以蒙古高原岩黄芪属植物为对象, 应用徐克学的最大同步法, 探讨了蒙古高原岩黄芪属(豆科)植物的系统演化, 并根据分支分类结果对蒙古高原岩黄芪属进行了系统学处理。作者首次将蒙古高原岩黄芪属分为岩黄芪亚属、半灌木岩黄芪亚属(新拟)和无刺岩黄芪组、丛枝岩黄芪组、无茎岩黄芪组、半灌木岩黄芪组等4 个组。本文对蒙古高原岩黄芪组的划分符合苏联植物志(1945)中的观点。  相似文献

Hydrostatic pressures in the range of hundreds of atmospheres are known to disrupt cytoskeletal organization in tissue culture cells, with profound changes in cell shape. The molecular mechanisms of these effects are poorly understood. To determine the effect of pressure on the cytoskeleton, and thus to provide better indicators of the molecular mechanisms, we used fluorescent antibody staining to compare the organizations of seven different cytoskeletal proteins in HeLa cells and rat osteosarcoma cells (ROS-17/2.8) subjected to different pressures up to 400 atm. Pressures of 300 atm or more caused cells of both lines to “round up” and to withdraw their lamellar extensions. However, this response varied within a population of cells, with some cells remaining spread at pressures that caused their neighbors to round up. The most resistant to rounding were those cells touching other cells, and the occasional giant cells. As expected, the rounded cells showed disruption of actin stress fibers and of vinculin and talin at focal contacts. The unrounded cells showed less disruption in the organization of these same proteins. Microtubules and myosin II filaments appeared resistant to 400 atm pressure in both cell types, whether rounded or unrounded. However, in HeLa cells, the intermediate filaments, vimentin and cytokeratin, depolymerized and formed small vesicles when pressures exceeded 200 atm, and this occurred in rounded as well as unrounded cells. In osteosarcoma cells, which do not have cytokeratin, vimentin did not depolymerize. We discuss different mechanisms that might explain these responses to pressure, including direct effects on the equilibria of protein polymerization and less direct effects on regulatory mechanisms, such as phosphorylation pathways, that control cytoskeletal organization. The latter type of explanation seems more consistent with both the variability of response within cell populations and the difference in vimentin's response in one cell line compared with the other.  相似文献

The potassium content of single human red cells was measured with an electron probe. Cells were placed on beryllium discs and coated with a thin layer of dibutyl pthalate to prevent loss of cellular contents. Samples were stable under the electron beam during analysis for more than 15 minutes and could be stored for long periods of time. Primary standards were prepared by loading red cells with varying known amounts of potassium in order to circumvent the corrections for absorption. The X-ray intensity was found to be directly proportional to the potassium content of the cells.  相似文献

Tail fin disks removed from tadpoles of Rana pipiens that were immersed in thyroxine or triiodothyronine for 3 days (referred to as donors) were fused to premetamorphic tail fin blocks that had never been exposed to this hormone (referred to as recipients) so that triplets were formed, consisting of one recipient tail block sandwiched between two donor tail fin blocks. Such recipient tail blocks responded with characteristic resorptive activity within 24 or 48 hr, instead of the minimum 72-hr latent period normally intervening in donor blocks, until shrinkage was initiated in response to triiodothyronine (T3) or tetraiodothyronine (T4). The presence of T3 or T4 hormone was not required continuously throughout the latent period. Hormone could be withdrawn after 30 hr contact in vivo and after 24 hr contact in vitro without interfering with the rate of tissue regression of tadpole tail fins, suggesting that the “latent period” probably does not coincide with the “critical period” during which subtle biochemical changes presumably occur that precede regression of the tadpole tail during metamorphosis. It is suggested that during the latent period active intermediates may be synthesized that are subsequently transferred from donor tail fins to recipients, thus reducing the latent period of the latter.  相似文献

