The Cape Gannet Morus capensis is one of several seabird species endemic to the Benguela upwelling ecosystem (BUS) but whose population has recently decreased, leading to an unfavourable IUCN Red List assessment. Application of ‘JARA’ (‘Just Another Red-List Assessment,’ a Bayesian state-space tool used for IUCN Red List assessments) to updated information on the areas occupied by Cape Gannets and the nest densities of breeding birds at their six colonies, suggested that the species should be classified as Vulnerable. However, the rate of decrease of Cape Gannets in their most-recent generation exceeded that of the previous generation, primarily as a result of large decreases at Bird Island, Lambert’s Bay, and Malgas Island, off South Africa’s west coast (the western part of their range). Since the 1960s, there has been an ongoing redistribution of the species from northwest to southeast around southern Africa, and ~70% of the population now occurs on the south coast of South Africa, at Bird Island in Algoa Bay, on the eastern border of the BUS. Recruitment rather than adult survival may be limiting the present population; however, information on the seabird’s demographic parameters and mortality in fisheries is lacking for colonies in the northern part of the BUS. Presently, major threats to Cape Gannet include: substantially decreased availability of their preferred prey in the west; heavy mortalities of eggs, chicks and fledglings at and around colonies, inflicted by Cape Fur Seals Arctocephalus pusillus and other seabirds; substantial disturbance at colonies caused by Cape Fur Seals attacking adult gannets ashore; oiling; and disease. 相似文献

The red and pink aerobic muscle fibers are used to power steady swimming in fishes. We examined red and pink muscle recruitment and function during swimming in scup, Stenotomus chrysops, through electromyography and high-speed ciné. Computer analysis of electromyograms (EMGs) allowed determination of initial speed of muscle recruitment and duty cycle and phase of muscle electromyographic activity for both fiber types. This analysis was carried out for three longitudinal positions over a range of swimming speeds. Fiber type and longitudinal position both affected swimming speed of initial recruitment. Posterior muscle is recruited at the lowest swimming speed, whereas more anterior muscle is not initially recruited until higher speeds. At more anterior positions, the initial recruitment of pink muscle occurs at a higher swimming speed than the recruitment of red muscle. The duty cycle of pink muscle EMG activity is significantly shorter than that of red muscle, reflecting a difference in the onset time of activation during each cycle of length change: pink muscle onset time follows that of red. The different patterns of usage of red and pink muscle reflect differences in their contraction kinetics. Because pink muscle generates force more rapidly than red muscle, it can be activated later in each tailbeat cycle. Pink muscle is used to augment red muscle power production at higher swimming speeds, allowing a higher aerobically based steady swimming speed than that possible by red muscle alone. 相似文献

BACKGROUND: The regulated assembly of microtubules is essential for bipolar spindle formation. Depending on cell type, microtubules nucleate through two different pathways: centrosome-driven or chromatin-driven. The chromatin-driven pathway dominates in cells lacking centrosomes. RESULTS: Human RHAMM (receptor for hyaluronic-acid-mediated motility) was originally implicated in hyaluronic-acid-induced motility but has since been shown to associate with centrosomes and play a role in astral spindle pole integrity in mitotic systems. We have identified the Xenopus ortholog of human RHAMM as a microtubule-associated protein that plays a role in focusing spindle poles and is essential for efficient microtubule nucleation during spindle assembly without centrosomes. XRHAMM associates both with gamma-TuRC, a complex required for microtubule nucleation and with TPX2, a protein required for microtubule nucleation and spindle pole organization. CONCLUSIONS: XRHAMM facilitates Ran-dependent, chromatin-driven nucleation in a process that may require coordinate activation of TPX2 and gamma-TuRC. 相似文献

The shutter region of serpins consists of a number of highly conserved residues that are critical for both stability and function. Several variants of antithrombin with substitutions in this region are unstable and predispose the carrier to thrombosis. Although most mutations in the shutter region investigated to date are deleterious with respect to serpin stability and function, the substitution of Phe51 by Leu in alpha(1)-antitrypsin results in enhanced stability. Here, we have investigated the effects of introducing an analogous mutation into antithrombin (Phe 77 to Leu). The mutation did not affect the kinetics of interaction with proteases. Strikingly, however, the thermostability of the protein was markedly decreased, with the serpin displaying a 13 degrees C decrease in melting temperature as compared to wild-type recombinant antithrombin. Further studies revealed that in contrast to wild-type antithrombin, the mutant adopted the latent (inactive) conformation upon mild heating. Previous studies on shutter region mutations that destabilize antithrombin revealed that such variants possess enhanced affinity for both heparin pentasaccharide and full-length heparin. The N135A/F77L mutant had unchanged affinity for heparin pentasaccharide, but the affinity for full-length heparin was increased. We suggest that the Phe77Leu mutation causes conformational changes around the top of the D-helix in antithrombin, in particular, to the arginine 132 and 133 residues that may mediate additional antithrombin/heparin interactions. This paper also demonstrates that there are major differences between the shutter regions of antithrombin and alpha(1)-antitrypsin since a stabilizing mutation in antitrypsin has the converse effect in antithrombin. 相似文献

