Several oncogenes and tumor-suppressor genes are involved either as early or late event in thyroid gland carcinogenesis. Human FHIT (fragile histidine triad) gene is highly conserved gene whose loss of function may be important in the development and/or progression of various types of cancer. We undertook this study to analyze FHIT and p53 gene status in different benignant and malignant thyroid tumors. Status of these genes as well as intensity of apoptosis was analyzed in tumor tissues by molecular genetic methods, immunohistochemistry, and FACS-scan analysis. The majority of the malignant thyroid cancers displayed aberrant expression of FHIT gene, concominant with p53 gene inactivation. This is followed by low rate of apoptosis, which may be important in the development and/or progression of thyroid cancer. We found higher incidence of p53 mutation and aberrant processing of FHIT mRNA in malignant tumors (papillary, follicular, medullary and anaplastic carcinomas) and in those tumors with distant metastasis. The growth of p53(-)/FHIT(-) follicular carcinoma of human origin was much faster in nude mice than p53(+)/FHIT(+) follicular carcinoma, and mice had shorter survival rate. Our results show a correlation between aberrant FHIT and p53 expression, low rate of apoptosis, and malignancy. Concomitant aberration of FHIT gene and p53 could be responsible for development of highly malignant types of thyroid cancer and may be considered as a prognostic marker for these tumors.  相似文献

The red seaweeds Gelidiella acerosa and Gracilaria mammillaris growing along the coast of Venezuela were investigated as potential economic sources of agar. Agar extracted from Gracilaria mammillaris accounted for 27% of the algal dry weight but had quite a poor gelling ability because of the presence of alkali-stable sulphate groups on the D-galactose residue. However, the gel strength of its aqueous solutions was considerably enhanced by the addition of potassium, sodium and calcium ions (up to 1N). On the contrary, the galactan from Gelidiella acerosa (yield of about 20% w/w) exhibited quite good properties. In particular, the gel strength was comparable to that of commercial agaroses. Sulphate esters were not detectable by chemical methods and NMR spectroscopy revealed an agarose backbone with a high degree of methylation on both D and L-galactose residues.  相似文献

The taxonomic status of the pheasant superspecies Lophura leucomelanos and Lophura nycthemera has been unclear since 1948. Molecular techniques provided the opportunity to clarify the situation. Using sequences of mitochondrial DNA (800 nucleotides from the D-loop, plus 400 from the cyt b ) from 49 specimens belonging to 10 subspecies (plus two outgroups), we constructed a phylogeny of the subspecies of L. nycthemera and L. leucomelanos . Our data support the monophyly of both species. L. l. lineata and L. l. crawfurdi belong to L. leucomelanos and not to L. nycthemera (suggested by other authors). Our data also confirm a northern locality of origin (Central Buthan) for L. l. moffitti , and have clarified the relationships between subspecies within each species: there are three groups within L. leucomelanos and two within L. nycthemera .  相似文献

Matrix-assisted laser desorption/ionization (MALDI) imaging of proteolytic peptides from formalin-fixed paraffin embedded (FFPE) tissue sections could be integrated in the portfolio of molecular pathologists for protein localization and tissue classification. However, protein identification can be very tedious using MALDI-time-of-flight (TOF) and post-source decay (PSD)-based fragmentation. Hereby, we implemented an R package and Shiny app to exploit liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based proteomic biomarker discovery data for more specific identification of peaks observed in bottom-up MALDI imaging data. Abstract This survey evaluates the antibody band patterns of sera taken from clinically defined cases of Lyme borreliosis, towards three locally isolated strains of Borrelia burgdorferi , belonging to the three species: Borrelia sensu stricto, Borrelia garinii and Borrelia afzelii , by means of Western blot. The sera were taken from patients resident in a limited area of Friuli Venezia Giulia (FVG) region. The data indicated that, besides a different feature of the band reactivity which correlated to the different stages of Lyme borreliosis, there was a preferential reactivity to the species Borrelia afzelii and Borrelia garinii . An immunodominant band at 51 kDa, corresponding to a protein visible in the electrophoretic profile of strain BL3 ( B. afzelii ), behaved like a marker of an early infection, because it was present exclusively in the sera of patient with ECM. The overall findings would indicate that B. afzelii and B. garinii are the prevalent genospecies in the FVG area, even if strains belonging to B. sensu stricto have been also isolated in this area. Consequently strains representative of these two species must be used as antigens in Western blot.  相似文献

