The packing of lipopolysaccharide aggregates from rough strains of Escherichia coli was examined at different pH values. Lipopolysaccharide head-group motion, measured with an electron spin resonance probe, was found to be dependent on pH, and indicated the existence of multiple ionizable groups. Lipopolysaccharide from a rough (Ra) and a heptose-less (Re) mutant were more rigid at pH 5 than at pH 10.5. In addition, head-group mobility of the magnesium salt of Ra lipopolysaccharide was substantially less than that of the sodium salt at pH 7.0, whereas at high pH (pH 12) the two salts were equally fluid. Changes in head-group packing were also reflected in pH-dependent changes in the phase transition measured with differential scanning calorimetry. The enthalpy of the transition, delta Ht, for the sodium salt of Re lipopolysaccharide was greatest at pH 7.5 and approached zero in both the acidic and the basic pH ranges. We propose that fixed charges in the core and lipid A regions significantly influence lipopolysaccharide head-group motion and the lipopolysaccharide aggregation state. Furthermore, ionic bridging among phosphate groups dramatically rigidifies head group interactions in the neutral to acidic pH ranges.  相似文献

Embryonic development of the mouse superior cervical ganglion (SCG) is defined in vivo and in vitro using morphologic, morphometric, and biochemical approaches. Catecholamine fluorescence was present in the SCG on Day 14 of gestation and underwent characteristic changes in distribution among neurons between this time and adulthood. During prenatal ontogeny, choline acetyltransferase (ChAc) activity increased 2-fold, while tyrosine hydroxylase (T-OH) activity rose 30-fold and total protein increased 4-fold. Ganglionic explants from 14-day embryos extended neurites and exhibited specific biochemical development in medium without added nerve growth factor (NGF). However, the addition of NGF further stimulated neuronal development: Ganglia exhibited significant increases in ChAc and T-OH activities and in total protein compared to controls grown in medium without added NGF. The presence of target submandibular gland radically altered development of T-OH activity in cultured sympathetic ganglia. By 5 days in culture, ganglia grown with target tissue, even in the presence of anti-NGF, exhibited a 10- to 15-fold increase in T-OH activity compared to zero-time controls, and a 2-fold increase over ganglia grown alone or with nontarget tissue. Ganglia grown with target salivary glands showed a correspondingly greater elaboration and directionality of nerve fiber outgrowth, even in the presence of anti-NGF.  相似文献

Receiver operating characteristic (ROC) curve analysis is a simple and effective means to compare the accuracies of indicator variables of bacterial beach water quality. The indicator variables examined in this study were previous day's Enterococcus density and antecedent rainfall at 24, 48, and 96 h. Daily Enterococcus densities and 15-min rainfall values were collected during a 5-year (1996 to 2000) study of four Boston Harbor beaches. The indicator variables were assessed for their ability to correctly classify water as suitable or unsuitable for swimming at a maximum threshold Enterococcus density of 104 CFU/100 ml. Sensitivity and specificity values were determined for each unique previous day's Enterococcus density and antecedent rainfall volume and used to construct ROC curves. The area under the ROC curve was used to compare the accuracies of the indicator variables. Twenty-four-hour antecedent rainfall classified elevated Enterococcus densities more accurately than previous day's Enterococcus density (P = 0.079). An empirically derived threshold for 48-h antecedent rainfall, corresponding to a sensitivity of 0.75, was determined from the 1996 to 2000 data and evaluated to ascertain if the threshold would produce a 0.75 sensitivity with independent water quality data collected in 2001 from the same beaches.  相似文献