Of five strains of Aureobasidium pullulans studied, NRRL Y-2311-1 yielded the highest titer (26.2 g/L) of pullulan and formed the lowest amount of melanin-like pigment. Sucrose was superior to glucose as the carbon and energy source on the basis of yield and titer of pullulan produced. Pullulan titer was higher (26.2 vs 5.1 g/L), biomass concentration was lower (6.9 vs 12.7 g/L), and DO was lower (0 vs 60% of saturation) when the fermenter was agitated by a marine propeller compared to Rushton impellers. Pullulan produced by strain NRRL Y-2311-1 ranged in weight-average molar mass (M(w)) from 486 KDa and number-average molar mass (M(n)) from 220 Da on day 1 of growth to 390 KDa and 690 Da on day 6; M(w) declined by about 35% from day 1 to day 3, the day of maximum pullulan titer. A tensegrity structure composed of six struts interconnected with 24 elastic cables is used as a quantitative model of the steady-state elastic response of cells, with the struts and cables representing microtubules and actin filaments, respectively. The model is stretched uniaxially and the Young's modulus (E0) is obtained from the initial slope of the stress versus strain curve of an equivalent continuum. It is found that E0 is directly proportional to the pre-existing tension in the cables (or compression in the struts) and inversely proportional to the cable (or strut) length square. This relationship is used to predict the upper and lower bounds of E0 of cells, assuming that the cable tension equals the yield force of actin (approximately 400 pN) for the upper bound, and that the strut compression equals the critical buckling force of microtubules for the lower bound. The cable (or strut) length is determined from the assumption that model dimensions match the diameter of probes used in standard mechanical tests on cells. Predicted values are compared to reported data for the Young's modulus of various cells. If the probe diameter is greater than or equal to 3 microns, these data are closer to the lower bound than to the upper bound. This, in turn, suggests that microtubules of the CSK carry initial compression that exceeds their critical buckling force (order of 10(0)-10(1) pN), but is much smaller than the yield force of actin. If the probe diameter is less than or equal to 2 microns, experimental data fall outside the region defined by the upper and lower bounds. 相似文献

Boron (B) is widely distributed in surface and groundwaters predominantly as undissociated boric acid, and is found in ocean waters at a mean level of approx 4.6 mg B/L. A recent analysis of US surface waters indicated that the 10th, 50th, and 90th percentile B levels were 0.010, 0.076, and 0.387 mg B/L, respectively. The same study found that the 50th and 90th percentile B levels in California drinking waters were 0.10 and 0.40 mg B/L, respectively. The overall mean B concentration in Canadian surface waters in 1988 was reported to be 0.16 mg B/L, with a few stations reporting concentrations above 2.0 mg B/L. Unusually high levels of B have been measured in human water supplies of northern Chile, with concentrations ranging from 0.31 to 15.2 mg B/L. River water sources of B in the UK and Northern Italy were found to range from 0.002 to 0.87 mg B/L, respectively, whereas German drinking waters had median and maximum B levels of 0.02 and 0.18 mg B/L, respectively. Bottled mineral water also represents a source of B exposure based on the largest reported survey of US and European products, with a mean level of 0.75 mg B/L and a range from <0.005 to 4.35 mg B/L. Thus, B intake from drinking water is highly variable and dependent on the geographic source, the quantities of water consumed and the water sources used to bottle other beverages. 相似文献

Zebrafish (Danio rerio) embryos have proven to be a powerful model for studying a variety of developmental and disease processes. External development and optical transparency make these embryos especially amenable to microscopy, and numerous transgenic lines that label specific cell types with fluorescent proteins are available, making the zebrafish embryo an ideal system for visualizing the interaction of vascular, hematopoietic, and other cell types during injury and repair in vivo. Forward and reverse genetics in zebrafish are well developed, and pharmacological manipulation is possible. We describe a mechanical vascular injury model using micromanipulation techniques that exploits several of these features to study responses to vascular injury including hemostasis and blood vessel repair. Using a combination of video and timelapse microscopy, we demonstrate that this method of vascular injury results in measurable and reproducible responses during hemostasis and wound repair. This method provides a system for studying vascular injury and repair in detail in a whole animal model. 相似文献

Google Scholar (GS), a commonly used web-based academic search engine, catalogues between 2 and 100 million records of both academic and grey literature (articles not formally published by commercial academic publishers). Google Scholar collates results from across the internet and is free to use. As a result it has received considerable attention as a method for searching for literature, particularly in searches for grey literature, as required by systematic reviews. The reliance on GS as a standalone resource has been greatly debated, however, and its efficacy in grey literature searching has not yet been investigated. Using systematic review case studies from environmental science, we investigated the utility of GS in systematic reviews and in searches for grey literature. Our findings show that GS results contain moderate amounts of grey literature, with the majority found on average at page 80. We also found that, when searched for specifically, the majority of literature identified using Web of Science was also found using GS. However, our findings showed moderate/poor overlap in results when similar search strings were used in Web of Science and GS (10–67%), and that GS missed some important literature in five of six case studies. Furthermore, a general GS search failed to find any grey literature from a case study that involved manual searching of organisations’ websites. If used in systematic reviews for grey literature, we recommend that searches of article titles focus on the first 200 to 300 results. We conclude that whilst Google Scholar can find much grey literature and specific, known studies, it should not be used alone for systematic review searches. Rather, it forms a powerful addition to other traditional search methods. In addition, we advocate the use of tools to transparently document and catalogue GS search results to maintain high levels of transparency and the ability to be updated, critical to systematic reviews. 相似文献
