Background

Obesity invokes a range of metabolic disturbances, but the transition from a poor to excessive nutritional environment may exacerbate adult metabolic dysfunction. The current study investigated global maternal nutrient restriction during early or late gestation on glucose tolerance and insulin sensitivity in the adult offspring when lean and obese.

Methods/Principal Findings

Pregnant sheep received adequate (1.0M; CE, n = 6) or energy restricted (0.7M) diet during early (1–65 days; LEE, n = 6) or late (65–128 days; LEL, n = 7) gestation (term ∼147 days). Subsequent offspring remained on pasture until 1.5 years when all received glucose and insulin tolerance tests (GTT & ITT) and body composition determination by dual energy x-ray absorptiometry (DXA). All animals were then exposed to an obesogenic environment for 6–7 months and all protocols repeated. Prenatal dietary treatment had no effect on birth weight or on metabolic endpoints when animals were ‘lean’ (1.5 years). Obesity revealed generalised metabolic ‘inflexibility’ and insulin resistance; characterised by blunted excursions of plasma NEFA and increased insulinAUC (from 133 to 341 [s.e.d. 26] ng.ml−1.120 mins) during a GTT, respectively. For LEL vs. CE, the peak in plasma insulin when obese was greater (7.8 vs. 4.7 [s.e.d. 1.1] ng.ml−1) and was exacerbated by offspring sex (i.e. 9.8 vs. 4.4 [s.e.d. 1.16] ng.ml−1; LEL male vs. CE male, respectively). Acquisition of obesity also significantly influenced the plasma lipid and protein profile to suggest, overall, greater net lipogenesis and reduced protein metabolism.

Conclusions

This study indicates generalised metabolic dysfunction with adult-onset obesity which also exacerbates and ‘reveals’ programming of glucose-insulin sensitivity in male offspring prenatally exposed to maternal undernutrition during late gestation. Taken together, the data suggest that metabolic function appears little compromised in young prenatally ‘programmed’ animals so long as weight is adequately controlled. Nutritional excess in adulthood exacerbates any programmed phenotype, indicating greater vigilance over weight control is required for those individuals exposed to nutritional thrift during gestation.  相似文献

Climate change scenarios predict a doubling of the atmospheric CO2 concentration by the end of this century. Yet, how rising CO2 will affect the species composition of aquatic microbial communities is still largely an open question. In this study, we develop a resource competition model to investigate competition for dissolved inorganic carbon in dense algal blooms. The model predicts how dynamic changes in carbon chemistry, pH and light conditions during bloom development feed back on competing phytoplankton species. We test the model predictions in chemostat experiments with monocultures and mixtures of a toxic and non-toxic strain of the freshwater cyanobacterium Microcystis aeruginosa. The toxic strain was able to reduce dissolved CO2 to lower concentrations than the non-toxic strain, and became dominant in competition at low CO2 levels. Conversely, the non-toxic strain could grow at lower light levels, and became dominant in competition at high CO2 levels but low light availability. The model captured the observed reversal in competitive dominance, and was quantitatively in good agreement with the results of the competition experiments. To assess whether microcystins might have a role in this reversal of competitive dominance, we performed further competition experiments with the wild-type strain M. aeruginosa PCC 7806 and its mcyB mutant impaired in microcystin production. The microcystin-producing wild type had a strong selective advantage at low CO2 levels but not at high CO2 levels. Our results thus demonstrate both in theory and experiment that rising CO2 levels can alter the community composition and toxicity of harmful algal blooms.  相似文献

The hyperaccumulator Thlaspi caerulescens was grown with Hordeum vulgare and Lepidium heterophyllum in a split pot experiment to examine the effect of rhizosphere interactions on metal uptake. The objective was to assess the viability of such intercropping as either (1) a system of ‘phytoprotection’ for nonaccumulating plants or (2) a means of enhancing phytoextraction with large-biomass crops through increased metal mobilization within the shared rhizosphere. The plants were separated by (1) an impermeable barrier, (2) a permeable root barrier, or (3) no physical barrier to allow different degrees of root interaction. Studies of rhizosphere effects using split pot experiments are subject to considerable uncertainty by the need to relate test results to appropriate control plants. This was resolved by comparing plant metal concentrations to ‘equivalent’ control plants, with the same yield, based on the observed variation in metal concentration with yield under similar growing conditions.