Mitosis requires precise control of microtubule dynamics. The KinI kinesin MCAK, a microtubule depolymerase, is critical for this regulation. In a screen to discover previously uncharacterized microtubule-associated proteins, we identified ICIS, a protein that stimulates MCAK activity in vitro. Consistent with this biochemical property, blocking ICIS function in Xenopus extracts with antibodies caused excessive microtubule growth and inhibited spindle formation. Prior to anaphase, ICIS localized in an MCAK-dependent manner to inner centromeres, the chromosomal region located in between sister kinetochores. From Xenopus extracts, ICIS coimmunoprecipitated MCAK and the inner centromere proteins INCENP and Aurora B, which are thought to promote chromosome biorientation. By immunoelectron microscopy, we found that ICIS is present on the surface of inner centromeres, placing it in an ideal location to depolymerize microtubules associated laterally with inner centromeres. At inner centromeres, MCAK-ICIS may destabilize these microtubules and provide a mechanism that prevents kinetochore-microtubule attachment errors.  相似文献

Salmonids undergo a developmental transition from parr to smoltthat involves a number of physiological and morphological changes.In recent years, my laboratory has studied shifts in red musclefunction at this parr-smolt transformation (PST) in rainbowtrout, Oncorhynchus mykiss. Parr red muscle has faster contractionkinetics than smolts, including faster rates of activation andrelaxation and a faster maximum shortening velocity. At PST,a transition in swimming behavior is also observed, with lowertailbeat frequencies and longer EMG duty cycles in the oldersmolts. Lastly, there is molecular correlate to changes in kineticsand behavior. During PST, there is a developmental reductionin the number of myosin heavy chain (MHC) isoforms in the redmuscle of rainbow trout. Since MHC composition of muscle candetermine contractile properties, these molecular results suggesta mechanism for the transition in red muscle kinetics and steadyswimming. The red muscle of parr is more likely to contain thefast-twitch or white isoform of MHC, resulting in faster contractileproperties of that muscle and higher tailbeat frequencies duringsteady swimming. Lastly, experimental work supports the conclusionthat the shift in kinetics causes the observed shift in swimmingbehavior.  相似文献

Can labels suggestively influence sensory perceptions and taste? Using a “ Phantom Ingredient” taste test, we show that the presence or absence of a labeled ingredient (soy) and the presence or absence of a health claim negatively bias taste perceptions toward a food erroneously thought to contain soy. We found a label highlighting soy content made health claims believable but negatively influenced perceptions of taste for certain segments of consumers. Our results and discussion provide better direction for researchers who work with ingredient labeling as well as for those who work with soybean products.  相似文献

Membranes from dormant and heat-activated spores were labelled with the fatty acid spin probe 5-doxyl stearate and analyzed using electron spin resonance spectroscopy. Membranes from dormant spores were slightly less fluid above 23° than membranes from heat-activated spores. Also L-proline caused a much larger increase in the upper transition temperature than did D-proline when added to membranes from heat-activated spores. Thus a compound known to trigger germination in this strain may interact stereospecifically to alter the biophysical properties of the spore membranes.  相似文献

We examined the spatial and temporal control of actin assembly in living Xenopus eggs. Within minutes of egg activation, dynamic actin-rich comet tails appeared on a subset of cytoplasmic vesicles that were enriched in protein kinase C (PKC), causing the vesicles to move through the cytoplasm. Actin comet tail formation in vivo was stimulated by the PKC activator phorbol myristate acetate (PMA), and this process could be reconstituted in a cell-free system. We used this system to define the characteristics that distinguish vesicles associated with actin comet tails from other vesicles in the extract. We found that the protein, N-WASP, was recruited to the surface of every vesicle associated with an actin comet tail, suggesting that vesicle movement results from actin assembly nucleated by the Arp2/3 complex, the immediate downstream target of N-WASP. The motile vesicles accumulated the dye acridine orange, a marker for endosomes and lysosomes. Furthermore, vesicles associated with actin comet tails had the morphological features of multivesicular endosomes as revealed by electron microscopy. Endosomes and lysosomes from mammalian cells preferentially nucleated actin assembly and moved in the Xenopus egg extract system. These results define endosomes and lysosomes as recruitment sites for the actin nucleation machinery and demonstrate that actin assembly contributes to organelle movement. Conversely, by nucleating actin assembly, intracellular membranes may contribute to the dynamic organization of the actin cytoskeleton.  相似文献
