Synchronous spiking during cerebellar tasks has been observed across Purkinje cells: however, little is known about the intrinsic cellular mechanisms responsible for its initiation, cessation and stability. The Phase Response Curve (PRC), a simple input-output characterization of single cells, can provide insights into individual and collective properties of neurons and networks, by quantifying the impact of an infinitesimal depolarizing current pulse on the time of occurrence of subsequent action potentials, while a neuron is firing tonically. Recently, the PRC theory applied to cerebellar Purkinje cells revealed that these behave as phase-independent integrators at low firing rates, and switch to a phase-dependent mode at high rates. Given the implications for computation and information processing in the cerebellum and the possible role of synchrony in the communication with its post-synaptic targets, we further explored the firing rate dependency of the PRC in Purkinje cells. We isolated key factors for the experimental estimation of the PRC and developed a closed-loop approach to reliably compute the PRC across diverse firing rates in the same cell. Our results show unambiguously that the PRC of individual Purkinje cells is firing rate dependent and that it smoothly transitions from phase independent integrator to a phase dependent mode. Using computational models we show that neither channel noise nor a realistic cell morphology are responsible for the rate dependent shift in the phase response curve.  相似文献

Changes in coastline configuration and sand movements on Portuguese Island, a small island lying close to Inhaca Island 35 km east of Maputo, Mozambique have resulted in large areas of the Island being eroded away and consequent restricted tidal recharge of the mangrove. Measurements made during 1989 show that flooding of the mangrove occurred on only three occasions. Percentage mortality of the dominant species,Ceriops tagal (Perr.) C. B. Robinson., is high. However, at the time of the study, the co-dominant,Bruguiera gymnorrhiza (L.) Lam., showed no signs of stress. Halophytic conditions are indicated by an invasive spread of salt-tolerant herbs. All species of crustaceans and molluscs have disappeared from the mangrove.Although mortality ofC. tagal is high, the regeneration status of this species is also high, comparable to that in a regularly recharged mangrove on Inhaca Island. It is suggested thatC. tagal propagules were shed under conditions when seedling establishment was favorable i.e. flooding. These seedlings may represent the last cohort which may also suffer stress and death unless tidal recharge is re-established.  相似文献

Most molecular analyses, including phylogenetic inference, are based on sequence alignments. We present an algorithm that estimates relatedness between biomolecules without the requirement of sequence alignment by using a protein frequency matrix that is reduced by singular value decomposition (SVD), in a latent semantic index information retrieval system. Two databases were used: one with 832 proteins from 13 mitochondrial gene families and another composed of 1000 sequences from nine types of proteins retrieved from GenBank. Firstly, 208 sequences from the first database and 200 from the second were randomly selected and compared using edit distance between each pair of sequences and respective cosines and Euclidean distances from SVD. Correlation between cosine and edit distance was -0.32 (P < 0.01) and between Euclidean distance and edit distance was +0.70 (P < 0.01). In order to check the ability of SVD in classifying sequences according to their categories, we used a sample of 202 sequences from the 13 gene families as queries (test set), and the other proteins (630) were used to generate the frequency matrix (training set). The classification algorithm applies a voting scheme based on the five most similar sequences with each query. With a 3-peptide frequency matrix, all 202 queries were correctly classified (accuracy = 100%). This algorithm is very attractive, because sequence alignments are neither generated nor required. In order to achieve results similar to those obtained with edit distance analysis, we recommend that Euclidean distance be used as a similarity measure for protein sequences in latent semantic indexing methods.  相似文献

Background  

Parkinson's disease (PD) is the second most common neurodegenerative disorder. One of the most widely used techniques to diagnose PD is a Single Photon Emission Computer Tomography (SPECT) scan to visualise the integrity of the dopaminergic pathways in the brain. Despite this there remains some discussion on the value of SPECT in the differential diagnosis of PD. We did a meta-analysis of all the existing literature on the diagnostic accuracy of both pre- and post-synaptic SPECT imaging in the differential diagnosis of PD.  相似文献

Astroviruses (AstVs) are positive sense, single-stranded RNA viruses transmitted to a wide range of hosts via the fecal-oral route. The number of AstV-infected animal hosts has rapidly expanded in recent years with many more likely to be discovered because of the advances in viral surveillance and next generation sequencing. Yet no study to date has identified human AstV genotypes in animals, although diverse AstV genotypes similar to animal-origin viruses have been found in children with diarrhea and in one instance of encephalitis. Here we provide important new evidence that non-human primates (NHP) can harbor a wide variety of mammalian and avian AstV genotypes, including those only associated with human infection. Serological analyses confirmed that >25% of the NHP tested had antibodies to human AstVs. Further, we identified a recombinant AstV with parental relationships to known human AstVs. Phylogenetic analysis suggests AstVs in NHP are on average evolutionarily much closer to AstVs from other animals than are AstVs from bats, a frequently proposed reservoir. Our studies not only demonstrate that human astroviruses can be detected in NHP but also suggest that NHP are unique in their ability to support diverse AstV genotypes, further challenging the paradigm that astrovirus infection is species-specific.  相似文献

Freshwater lakes represent large methane sources that, in contrast to the Ocean, significantly contribute to non-anthropogenic methane emissions to the atmosphere. Particularly mixed lakes are major methane emitters, while permanently and seasonally stratified lakes with anoxic bottom waters are often characterized by strongly reduced methane emissions. The causes for this reduced methane flux from anoxic lake waters are not fully understood. Here we identified the microorganisms and processes responsible for the near complete consumption of methane in the anoxic waters of a permanently stratified lake, Lago di Cadagno. Interestingly, known anaerobic methanotrophs could not be detected in these waters. Instead, we found abundant gamma-proteobacterial aerobic methane-oxidizing bacteria active in the anoxic waters. In vitro incubations revealed that, among all the tested potential electron acceptors, only the addition of oxygen enhanced the rates of methane oxidation. An equally pronounced stimulation was also observed when the anoxic water samples were incubated in the light. Our combined results from molecular, biogeochemical and single-cell analyses indicate that methane removal at the anoxic chemocline of Lago di Cadagno is due to true aerobic oxidation of methane fuelled by in situ oxygen production by photosynthetic algae. A similar mechanism could be active in seasonally stratified lakes and marine basins such as the Black Sea, where light penetrates to the anoxic chemocline. Given the widespread occurrence of seasonally stratified anoxic lakes, aerobic methane oxidation coupled to oxygenic photosynthesis might have an important but so far neglected role in methane emissions from lakes.  相似文献
