Controlled addition of small amounts of oxygen with membranes

Summary In some fermentations micro-aerobic conditions are desirable. The controlled addition of trace amounts of oxygen may be achieved by bubble free oxygen supply using silicon membrane tube, operated in dead-end mode. The small oxygen uptake rate is accurately determined with this self-constructable device.     

Organization of the murine Mx gene and characterization of its interferon- and virus-inducible promoter.

Specific resistance of Mx+ mice to influenza virus is due to the interferon (IFN)-induced protein Mx. The Mx gene consists of 14 exons that are spread over at least 55 kilobase pairs of DNA. Surprisingly, the Mx gene promoter is induced as efficiently by Newcastle disease virus as it is by IFN. The 5' boundary of the region required for maximal induction by both IFN and Newcastle disease virus is located about 140 base pairs upstream of the cap site. This region contains five elements of the type GAAANN, which occurs in all IFN- and virus-inducible promoters. The consensus sequence purine-GAAAN(N/-)GAAA(C/G)-pyrimidine is found in all IFN-inducible promoters.     

Electrical stability of erythrocytes in the presence of divalent cations

Erythrocytes suspended in a medium of low ionic strength lyse under the effect of an exponential electrical pulse. The percentage of haemolysed cells decreases several-fold in the presence of divalent cations. The protective action of the ions studied increases in the following order: Ca⁺⁺, Mg⁺⁺, Zn⁺⁺. It is assumed that divalent ions bind to the negative charges of the lipid and protein molecules and reduce their electrostatic repulsion, which results in stabilization of the membranes.     

Microspectrofluorimetric analysis of the formaldehyde-induced fluorophores of 5-hydroxytryptamine and dopamine in intrapulmonary neuroepithelial bodies after administration ofl-hydroxytryptophan andl-DOPA

Summary To demonstrate the intracellular store of 5-hydroxytryptamine and dopamine in pulmonary neuroepithelial bodies of the neonatal rabbit after treatment with the corresponding amino-acid precursorsl-5-hydroxytryptophan orl-3,4-dihydroxyphenylalanine, formaldehyde-induced flourescence in combination with microspectrofluorimetric analysis has been used. Emission spectra and excitation spectra in an extended wavelength range from 240 to 460 nm, the displacement of excitation peaks after exposure to hydrochloric acid vapour, and calculation of peak ratio values 410/260, 380/320, 320/260 for phenylethylamine fluorophores and 385/315 for indolylethylamine fluorphores were performed. Thus, the presence of 5-hydroxytryptamine without occurrence of 5-hydroxytryptophan was demonstrated in pulmonary neuroepithelial bodies after administration of the corresponding biological precursor, while dopamine combined with 5-hydroxytryptamine were clearly revealed after administration ofl-3,4-dihydroxyphenylalanine. Fluorescence studies on aged and young erythrocyte populations

Structural changes in red blood cell (RBC) membrane are investigated by fluorescence techniques. Results obtained with three probes (DPH, 3-PM and fluorescamine) indicate a significant increase in membrane rigidity associated with aging of RBCs. Discrepancies between our observations and published data could arise from utilization of experimental conditions closer to physiological conditions in our study. Use of intact RBCs continuously manipulated in a 37 degrees C environment could represent experimental conditions favourable to the identification of rheologic membrane changes in senescent RBCs.     

Loss of urease activity in Helicobacter mustelae

Urease-negative variants of Helicobacter mustelae were isolated after spontaneous loss of activity during sub-culture. The whole-cell protein patterns showed that the loss of urease activity was linked to the absence of two polypeptides of 29·1 and 65·4 kDa. Restriction endonuclease analysis of chromosomal DNA indicated no substantial differences between the urease negative and positive variants. It is likely that a change in the expression of the gene for urease was responsible for these observations.     

Occurrence of dinucleosidetriphosphatase in the cytosol and particulate fractions from rat liver

Dinucleosidetriphosphatase (EC 3.6.1.29) is present in both the 37,000 g rat liver supernatant and precipitate (50 mU/g each fraction). These two activities show matching molecular weights, isoelectric points, substrate specificities, Km values, bivalent cation requirements and inhibition by zinc (II). The particulate triphosphatase and a residual dinucleosidetetraphosphatase (EC 3.6.1.17) are solubilized by freeze-thawing or by Triton X-100. Detergent treatment also extracts an unspecific phosphodiesterase I activity (EC 3.1.4.1) which also splits dinucleoside polyphosphates. The above findings suggest the occurrence of cytosolic and particulate degradative pathways for dinucleoside polyphosphates.     

Heart structure and ventricular ultrastructure of hemoglobin- and myoglobin-free icefish Channichthys rhinoceratus

Summary The structural and ultrastructural characteristics of the heart of Channichthys rhinoceratus, an antarctic teleost devoid of respiratory pigments, are described and compared with those obtained from the red-blooded related species Notothenia rossii.The heart of the icefish is characterized by a spongy myocardium supplied with a highly developed arterial coronary system. This vasculature includes a subepicardial system and an extensive intratrabecular capillary network. Arterial hilar network and Thebesian vessels may also be present. The bulbus arteriosus shows unusually large spheroid structures located in the middle layer of the wall.Both white- and red-blooded species display comparable myocardial cell morphology and organelle distribution. However, the mitochondrial cristae of the former are more densely packed and the sarcolemma possesses numerous caveolae. A large proportion of non-contractile cells is also found in the icefish ventricular wall.     

Isolation of astrocytes,neurons, and synaptosomes of rat brain cortex

A simplified method was developed for the bulk separation of neuronal perikarya and astroglial celis from adult rat brain without the involvement of density gradients. Activities of various enzymes involved in glutamate metabolism were estimated and compared with those of synaptosomes. The activities of glutamate dehydrogenase and aspartate aminotransferase were higher in synaptosomes than in neuronal perikarya or glia. Glutamine synthetase was distributed in all the three fractions while glutaminase activity was higher in astrocytes than in synaptosomes and was not detectable in neuronal perikarya. The significance of these results in relation to metabolic compartmentation was discussed.     

A neural cocktail-party processor

Sensory segmentation is an outstanding unsolved problem of theoretical, practical and technical importance. The basic idea of a solution is described in the form of a model. The response of neurons within the sensory field is temporally unstable. Segmentation is expressed by synchronization within segments and desynchronization between segments. Correlations are generated by an autonomous pattern formation process. Neuronal coupling is the result both of peripheral evidence (similarity of local quality) and of central evidence (common membership in a stored pattern). The model is consistent with known anatomy and physiology. However, a new physiological function, synaptic modulation, has to be postulated. The present paper restricts explicit treatment to the peripheral evidence represented by amplitude modulations globally present in all components of a sound spectrum. Generalization to arbitrary sensory qualities will be the subject of a later paper.
