Infectious bursal disease virus (IBDV), a member of the Birnaviridae group, is a commercially important pathogen of chickens. From electron micrographs of frozen, hydrated, unstained specimens, we have computed a three-dimensional map of IBDV at about 2 nm resolution. The map shows that the structure of the virus is based on a T=13 lattice and that the subunits are predominantly trimer clustered. The subunits close to the fivefold symmetry axes are at a larger radius than those close to the two- or threefold axes, giving the capsid a markedly nonspherical shape. The trimer units on the outer surface protrude from a continuous shell of density. On the inner surface, the trimers appear as Y-shaped units, but the set of units surrounding the fivefold axes appears to be missing. It is likely that the outer trimers correspond to the protein VP2, carrying the dominant neutralizing epitope, and the inner trimers correspond to protein VP3, which has a basic carboxy-terminal tail expected to interact with the packaged RNA.  相似文献

Aim The physiological requirements and tolerances of a species partially determine both its habitat preferences within a community and its broader geographic range. Therefore, we predicted that local ecology should be correlated with geographic distribution. We tested for a correlation between local ecology and range size, and we attempted to account for this correlation by the climate of the range. Location Bishop Creek Watershed, on the eastern side of the Sierra Nevada, California. Methods We recorded all plant species growing in each of 263 plots in the montane to alpine zones of the watershed. The local habitat preferences of 282 species were described in terms of wetness, elevation, soil, and amount of shade. The size and centre of the geographic range for each species were determined from regional floras. Results Wetness preference within the watershed was significantly correlated with range size. Specifically, plants of wet sites had larger ranges that extend to the north, whereas plants of dry sites tended to have smaller ranges centred to the east. The correlation between local wetness preference and range size was entirely explained by the location of the range centre of the species. Main conclusions A possible reason for the large ranges of mesophilic plants in our study area is that mesic habitats are continuous throughout the western Cordillera, while dry alpine habitats are isolated by valleys to the east. The correspondence between local ecology and geographic distributions implies evolutionary stasis in the niches of these plant species.  相似文献

The coastal heathlands of the Royal National Park are impacted by both fire and herbivory by introduced deer, and to date these two factors have been dealt with independently in the management of natural areas. In recent years, there has been increasing recognition for a more integrated approach to manage these two disturbance agents. Fire and its role in Australian heathlands are well known, while impacts from introduced deer and the combined effects of fire and introduced deer are still poorly understood. In this study, we investigated the effects of fire and Javan rusa deer (Cervus timorensis) on both vegetation cover and floristics. The percentage cover of plants at different height layers and the presence/absence of individual species were recorded at sites representing two different burn histories (1993/1994 and 2000/2001) and deer presence or absence. Fire significantly reduced vegetation cover at low (<50 cm) and intermediate heights (50–100 cm), while deer presence affected grasses and sedges, and low vegetation at more recently burnt sites. Rusa deer also affected composition of the plant species assemblages, but no such effect was found for fire. Understanding the influence of each disturbance factor independently and together in the heathlands will be critical for implementing a more robust framework for future management.  相似文献

Background

Current international guidelines advocate achieving at least a 30 % reduction in maximum plantar pressure to reduce the risk of foot ulcers in people with diabetes. However, whether plantar pressures differ in cases with foot ulcers to controls without ulcers is not clear. The aim of this study was to assess if plantar pressures were higher in patients with active plantar diabetic foot ulcers (cases) compared to patients with diabetes without a foot ulcer history (diabetes controls) and people without diabetes or a foot ulcer history (healthy controls).

Methods

Twenty-one cases with diabetic foot ulcers, 69 diabetes controls and 56 healthy controls were recruited for this case-control study. Plantar pressures at ten sites on both feet and stance phase duration were measured using a pre-established protocol. Primary outcomes were mean peak plantar pressure, pressure-time integral and stance phase duration. Non-parametric analyses were used with Holm’s correction to correct for multiple testing. Binary logistic regression models were used to adjust outcomes for age, sex and body mass index. Median differences with 95 % confidence intervals and Cohen’s d values (standardised mean difference) were reported for all significant outcomes.

Results

The majority of ulcers were located on the plantar surface of the hallux and toes. When adjusted for age, sex and body mass index, the mean peak plantar pressure and pressure-time integral of toes and the mid-foot were significantly higher in cases compared to diabetes and healthy controls (p?<?0.05). The stance phase duration was also significantly higher in cases compared to both control groups (p?<?0.05). The main limitations of the study were the small number of cases studied and the inability to adjust analyses for multiple factors.

Conclusions

This study shows that plantar pressures are higher in cases with active diabetic foot ulcers despite having a longer stance phase duration which would be expected to lower plantar pressure. Whether plantar pressure changes can predict ulcer healing should be the focus of future research. These results highlight the importance of offloading feet during active ulceration in addition to before ulceration.

