In the last years, we have developed a method for the study of random RNA sequences with the aim of investigating their capacity to assume folded and possibly functionally active (ribozymes) structures. The RNA Foster assay developed in our laboratory is a powerful, simple, and fast method to investigate the structural properties of RNA by assessing the presence of secondary domains and their thermal stability through the combination of S1 nuclease activity at different temperatures. In this work, we investigated the structural properties of totally de novo random RNAs, 97‐nucleotides‐long, of which 60 nucleotides were completely random. The rationale behind this was to assess whether and to what extent random RNAs would acquire a stable fold as a prerequisite for catalytic activity. A library of random sequences was created, and 18 sequences were randomly selected and analyzed. Surprisingly, most of the RNAs were resistant to S1 nuclease digestion at up to 50°, and two sequences were resistant even at 70°, suggesting that these totally random RNA molecules could posses a stable secondary structure over a broad range of temperatures under the conditions used. We discuss the possible relevance of these data for the general scenario of the RNA world.  相似文献

The aim of the present study was to evaluate the expression of innate immunity receptors belonging to the Toll-like family in the neural plexuses of the different tracts of murine intestine, of the human ileum, and in lower dorsal root ganglia (DRGs) from where extrinsic afferents to these plexuses originate. Results obtained by immunohistochemistry and immunofluorescence on paraffin-embedded tissue and whole-mount preparations show that Toll-like receptors (TLRs) -3 and -7, recognizing viral RNA, and TLR4, recognizing lipopolysaccharide (membrane component of Gram-negative bacteria), are expressed in the myenteric and submucous plexuses of murine intestine and human ileum, and in DRGs primary sensory neurons. They also show that TLR4 immunostaining is stronger in murine distal large bowel. In murine tissue, expression of TLRs was present in both neurons and glial cells. These observations indicate that the enteric neural network might be directly activated by bacterial and viral components and is therefore more in the forefront than previously envisaged in defense responses of the intestinal wall and in the cross-talk with intestinal microbiota. They also highlight the presence of a peripheral neural network that by way of hardwired neurotransmission could potentially convey to the central nervous system specific information on our microbial counterpart and invading or potentially invading pathogens. The vibrational circular dichroism (VCD) spectra of dicarvone ( 1 ), dipinocarvone ( 2 ), and dimenthol ( 3 ) have been recorded in the range 900–3200 cm−1, encompassing the mid-infrared (mid-IR), the CO stretching, and the CH-stretching regions. For compound 3 also, the fundamental and the first overtone OH stretching regions have been investigated by IR/NIR absorption and VCD. Density functional theory (DFT) calculations allow one to interpret the IR and VCD spectra and to confirm the configuration/conformational studies previously conducted by X-ray diffraction. The most intense VCD signals are associated with the vibrational normal modes involving symmetry-related groups close to the CC bond connecting covalently the two molecular units. The vibrational exciton (VCDEC) model is fruitfully tested on the VCD data of compounds 1 and 2 for the spectroscopic regions at ~1700 cm−1, and the local mode model is tested on compound 3 at ~3500 and ~6500 cm−1. For compounds 1 and 2 also, ECD spectra are reported, and the exciton mechanism is tested also there, and connections to the VCDEC model are examined.  相似文献

The mating behavior of Mahanarva spectabilis (Distant) was studied under laboratory conditions. Virgin adults were individually maintained in transparent cages for observation of the mating sequence. Mating behavior of 23 couples was monitored during the photophase portion of the day. Copulation was observed in 17 of the 23 pairs. The mating sequence includes male approaching the female, mounting of the female, and copulation. Of the 17 mating pairs, 70.6 % mated on the second day after emergence. Most of the copulations began between 8 and 9 h after the onset of photophase. The average duration of copulation was 268?±?24.9 min, and most pairs mated once or twice during their lifetimes. The mating behavior of M. spectabilis is important to understand because it determines the age and ideal time for further behavioral testing, which is essential for determining the cues involved in the communication system of the species.  相似文献

Butler’s idea of evolution was developed over the publication of four books, several articles and essays between 1863 and 1890. These publications, although never achieving the success expected by Butler, proposed a psychological elaboration of evolution (robustly enforced by Lamarck’s philosophy), called ‘unconscious memory’. This was strongly in contrast with the materialistic approach suggested by Darwin’s natural selection. Starting with a historical introduction, this paper aspires to ascertain the logic, meaning and significance of Butler’s idea of ‘unconscious memory’ in the post-Darwinian physiological and psychological Pan-European discussion. Particular attention is devoted to demonstrating that Butler was not only a populariser of science but also an active protagonist in the late Victorian psychological debate.  相似文献

From a pharmacological point of view, organoseleniums are compounds with important and interesting antioxidant and biological activities. The aim of this study was to evaluate the hepatoprotective effect of bis(4‐methylbenzoyl) diselenide (BMD) against carbon tetrachloride (CCl4)–induced oxidative damage in mice. The animals received BMD (25 mg/kg p.o., for 3 days), and after 1 day, CCl4 (1 mg/kg body weight) was administered by intraperitoneal route. One day after the CCl4 exposure, the animals were euthanized for biochemical and histological analysis. Treatment with BMD (25 mg/kg p.o.) protected against aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, gamma‐glutamyl transferase and lactate dehydrogenase activity increases induced by CCl4 plasma exposure. Treatment with BMD (25 mg/kg) protected against increases in thiobarbituric reactive species and decreasing non‐protein thiols and ascorbic acid levels in liver of mice. Catalase and superoxide dismutase activity inhibition in the liver caused by CCl4 were protected by treatment with BMD (25 mg/kg). Glutathione S‐transferase activity was inhibited by CCl4 and remained unaltered even after treatment with BMD. Sections of liver from CCl4‐exposed mice presented an intense infiltration of inflammatory cells and loss of the cellular architecture. BMD (25 mg/kg) attenuated CCl4‐induced hepatic histological alterations.
