The mycorrhizal associations established between plants and fungi have multiple effects on plant growth, directly affecting stress tolerance. This work aimed to explore arbuscular mycorrhizal (AM) effects on carbon and nitrogen relationships of Aster tripolium L. and consequently on its flooding tolerance. Mycorrhizal and non-mycorrhizal juvenile plants were submitted to non-flooding and tidal flooding conditions for 56 d. Tidal flooding reduced biomass, but the presence of mycorrhiza had an ameliorating effect. The AM symbioses seem to have, like flooding, a stressful effect on A. tripolium at an early stage of plant development. However, once the plant was established, an improvement of growth performance of plants with mycorrhiza under flooding conditions was observed. The better tolerance of AM plants to flooding was mediated through an improvement of the osmotic adjustment of the plant tissues (higher concentrations of soluble sugars and proline) and through the increment of nitrogen acquisition in tidal-flooded plants.  相似文献

We evaluated a year-long treatment regime testing synthetic, 10-component, honey bee, Apis mellifera L. (Hymenoptera: Apidae), brood pheromone (SuperBoost; Contech Enterprises Inc., Delta, BC, Canada) on the productivity and vigor of package bee colonies in the lower Fraser Valley of British Columbia, Canada. Fifty-eight newlyestablished 1.3-kg (3-lb) colonies treated three times with SuperBoost at 5-wk intervals starting 30 April 2009 were compared with 52 untreated control colonies. Treated colonies produced 84.3% more honey than untreated control colonies. By 8 September 2009, SuperBoost-treated colonies had 35.4% more adults than untreated colonies. By 28 September, net survival of treated and control colonies was 72.4 and 67.3%, respectively. On 5 October, treated and control colonies were divided into two additional groups, making up four cohorts: SuperBoost-treated colonies treated again during fall and spring build-up feeding with pollen substitute diet (BeePro, Mann Lake Ltd., Hackensack, MN; TIT); controls that remained untreated throughout the year (CCC); colonies treated with SuperBoost in spring-summer 2009 but not treated thereafter (TCC); and original control colonies treated with SuperBoost during the fall and spring build-up feeding periods (CTT). There was no difference among cohorts in consumption of BeePro during fall feeding, but TTT colonies (including daughter colonies split off from parent colonies) consumed 50.8% more diet than CCC colonies during spring build-up feeding. By 21 April, the normalized percentages of the original number of colonies remaining (dead colonies partially offset by splits) were as follows: CCC, 31.4%; CTT, 43.8%; TCC, 53.59%; and TTT, 80.0%. The net benefit of placing 100 newly established package bee colonies on a year-long six-treatment regime with SuperBoost would be US$6,202 (US$62.02 per colony). We conclude that treatment with SuperBoost enhanced the productivity and survival of package bee colonies and hypothesize that similar results could be achieved with established colonies.  相似文献

The codling moth (Cydia pomonella (L.)) is an invasive pest of pome fruits introduced to the Americas in the 19–20th centuries. This pest is widespread on both sides of the Andes range separating Argentina and Chile. We performed an analysis of the population genetic variability and structure of Cpomonella in Argentina and Chile using 13 microsatellite markers and sampled C. pomonella from apple as the main host plant along its distribution area (approx. 1,800 km). A total of 22 locations (11 from Chile and 11 from Argentina) were sampled. Significant genetic differentiation was observed among samples from Argentina and Chile (FSC = 0.045) and between all localities (FST = 0.085). Significant isolation by distance (IBD) was found for each country and when samples from both sides of the Andes range were pooled, although a lower correlation coefficient was observed. The Mantel test showed that the geographic distance and highest altitude of the mountains between locations were significantly associated with the pairwise FST when samples from both sides of the Andes range were pooled. According to a Bayesian assignment test (STRUCTURE), samples from Argentina and Chile conformed to two distinct genetic clusters. Our results also suggest that the recent invasion of Cpomonella in the southernmost localities (Aysén Region in Chile and Santa Cruz Province in Argentina) originated in populations from the respective sides of the Andes range. Our results indicate a genetic exchange of Cpomonella within each country and significant genetic differentiation between countries, which could be explained by dispersal mediated by human activities related to fruit production within each country with little exchange between them. A possible explanation is that the Andes range could be a significant barrier for dispersal by flight, and quarantine barriers could prevent the movement of plant material or infested fruit between countries.  相似文献