Australia’s native wild dog, the dingo (Canis dingo), is threatened by hybridization with feral or domestic dogs. In this study we provide the first comprehensive three dimensional geometric morphometric evaluation of cranial shape for dingoes, dogs and their hybrids. We introduce a novel framework to assess whether modularity facilitates, or constrains, cranial shape change in hybridization. Our results show that hybrid and pure dingo morphology overlaps greatly, meaning that hybrids cannot be reliably distinguished from dingoes on the basis of cranial metrics. We find that dingo morphology is resistant, with observed hybrids exhibiting morphology closer to the dingo than to the parent group dog. We also find that that hybridization with dog breeds does not push the dingo cranial morphology towards the wolf phenotype. Disparity and integration analyses on the ten recovered modules provided empirical support for modularity facilitating shape change over short evolutionary time scales. However, our results show that this is may not be the case in hybridization events, which were not influenced by module integration or disparity levels. We conclude that although hybridization events may introduce breed dog DNA to the dingo population, the native cranial morphology, and therefore likely the feeding eco-niche, of the dingo population is resistant to change. Our results have implications for conservation and management of dingoes and, more broadly, for the influence of integration patterns over ecological time scales in relation to selection pressure.  相似文献

Clathrin triskelions form polyhedral cages with hexagonal and pentagonal faces when dialyzed against suitable assembly buffers. However, when the buffer is made 12% saturated in ammonium sulfate and the dialysis is performed at 4 degrees C, clathrin polymerizes into cubes. The cube is constructed from eight triskelions with one at each corner. The edge length of the cube is approximately 45 nm, equivalent to the length of the leg of a triskelion. Thus, each edge of the cube is composed of two antiparallel legs overlapping over their whole length. The interactions between the legs in the cube are a subset of those postulated to occur in cages. Indeed, the cube can be derived from a pentagonal dodecahedron by removing 12 of the 20 triskelions with only slight adjustment of the legs of the remaining triskelions. The cube forms regular arrays and appears to be a favorable species for crystallization of clathrin.  相似文献

DNA was extracted from 12 clinical isolates of theMycobacterium avium-M. intracellulare-M. scrofulaceum serocomplex (including serovars 1, 2, 3, 4, 8, 42A, and 43), as well as from nine reference strains of other mycobacterial species. Rapid nitrogen decompression was used to weaken the cells for subsequent DNA extraction by a modified Marmur procedure. The DNA was hydrolyzed with phosphodiesterase, and the concentration of released deoxyribonucleotides was determined by reversed-phase high-performance liquid chromatography. The data were used to calculate the mole percentage guanine plus cytosine. Values ranged from 58.6 forM. leprae to 71.5 forM. avium serovar 3. The lowest value for theM. avium-M. intracellulare-M. scrofulaceum serocomplex was 66.3 forM. scrofulaceum serovar 42A. The data indicate that there is greater genetic variability for this group of organisms than is suggested by diagnostic clinical tests and seroagglutination reactions.  相似文献

We have observed ultrastructural features of muscle differentiation in the muscle lineage cells of cleavage-arrested whole embryos and partial embryos of ascidians. Whole embryos of Ciona intestinalis and Ascidia ceratodes were cleavage-arrested with cytochalasin B at the 8-cell stage and reared to an age equivalent to several hours after hatching; these embryos formed extensive myofilaments which were often further organized into myofibrils of different sizes and densities in the peripheral cytoplasm of the two muscle lineage blastomeres (B4.1 pair). Developing myofibrils in cleavage-arrested embryos resembled the muscle elements observed in normal hatched larvae, but were less uniformly organized. A similar development of myofilaments and myofibrils occurred in the muscle lineage cells of multicellular partial embryos reared to "hatching" age. These partial embryos resulted from the isolated muscle lineage pair (B4.1) of blastomeres of the 8-cell stage (Ciona and Ascidia), and from a muscle lineage blastomere pair (B5.2) isolated at the 16-cell stage (Ascidia). Muscle lineage cells in the partial embryos were readily identified by the dense aggregates of mitochondria in their cytoplasm. Taken together, these results from the two kinds of partial embryo effectively eliminate inductive interactions with embryonic tissues other than mesodermal as a necessary factor in the onset of self-differentiation in muscle lineage cells. The relative complexity of muscle phenotype expressed in cleavage-arrested and partial embryos attests to an unusually strong developmental autonomy in the ascidian muscle lineages. This autonomy lends further support to the theory that a localized and segregated egg cytoplasmic determinant is responsible for larval muscle development in ascidian embryos.  相似文献