Cadmium concentration in H. vulgare was increased by a factor of 2.4 when it was grown alongside T. caerulescens without a barrier. In contrast, the uptake of zinc by H. vulgare was significantly decreased, probably through metal depletion within the zone of the Zn-hyperaccumulator's rhizosphere. T. caerulescens also apparently increased the concentration of Cd in H. vulgare by a factor of 1.4 when the roots of the two plants were separated by a permeable barrier that allowed movement of soil solution but prevented physical mixing of roots. The concentrations of all the metals studied (Cd, Zn, Cu, Pb, Ni) were greater in T. caerulescens when the hyperaccumulator was grown alongside either L. heterophyllum or H. vulgare without a root barrier — probably through successful exploitation of a greater volume of soil. However, this effect was not seen in the presence of a partial barrier, except in the case of Cu when T. caerulescens was grown alongside H. vulgare.

These results suggest that T. caerulescens may alter conditions in shared rhizospheres and thereby affect the availability of selected metals to neighboring plants. Thus, it is possible that under-sowing some plants with small hyperaccumulators may potentially offer an alternative form of management for marginally contaminated soils. There was limited evidence of an intercropped hyperaccumulator mobilizing selected metals and restricting the availability of others. However, changes in uptake of selected metals by the larger plant may be quite small compared with the requirements of crop protection or the short-term requirements of many land remediation programs.  相似文献

Tempo and mode of concerted evolution in the L1 repeat family of mice   总被引:10,自引:0,他引:10  
A 300-bp DNA sequence has been determined for 30 (10 from each of three species of mice) random isolates of a subset of the long interspersed repeat family L1. From these data we conclude that members of the L1 family are evolving in concert at the DNA sequence level in Mus domesticus, Mus caroli, and Mus platythrix. The mechanism responsible for this phenomenon may be either duplicative transposition, gene conversion, or a combination of the two. The amount of intraspecies divergence averages 4.4%, although between species base substitutions accumulate at the rate of approximately 0.85%/Myr to a maximum divergence of 9.1% between M. platythrix and both M. domesticus and M. caroli. Parsimony analysis reveals that the M. platythrix L1 family has evolved into a distinct clade in the 10-12 Myr since M. platythrix last shared a common ancestor with M. domesticus and M. caroli. The parsimony tree also provides a means to derive the average half-life of L1 sequences in the genome. The rates of gain and loss of individual copies of L1 were estimated to be approximately equal, such that approximately one-half of them turn over every 3.3 Myr.   相似文献

The genus Dasylirion is a group of plants typically present in the Chihuahuan Desert, perennial, with a dioecious sexual behavior and commonly called sotoles. This genus has been little studied from the biological point of view, and the bases of its reproductive response remain unknown. In this work we studied the chromosome number and meiotic response of Dasylirion cedrosanum in the county of Saltillo, Coahuila, located at the North East of Mexico. For the preparation of mitotic chromosomes, we used a technique based on enzymatic treatment with pectolyase and cellulase, as well as staining with acetocarmin dye. For the study of meiosis, male flower buds were collected, fixed and stained for analysis with the same dye. As a result, the gametic (n = x = 19) and somatic chromosome (2n = 38) numbers of D. cedrosanum are reported for the first time, being consistent with previous findings in other Dasylirion species, which points to a constant ploidy level across the genus. Variation was observed in the morphology and size of the somatic chromosomes, with types ranging from submetacentric to subtelocentric, and sizes oscillating in a range of 4.43 µm, with an average total length of 112.38 µm for the diploid chromosome complement. This shows that the chromosome complement of D. cedrosanom would belong to a 3B classification of Stebins, with a medium variation between chromosome lengths and low chromosome asymmetry. This variation indicates the feasibility of constructing a chromosome ideotype for this species. The meiotic chromosome pairing showed a chromosome behavior consistent with a disomic inheritance characteristic of a diploid species, with prevalence of ring and chain bivalents, typically without pairing abnormalities. Bivalent configurations in all cases were symmetrical.The normal and symmetrical meiotic pairing indicates a balanced production of gametes, and suggests the absence of heteromorphic sex determination.  相似文献