Stress granules (SGs) are dynamic condensates associated with protein misfolding diseases. They sequester stalled mRNAs and signaling factors, such as the mTORC1 subunit raptor, suggesting that SGs coordinate cell growth during and after stress. However, the molecular mechanisms linking SG dynamics and signaling remain undefined. We report that the chaperone Hsp90 is required for SG dissolution. Hsp90 binds and stabilizes the dual‐specificity tyrosine‐phosphorylation‐regulated kinase 3 (DYRK3) in the cytosol. Upon Hsp90 inhibition, DYRK3 dissociates from Hsp90 and becomes inactive. Inactive DYRK3 is subjected to two different fates: it either partitions into SGs, where it is protected from irreversible aggregation, or it is degraded. In the presence of Hsp90, DYRK3 is active and promotes SG disassembly, restoring mTORC1 signaling and translation. Thus, Hsp90 links stress adaptation and cell growth by regulating the activity of a key kinase involved in condensate disassembly and translation restoration.  相似文献

Telomere attrition has been proposed as a biomarker and causal factor in aging. In addition to causing cellular senescence and apoptosis, telomere shortening has been found to affect gene expression in subtelomeric regions. Here, we analyzed the distribution of age‐related differentially expressed genes from the GTEx RNA sequencing database of 54 tissue types from 979 human subjects and found significantly more upregulated than downregulated genes in subtelomeric regions as compared to the genome‐wide average. Our data demonstrate spatial relationships between telomeres and gene expression in aging.  相似文献

Human gait analysis is often conducted in clinical and basic research, but many common approaches (e.g., three-dimensional motion capture, wearables) are expensive, immobile, data-limited, and require expertise. Recent advances in video-based pose estimation suggest potential for gait analysis using two-dimensional video collected from readily accessible devices (e.g., smartphones). To date, several studies have extracted features of human gait using markerless pose estimation. However, we currently lack evaluation of video-based approaches using a dataset of human gait for a wide range of gait parameters on a stride-by-stride basis and a workflow for performing gait analysis from video. Here, we compared spatiotemporal and sagittal kinematic gait parameters measured with OpenPose (open-source video-based human pose estimation) against simultaneously recorded three-dimensional motion capture from overground walking of healthy adults. When assessing all individual steps in the walking bouts, we observed mean absolute errors between motion capture and OpenPose of 0.02 s for temporal gait parameters (i.e., step time, stance time, swing time and double support time) and 0.049 m for step lengths. Accuracy improved when spatiotemporal gait parameters were calculated as individual participant mean values: mean absolute error was 0.01 s for temporal gait parameters and 0.018 m for step lengths. The greatest difference in gait speed between motion capture and OpenPose was less than 0.10 m s−1. Mean absolute error of sagittal plane hip, knee and ankle angles between motion capture and OpenPose were 4.0°, 5.6° and 7.4°. Our analysis workflow is freely available, involves minimal user input, and does not require prior gait analysis expertise. Finally, we offer suggestions and considerations for future applications of pose estimation for human gait analysis.  相似文献

Alzheimer's disease (AD) is characterized by cognitive disorders and alterations of behavioral traits such as anhedonia and anxiety. Contribution of nonphysiological forms of amyloid and tau peptides to the onset of neurological dysfunctions remains unclear because most preclinical models only present one of those pathological AD-related biomarkers. A more recently developed model, the TgF344-AD rat has the advantage of overexpressing amyloid and naturally developing tauopathy, thus making it close to human familial forms of AD. We showed the presence of a learning dysfunction in a reference memory test, without spatial working memory impairment but with an increase in anxiety levels and a decrease in motivation to participate in the test. In the sucrose preference test, TgF344-AD rats did not show signs of anhedonia but did not increase the volume of liquid consumed when the water was replaced by sucrose solution. These behavioral phenomena were observed at an age when tau accumulation are absent, and where amyloid deposits are predominant in the hippocampus and the entorhinal cortex. Within the hippocampus itself, amyloid accumulation is heterogenous between the subiculum, the dorsal hippocampus and the ventral hippocampus. Thus, our data demonstrated heterogeneity in the appearance of various behavioral and neurochemical markers in the TgF344-AD rat. This multivariate analysis will therefore make it possible to define the stage of the pathology, to measure its evolution and the effects of future therapeutic treatments.  相似文献
