Parkinson's disease is characterized by progressive cell death in the substantia nigra pars compacta, which leads to dopamine
depletion in the striatum and indirectly to cortical dysfunction. Increased glutamatergic transmission in the basal ganglia
is implicated in the pathophysiology of Parkinson's disease and glutamate receptor mediated excitotoxicity has been suggested
to be one of the possible causes of the neuronal degeneration. In the present study, the effects of serotonin, gamma-aminobutyric
acid and bone marrow cells infused intranigrally to substantia nigra individually and in combination on unilateral 6-hydroxydopamine
induced Parkinson's rat model was analyzed. Scatchard analysis of total glutamate and NMDA receptor binding parameters showed
a significant increase in Bmax (P < 0.001) in the cerebral cortex of 6-hydroxydopamine infused rat compared to control. Real Time PCR amplification of NMDA2B,
mGluR5, bax, and ubiquitin carboxy-terminal hydrolase were up regulated in cerebral cortex of 6-hydroxydopamine infused rats
compared to control. Gene expression studies of GLAST, ά-Synuclien and Cyclic AMP response element-binding protein showed
a significant (P < 0.001) down regulation in 6-OHDA infused rats compared to control. Behavioural studies were carried out
to confirm the biochemical and molecular studies. Serotonin and GABA along with bone marrow cells in combination showed reversal
of glutamate receptors and behaviour abnormality shown in the Parkinson's rat model. The therapeutic significance in Parkinson's

Variation in cognitive performance, which strongly predicts functional outcome in schizophrenia (SZ), has been associated with multiple immune‐relevant genetic loci. These loci include complement component 4 (C4A), structural variation at which was recently associated with SZ risk and synaptic pruning during neurodevelopment and cognitive function. Here, we test whether this genetic association with cognition and SZ risk is specific to C4A, or extends more broadly to genes related to the complement system. Using a gene‐set with an identified role in “complement” function (excluding C4A), we used MAGMA to test if this gene‐set was enriched for genes associated with human intelligence and SZ risk, using genome‐wide association summary statistics (IQ; N = 269 867, SZ; N = 105 318). We followed up this gene‐set analysis with a complement gene‐set polygenic score (PGS) regression analysis in an independent data set of patients with psychotic disorders and healthy participants with cognitive and genomic data (N = 1000). Enrichment analysis suggested that genes within the complement pathway were significantly enriched for genes associated with IQ, but not SZ. In a gene‐based analysis of 90 genes, SERPING1 was the most enriched gene for the phenotype of IQ. In a PGS regression analysis, we found that a complement pathway PGS associated with IQ genome‐wide association studies statistics also predicted variation in IQ in our independent sample. This association (observed across both patients and controls) remained significant after controlling for the relationship between C4A and cognition. These results suggest a robust association between the complement system and cognitive function, extending beyond structural variation at C4A. 相似文献

Biochemical dissection of the “acid-growth” process of plant cell walls led to the isolation of a new class of wall loosening
proteins, called expansins. These proteins affect the rheology of growing walls by permitting the microfibril-matrix network
to slide, thereby enabling the wall to expand. Molecular sequence analysis suggests that expansins might have a cryptic glycosyl
transferase activity, but biochemical results suggest that expansins disrupt noncovalent bonding between microfibrils and
the matrix. Recent discoveries of a new expansin family and gene expression in fruit, meristerms and cotton fibers have enlarged

A number of tumor cell lines have been examined that differentially produce human chorionic gonadotropin and the isolated alpha- or beta-subunits. It has been demonstrated that all of the cell lines studied to date contain genes for both alpha- and beta-subunits, indicating that differential and exclusive expression of one subunit is not the result of a particular cell line having lost the gene for the alternate subunit as a consequence of chromosome changes accompanying cell transformation. Because many of these established cell lines are aneuploid, it is also significant that no evidence was found for gene amplification in cell lines producing alpha-subunit at very high levels compared to those with very low level expression. Analysis of restriction endonuclease digests of tumor cell DNAs has demonstrated identical patterns for beta-subunit in KpnI digests and KpnI/HindIII double digests. Polymorphisms were observed for alpha-subunit in EcoRI and HindIII digests, but these did not correspond with expression of the alpha-subunit. Significant levels of either mRNA (as determined by dot blot and Northern transfer hybridization analysis) were accompanied by corresponding elevated levels of alpha- and beta-subunits (as determined by radioimmunoassay), suggesting that regulation of subunit production most likely occurs at a pretranslational stage. However, there were apparent differences in the relative ratio of alpha- and beta-subunits and their cognate mRNAs among the cell lines. 相似文献