We investigated the effects of thymoquinone (TQ) on the expression of liver microRNAs (miRNAs), liver histopathology and oxidative stress in Ehrlich acid solid tumor model induced mice. We used 24 male BALB/c mice divided randomly into three groups. Control (C) group mice were injected intraperitoneally (i.p.) with 0.5 ml saline for four weeks. Tumor (T) group mice were injected i.p. with 0.5 ml saline for four weeks, then Ehrlich acid tumor cells were injected subcutaneously into the neck to induce solid tumor formation. TQ (T + Tq) group mice injected i.p. with 10 mg/kg TQ for four weeks, then Ehrlich acid tumor cells were injected subcutaneously into the neck of the mice in this group to induce solid tumor formation. At the end of the study, liver from all groups were removed for histopathological and miRNAs analysis, and oxidative stress measurement. We found that the expression of miR-206b-3p was up-regulated and the oxidative stress and necrosis increased in the liver tissue of mice with Ehrlich acid solid tumor. TQ application decreased the oxidative stress, prevented necrosis, increased regeneration and down-regulated the expression of miR-206b-3p in the liver tissue.  相似文献

Modifications occurring on autoantigens during cell death have been proposed to have a role in the initiation of autoimmune diseases. Patients suffering from mixed connective tissue disease (MCTD) produce autoantibodies directed to U1 small nuclear ribonucleoprotein (snRNP), and antibodies against a 70 kDa protein component, the U1-70K (70K) protein, are the most prominent. During apoptosis, 70K is cleaved by caspase-3 to a 40 kDa product, which remains associated with the complex. Autoantibodies preferentially recognizing the apoptotic form of 70K have been described previously, and an apoptosis-specific epitope on 70K has been identified. This study shows that 29 of 53 (54%) MCTD sera preferentially recognize the apoptotic form of 70K over intact 70K. Moreover, we show that antibodies directed to an apoptosis-specific epitope on 70K are more specifically associated with MCTD than other anti-70K antibodies, suggesting that apoptotic 70K is a better antigen for the detection of these antibodies in MCTD patients. Longitudinal analysis of 12 MCTD patients showed in several patients that early sera are relatively enriched with antibodies recognizing an apoptosis-specific epitope, and that the levels of these apoptosis-specific antibodies decrease in time. These findings indicate that the early detection of apoptotic 70K is of considerable interest for anti-U1 snRNP-positive patients.  相似文献

This study examined the impact of season-long exposure to elevated carbon dioxide (CO2) and ozone (O3), individually and in combination, on leaf chlorophyll content and gas exchange characteristics in potato (Solanum tuberosum L. cv. Bintje). Plants grown in open-top chambers were exposed to three CO2 (ambient, 550 and 680 μmol mol-1) and two O3 treatments (ambient and elevated; 25 and 65 nmol mol-1, 8 h day-1 means, respectively) between crop emergence and maturity; plants were also grown in unchambered field plots. Non-destructive measurements of chlorophyll content and visible foliar injury were made for all treatments at 2-week intervals between 43 and 95 days after emergence. Gas exchange measurements were made for all except the intermediate 550 μmol mol-1 CO2 treatment. Season-long exposure to elevated O3 under ambient CO2 reduced chlorophyll content and induced extensive visible foliar damage, but had little effect on net assimilation rate or stomatal conductance. Elevated CO2 had no significant effect on chlorophyll content, but greatly reduced the damaging impact of O3 on chlorophyll content and visible foliar damage. Light-saturated assimilation rates for leaves grown under elevated CO2 were consistently lower when measured under either elevated or ambient CO2 than in equivalent leaves grown under ambient CO2. Analysis of CO2 response curves revealed that CO2-saturated assimilation rate, maximum rates of carboxylation and electron transport and respiration decreased with time. CO2-saturated assimilation rate was reduced by elevated O3 during the early stages of the season, while respiration was significantly greater under elevated CO2 as the crop approached maturity. The physiological origins of these responses and their implications for the performance of potato in a changing climate are discussed.  相似文献
