamens, inner stamens. The outer stamen whorl and the carpel show possible overlap in time of initiation. Equalization of organ size occurs only within the stamen whorls. The sepals retain distinction in size, and the petals exhibit an inverse size to age relationship. The keel petals postgenitally fuse along part of their abaxial margins; their bases, however, remain free. Soybean flowers initiated at cool day/night temperatures of 18/14 C exhibited abnormalities and intermediate organs in all whorls. The gynoecium consisted of one to ten carpels (usually three or four), and carpel connation varied. Fusion of keel petals was often lacking, and stamen filaments fused erratically. Multiple carpellate flowers developed into multiple pods that were separate or variously connate. Intermediate type organs had characteristics only of organs in adjacent whorls. These aberrant flowers demonstrate that the floral meristem of soybean is not fixed or limited in its developmental capabilities and that it has the potential to produce alternate morphological patterns.  相似文献

High performance liquid chromatography has been used to measure the quantities of caffeine, theobromine, and theophylline in aqueous extracts of endosperm from immature and mature fruits of Coffea arabica and six other species of Coffea. Caffeine was the alkaloid present in largest amounts and, with one exception, in concentrations that were broadly similar in immature and mature fruit. The highest concentrations of caffeine were found in C. canephora at 35.1 and 24.5 mg g−1, respectively, in immature and mature endosperm. The lowest concentrations were in C. bengalensis, where caffeine was not detected in extracts from mature fruit. [8-3H]Caffeine was metabolised relatively slowly by immature endosperm of C. arabica and C. canephora. In contrast, C. dewevrei, C. eugenioides, C. stenophylla, C. salvatrix and C. bengalensis all appeared to metabolise [8-3H]caffeine much more rapidly, as the percentage recovery of the applied label was much lower and there was more extensive incorporation of radioactivity into theobromine, theophylline, 3-methylxanthine and two unidentified polar metabolites. The endogenous caffeine concentrations and the metabolism data indicate that there may be marked differences in the rate of turnover of caffeine in the various species of Coffea. Potential sources of material for the production of naturally decaffeinated coffee are discussed.  相似文献
