Sipha maydis (Passerini) is a new aphid pest of cereals and cultivated and wild grasses in Argentina. This species was recently introduced into America, and nothing is known of its distribution or host range in South America. A better understanding of its biology is likely to facilitate control. This article records 1) the distribution and 2) the host range of S. maydis in Argentina. Over the period 2004-2006 samples were collected from 32 populations at several localities in Argentina. The number of S. maydis, accompanying aphid species, and the host from which they were collected were recorded. The distribution of S. maydis ranged from 32 degrees 52' to 42 degrees 03' S, and from 57 degrees 41' to 71 degrees 24' W, bounded by isothermals 18 and 10 degrees C and isohyets 200-400 and <1,200 mm. No S. maydis were found in the subtropical region, even in winter. In the field, the different populations showed very different host preferences. S. maydis was found mainly on cultivated barley and wheat and on wild Bromus spp. and Sorghum halepense (L.) Pers. No aphids were found on maize, Zea mays L. Most of the damage to winter cereal crops occurred at the seedling stage in early autumn and of adult plants when infestations occurred in late spring. In the 4 yr after the first record of S. maydis in Argentina, it colonized a huge area similar to that colonized by Diuraphis noxia (Mordvilko) in 10 yr. The wide range of regions, hosts and climatic conditions this species is adapted to is likely to make the control of this pest very difficult.  相似文献

Objectives

To determine whether the interleukin-33 (IL-33)-interleukin-1 receptor like 1 (IL-1RL1) signaling pathway is implicated in the risk of subclinical atherosclerosis in patients with rheumatoid arthritis (RA).

Methods

A total of 576 Spanish RA patients from Northern Spain were genotyped for 6 well-known IL33-IL1RL1 polymorphisms (IL33 rs3939286, IL33 rs7025417, IL33 rs7044343, IL1RL1 rs2058660, IL1RL1 rs2310173 and IL1RL1 rs13015714) by TaqMan genotyping assay. The presence of subclinical atherosclerosis was determined by the assessment of carotid intima-media thickness (cIMT) by carotid ultrasound (US).

Results

RA patients carrying the TT genotype of the IL33 rs3939286 polymorphism had lower cIMT values than those homozygous for the CC genotype (mean ± standard deviation (SD): 0.71 ± 0.14 mm versus 0.76 ± 0.16 mm, respectively) while patients carrying the CT genotype had intermediate cIMT values (mean ± SD: 0.73 ± 0.17 mm). Moreover, RA patients carrying the mutant allele T of the IL33 rs3939286 polymorphism exhibited significantly lower cIMT values than those carrying the wild allele C (mean ± SD: 0.72 ± 0.16 mm versus 0.75 ± 0.18 mm respectively; p = 0.04). The association of both genotype and allele frequencies of IL33 rs3939286 and cIMT levels remained statistically significant after adjustment for sex, age at the time of US study, follow-up and center (p = 0.006 and p = 0.0023, respectively), evidencing that the potential effect conferred by IL33 rs3939286 may be independent of confounder factors. No association with other IL33-IL1RL1 genetic variants was observed.

Conclusions

In conclusion, our results may suggest a potential protective effect of the IL33 rs3939286 allele T in the risk of subclinical atherosclerosis in patients with RA.  相似文献

Conformational criteria based on the Ramachandran plot are mainly used to distinguish bet ween distorted and adequately refined models. S-Adenosylmethionine serves as the methyl donor for many biological methylation reactions and provides the propylamine group for the synthesis of polyamines. S-Adenosylmethionine is synthesized from methionine and ATP by the enzyme methionine adenosyltransferase. The cellular factors regulating S-adenosylmethionine synthesis have not been well defined. Here we show that in rat hepatocytes S-nitrosoglutathione monoethyl ester, a cell-permeable nitric oxide donor, markedly reduces cellular S-adenosylmethionine content via inactivation of methionine adenosyltransferase by S-nitrosylation. Removal of the nitric oxide donor from the incubation medium leads to the denitrosylation and reactivation of methionine adenosyltransferase and to the rapid recovery of cellular S-adenosylmethionine levels. Nitric oxide inactivates methionine adenosyltransferase via S-nitrosylation of cysteine 121. Replacement of the acidic (aspartate 355) or basic (arginine 357 and arginine 363) amino acids located in the vicinity of cysteine 121 by serine leads to a marked reduction in the ability of nitric oxide to S-nitrosylate and inactivate hepatic methionine adenosyltransferase. These results indicate that protein S-nitrosylation is regulated by the basic and acidic amino acids surrounding the target cysteine.  相似文献

