The marine ecosystem response to climate change and demersal trawling was investigated using the coupled hydrodynamic-biogeochemical water column model GOTM-ERSEM-BFM for three contrasting sites in the North Sea. Climate change forcing was derived from the HadRM3-PPE-UK regional climate model for the UK for the period 1950–2100 using historical emissions and a medium emissions scenario (SRESA1B). Effects of demersal trawling were implemented as an additional mortality on benthic fauna, and changes in the benthic–pelagic nutrient and carbon fluxes. The main impacts of climate change were (i) a temperature-driven increase in pelagic metabolic rates and nutrient cycling, (ii) an increase in primary production fuelled by recycled nutrients, (iii) a decrease in benthic biomass due to increased benthic metabolic rates and decreased food supply as a result of the increased pelagic cycling, and (iv) a decrease in near-bed oxygen concentrations. The main impacts of trawling were (i) reduced benthic biomass due to the increased mortality, and (ii) the increased benthic–pelagic nutrient fluxes, with these effects counteracting each other, and relatively small changes in other variables. One important consequence was a large decrease in the de-nitrification flux predicted at the two summer-stratified sites because less benthic nitrate was available. The effects of trawling scaled linearly with fishing effort, with greatest sensitivity to fishing in summer compared to fishing in winter. The impacts of climate change and trawling were additive, suggesting little or no non-linear interactions between these disturbances.  相似文献

The anthraquinone dye reactive blue 2 was found to be a potent inhibitor of a protein kinase isolated and purified from thylakoids. This enzyme was also inhibited in situ, with corresponding inhibition of ATP-dependent quenching of the chlorophyll fluorescence. The mode of inhibition was noncompetitive, with a Ki of 8 microM for the membrane-bound kinase, and 6 microM for the purified kinase. The inhibitor did not modify the substrate preference of the endogenous kinase and could be removed from the membrane by washing. Unlike reactive blue 2, the enzyme did not partition into detergent micelles and is therefore presumably not a hydrophobic, intrinsic membrane protein.  相似文献

In shallow ecosystems, the short temporal variability of available underwater irradiance is considered a major process controlling submerged macrophytes development. Mechanistic models that estimate photosynthetically available radiation (PAR) in shallow ecosystems at very short time scales are needed for use in predicting submerged macrophyte growth and persistence. We coupled a 2D horizontal circulation model with, first, a 1D vertical numerical model of suspended solid (SS) re-suspension, diffusion and settling, and next, with a model of vertical extinction of irradiance, previously validated at the same site. The study site was the Vaccarès lagoon (France) where a large data set of high frequency bottom irradiance and SS concentration were available. SS and irradiance measurements were conducted at a vertical study station, monitored over a 6 month period (from December 1995 to May 1996) characterized by wide-ranging wind velocities (1.5–18 ms−1). In addition, grain-size analyses conducted over the whole lagoon, allowed adaptation of the 1D numerical model to the silt-sized (7 μm) and clay-sized (0.3 μm) fractions that prevail in the local sediment. First, model results showed that about 60% of the variance in bottom irradiance time series can be explained by our deterministic formulations, thus representing the same level of efficiency than those already obtained by a stochastic model previously developed with the same data set. Second, model results showed that the fit of the model to the field data (SS concentrations and bottom irradiance) depended mainly on storm occurrence and season (winter or spring). Finally, model results suggested that the underwater irradiance regime was controlled by seasonal succession of the horizontal circulation of turbid water in the lagoon, with increased solids concentrations in winter, followed by submerged canopy development and decreased solids concentrations in spring.  相似文献

Three experiments were carried out to examine the consequences of concurrent infections with Ascaridia galli and Escherichia coli in chickens raised for table egg production. Characteristic pathological lesions including airsacculitis, peritonitis and/or polyserositis were seen in all groups infected with E. coli. Furthermore, a trend for increased mortality rates was observed in groups infected with both organisms which, however, could not be confirmed statistically. The mean worm burden was significantly lower in combined infection groups compared to groups infected only with A. galli. It was also shown that combined infections of E. coli and A. galli had an added significant negative impact on weight gain.  相似文献

Background  

Mitochondrial and nuclear genes have generally been employed for different purposes in molecular systematics, the former to resolve relationships within recently evolved groups and the latter to investigate phylogenies at a deeper level. In the case of rapid and recent evolutionary radiations, mitochondrial genes like cytochrome b (CYB) are often inefficient for resolving phylogenetic relationships. One of the best examples is illustrated by Arvicolinae rodents (Rodentia; Muridae), the most impressive mammalian radiation of the Northern Hemisphere which produced voles, lemmings and muskrats. Here, we compare the relative contribution of a nuclear marker – the exon 10 of the growth hormone receptor (GHR) gene – to the one of the mitochondrial CYB for inferring phylogenetic relationships among the major lineages of arvicoline rodents.  相似文献

Tocopherols are lipophilic antioxidants synthesized exclusively by photosynthetic organisms and collectively constitute vitamin E, an essential nutrient for both humans and animals. Tocopherol cyclase (TC) catalyzes the conversion of various phytyl quinol pathway intermediates to their corresponding tocopherols through the formation of the chromanol ring. Herein, the molecular and biochemical characterization of TCs from Arabidopsis (VTE1 [VITAMIN E 1]), Zea mays (SXD1 [Sucrose Export Deficient 1]) and Synechocystis sp. PCC6803 (slr1737) are described. Mutations in the VTE1, SXD1, or slr1737 genes resulted in both tocopherol deficiency and the accumulation of 2,3-dimethyl-6-phytyl-1,4-benzoquinone (DMPBQ), a TC substrate. Recombinant SXD1 and VTE1 proteins are able to convert DMPBQ to gamma-tocopherol in vitro. In addition, expression of maize SXD1 in a Synechocystis sp. PCC6803 slr1737 knockout mutant restored tocopherol synthesis, indicating that TC activity is evolutionarily conserved between plants and cyanobacteria. Sequence analysis identified a highly conserved 30-amino acid C-terminal domain in plant TCs that is absent from cyanobacterial orthologs. vte1-2 causes a truncation within this C-terminal domain, and the resulting mutant phenotype suggests that this domain is necessary for TC activity in plants. The defective export of Suc in sxd1 suggests that in addition to presumed antioxidant activities, tocopherols or tocopherol breakdown products also function as signal transduction molecules, or, alternatively, the DMPBQ that accumulates in sxd1 disrupts signaling required for efficient Suc export in maize.  相似文献