【背景】宁夏贺兰山东麓葡萄产区忽视有机肥的施用,果树枝条焚烧污染环境,造成土壤养分缺失,土壤质量下降。【目的】为解决长期施用化肥对土壤造成的一系列问题,通过大田试验研究施肥及喷施不同浓度菌剂对土壤理化性质、真菌群落组成及多样性的影响,为酿酒葡萄可持续健康发展提供科学依据。【方法】以‘赤霞珠’葡萄根际土壤为试验对象,采用Illumina MiSeq高通量测序技术,测定并分析根际土壤理化性质、真菌群落组成和多样性在7个处理[常规施肥(CK)、蚯蚓粪+腐熟枝条+100倍菌剂(T1)、蚯蚓粪+腐熟枝条+200倍菌剂(T2)、蚯蚓粪+腐熟枝条+300倍菌剂(T3)、蚯蚓粪+未腐熟枝条+100倍菌剂(A1)、蚯蚓粪+未腐熟枝条+200倍菌剂(A2)和蚯蚓粪+未腐熟枝条+300倍菌剂(A3)]的变化。【结果】相较于CK,葡萄根际土壤理化性质差异明显,施肥处理增加了土壤有机质含量,土壤pH含量无明显变化,改良了土壤结构,活化了土壤有效养分。相较于CK,各处理真菌分类操作单元(operational taxonomic unit, OTU)数均降低,A2处理根际土壤丰富度及多样性均显著增加。真菌群落组成...  相似文献

在扫描电镜下观察了豆科黄华属Thermopsis 18种植物种子的表面纹饰,发现 T.alpina,T.bar- bata,T.inflata,T.lupinoides,T. licentiana,T.smithiana和T.turkestanica的种子表面为粗网状,T californica,T.divaricarpa,T. macrophylla,T.mollis的种子表面为细网状,T.gracilis,T.montana,T. fabacea的种子表面为相对平滑型纹饰,T.alterniflora的种子表面为不规则条形,T.chinensis的种子表 面为粘膜状,T.rhombifolia的种子表面为条形及 T.viuosa的种子表面为碎屑状纹饰。结果表明黄华属的种子表面特征对属下类群的划分有一定意义,对澄清某些混乱的种有一定价值。  相似文献

Pioglitazone (PIO) is a peroxisome proliferator-activated receptor-γ (PPARγ) agonist in clinical use for treatment of type 2 diabetes (T2DM). Accumulating evidence suggests PPARγ agonists may be useful for treating or delaying the onset of Alzheimer’s disease (AD), possibly via actions on mitochondria, and that dose strengths lower than those clinically used for T2DM may be efficacious. Our major objective was to determine if low doses of pioglitazone, administered orally, impacted brain activity. We measured blood-oxygenation-level dependent (BOLD) low-frequency fluctuations in conscious rats to map changes in brain resting-state functional connectivity due to daily, oral dosing with low-dose PIO. The connectivity in two neural circuits exhibited significant changes compared with vehicle after two days of treatment with PIO at 0.08 mg/kg/day. After 7 days of treatment with a range of PIO dose-strengths, connections between 17 pairs of brain regions were significantly affected. Functional connectivity with the CA1 region of the hippocampus, a region that is involved in memory and is affected early in the progression of AD, was specifically investigated in a seed-based analysis. This approach revealed that the spatial pattern of CA1 connectivity was consistent among all dose groups at baseline, prior to treatment with PIO, and in the control group imaged on day 7. Compared to baseline and controls, increased connectivity to CA1 was observed regionally in the hypothalamus and ventral thalamus in all PIO-treated groups, but was least pronounced in the group treated with the highest dose of PIO. These data support our hypothesis that PIO modulates neuronal and/or cerebrovascular function at dose strengths significantly lower than those used to treat T2DM and therefore may be a useful therapy for neurodegenerative diseases including AD.  相似文献

遗传和营养因素都能影响猪肉的品质。但是, 到目前为止同时研究遗传和营养因素对肉质影响的报道很少。在本研究中, 136头PIC5系杂交猪, 体重65 kg, 被随机分成4组, 各组分别给予不同日粮。在饲养35 d、体重大约90 kg时统一屠宰并且进行肉质测定、H-FABP基因分型及其与肉质性状的关联分析。结果表明: (1)所采用的3种日粮对肉色、屠宰后24 h的pH、肌内脂肪和肌肉蛋白含量有极显著的影响; (2)H-FABP基因型对肌内脂肪和肌肉蛋白含量存在极显著的影响; (3)H-FABP基因多态性和营养因素的交互作用对pH 和肌内脂肪含量均有显著的影响, 对照组的AA基因型具有最高pH值, 高维生素E组的AA基因型具有最高肌内脂肪值。实验结果提示在关于猪肉质的育种和生产过程中应该同时考虑营养因素和遗传因素。  相似文献