Wnt/β-catenin pathway controls biochemical processes related to cell differentiation. In committed cells the alteration of this pathway has been associated with tumors as hepatocellular carcinoma or hepatoblastoma. The present study evaluated the role of Wnt/β-catenin activation during human mesenchymal stem cells differentiation into hepatocytes. The differentiation to hepatocytes was achieved by the addition of two different conditioned media. In one of them, β-catenin nuclear translocation, up-regulation of genes related to the Wnt/β-catenin pathway, such as Lrp5 and Fzd3, as well as the oncogenes c-myc and p53 were observed. While in the other protocol there was a Wnt/β-catenin inactivation. Hepatocytes with nuclear translocation of β-catenin also had abnormal cellular proliferation, and expressed membrane proteins involved in hepatocellular carcinoma, metastatic behavior and cancer stem cells. Further, these cells had also increased auto-renewal capability as shown in spheroids formation assay. Comparison of both differentiation protocols by 2D-DIGE proteomic analysis revealed differential expression of 11 proteins with altered expression in hepatocellular carcinoma. Cathepsin B and D, adenine phosphoribosyltransferase, triosephosphate isomerase, inorganic pyrophosphatase, peptidyl-prolyl cis-trans isomerase A or lactate dehydrogenase β-chain were up-regulated only with the protocol associated with Wnt signaling activation while other proteins involved in tumor suppression, such as transgelin or tropomyosin β-chain were down-regulated in this protocol. In conclusion, our results suggest that activation of the Wnt/β-catenin pathway during human mesenchymal stem cells differentiation into hepatocytes is associated with a tumoral phenotype.  相似文献

The bacterium Sphingomonas paucimobilis, isolated from cinematographic films in an earlier project, was able to biodeteriorate the cellulose triacetate material (acetylation degree of 2.7). Film colonization was monitored by the indirect impedance technique and the production of carbon dioxide. The presence of ruggedness and irregularities on the surface of the film, produced when the plasticizer was extracted, accelerated the biodeterioration of the material. In contrast, cinematographic films with their layered structure made of photographic gelatine emulsion were protected and no colonization was observed. The cinematographic film without photographic emulsion reached a 5% level of biodeterioration after six weeks of incubation, confirming the possibility of biodeterioration of archival cinematographic materials if conservation conditions are not adequate. Through viscosity measurements, a decrease in relative viscosity was observed on the biodeteriorated sample, with respect to the original material, confirming a lower molecular weight as a result of enzymatic activity of S. paucimobilis. Also, using chemiluminescence, the film surface oxidation on the biodeteriorated sample was observed. This technique was very sensitive in detecting material oxidation by reactive oxygen species generated by bacteria, and could be useful to study microbiological biodeterioration in polymeric materials.  相似文献

Rat liver S-adenosylmethionine (AdoMet) synthetase appears as high-M(r) (tetramer) and low-M(r) (dimer) forms. Both are inhibited in the presence of GSSG at pH 8. The calculated Ki values are 2.14 and 4.03 mM for the high- and low-M(r) forms, respectively. No effect on enzyme activity was observed in the presence of GSH, but modulation of inhibition by GSSG can be obtained by addition of GSH. At a total glutathione concentration (GSH + GSSG) of 10 mM, a KOX of 1.74 was calculated for the high-M(r) form, whereas this constant was 2.85 for the low-M(r) AdoMet synthetase. No incorporation of [35S]GSSG was observed in either of the enzyme forms, and inhibition of enzyme activity was correlated with dissociation of both AdoMet synthetases to a monomer. The data obtained in the presence of GSSG seem to suggest that oxidation leads to the formation of an intrasubunit disulfide. The possible regulation of AdoMet synthetase activity by the GSH/GSSG ratio is discussed, as well as its in vivo significance.  相似文献