Ecological interactions often vary geographically. Work in salt marshes on the Atlantic Coast of the United States has documented community-wide latitudinal gradients in plant palatability and plant traits that may be driven in part by greater herbivore pressure at low latitudes. To determine if similar patterns exist elsewhere, we studied six taxa of saltmarsh plants ( Atriplex , Juncus , Limonium , Salicornia , Spartina and Suaeda ) at European sites at high (Germany and the Netherlands) and low (Portugal and Spain) latitudes. We conducted feeding assays using both native and non-native consumers, and documented patterns of herbivore damage in the field. As in the United States, high-latitude plants tended to be more palatable than low-latitude plants when offered to consumers in paired feeding assays in the laboratory, although assays with grasshopper consumers were less consistent than those with crab consumers, and plants in the field at low-latitude sites tended to experience greater levels of herbivore pressure than plants at high-latitude sites. Similarly, high-latitude leaf litter was more palatable than litter from low-latitude plants when offered to consumers in paired feeding assays in the laboratory. Latitudinal gradients in plant palatability and herbivore pressure may be a general phenomenon, and may contribute to latitudinal gradients in decomposition processes.  相似文献

The degradation of 2-chlorophenol vapours in air was performed in a trickling biofilter packed with ceramic material seeded with the bacterium Pseudomonas pickettii, strain LD1. The system performance was evaluated under varying operating conditions (inlet 2-chlorophenol air concentrations from 0.10 to 3.50 g m?3, and superficial air velocities of 30.0, 60.0, and 120.0 m h?1). For all air velocity the maximum degradation rate was obtained for loading rates of 40 g m?2 h?1. Higher loading conditions resulted in strong inhibition of microbial activity, particularly severe at high air velocity. Process analysis, performed using data on pollutant concentration profiles along the filter packing obtained under different conditions of inlet concentration and air velocity, proves that best performance (i.e. maximum degradation efficiency and capacity) can be obtained for a narrow range of operating conditions, which can be ensured by proper design of biofilter size (i.e. diameter and height). Kinetic analysis of experimental data confirms that 2-CP inhibits microbial activity in the biofilter bed. Experimental data are satisfactorily fitted by the Haldane kinetic equation up to a critical value of loading rate, beyond which the experimental degradation rate is overestimated by the kinetic model. The inhibition appears to be affected by the loading rate, and the estimated inhibition constant linearly increases with increasing empty bed residence time.  相似文献

Bovine spongiform encephalopathy (BSE), the prion disease in cattle, was widely believed to be caused by only one strain, BSE-C. BSE-C causes the fatal prion disease named new variant Creutzfeldt-Jacob disease in humans. Two atypical BSE strains, bovine amyloidotic spongiform encephalopathy (BASE, also named BSE-L) and BSE-H, have been discovered in several countries since 2004; their transmissibility and phenotypes in humans are unknown. We investigated the infectivity and human phenotype of BASE strains by inoculating transgenic (Tg) mice expressing the human prion protein with brain homogenates from two BASE strain-infected cattle. Sixty percent of the inoculated Tg mice became infected after 20 to 22 months of incubation, a transmission rate higher than those reported for BSE-C. A quarter of BASE strain-infected Tg mice, but none of the Tg mice infected with prions causing a sporadic human prion disease, showed the presence of pathogenic prion protein isoforms in the spleen, indicating that the BASE prion is intrinsically lymphotropic. The pathological prion protein isoforms in BASE strain-infected humanized Tg mouse brains are different from those from the original cattle BASE or sporadic human prion disease. Minimal brain spongiosis and long incubation times are observed for the BASE strain-infected Tg mice. These results suggest that in humans, the BASE strain is a more virulent BSE strain and likely lymphotropic.  相似文献

We evaluated the Plasmodium vivax polymorphism by studying the Pvmsp-3 gene's polymorphic region by PCR-RFLP in 55 samples from patients living in Tierralta, Colombia. Three different sizes of the Pvmsp-3 gene were found, type A (1,900 bp), type B (1,500 bp) and type C (1,100 bp); most of the samples were type A (96.4 %). The Pvmsp-3 gene exhibited high polymorphism. Seven restriction patterns were found when using Alu I, and nine were found with Hha I; 12 different alleles were obtained when these patterns were combined. The findings suggest that this gene could be used in Colombia as a molecular epidemiologic marker for genotyping P. vivax.  相似文献
